WHAT WAS THE PROTOCOL?

1. A 7um section of tissue was used.2. Embryos were fixed for 2.5 hours with 4% PFA in PBS.3. They were then incubated in 20% sucrose until they sunk.4. Immersion in cryoprotective solution.5. Flash frozen.6. Blocking solution contained 10% serum, 1% BSA in PBS, 0.5% triton x-100. 7. After O/N incubation with primary antibody, the section was incubated with

secondary antibody (Alexa 488, Invitrogen) for 1 hour. 8. No antigen retrieval was necessary.

N.B. Antigen retrieval procedures with EDTA buffer or Citrate buffer prevented the immunostaining.
