Six months to 10-old improvement

In the six months since the Ion PGM™

sequencer launched, its throughput hasincreased tenfold, its workow hasbecome the fastest in the industry, andit’s now the fastest-selling sequencerin the world, with placements in morethan 40 countries. In the second halfof the year you’ll see more of the same.

Read lengths will double to 200 basepairs and throughput will increaseby tenfold—again.

All of these advances are a direct resultof Ion’s semiconductor sequencingtechnology and native chemistry, whichmake sequencing faster, simpler, andmore scalable than ever before.

More Throughput, Better Value

The scalability of Ion semiconductorsequencing will enable a hundredfoldincrease in throughput this year.The new Ion 316™ chip has 100 Mbof throughput, tenfold more than theoriginal Ion 314™chip. The Ion 318™

chip, available in the fourth quarter,will deliver 1 Gb of throughput. Each

of the three chips will work on theIon PGM™ sequencer, giving usersthe exibility to buy only as muchthroughput as they need for anexperiment, from 10 Mb to 1 Gb.

Ion semiconductor sequencing offersexceptional value as well as perfor-mance. The Ion 314™ chip is nowavailable for just $99 USD, whichenables the sequencing community todevelop novel methods and applica-tions. Ion Torrent is able to lower theprice because of the signicant salesvolume generated by the Ion 314™ chipand the economies of scale inherent insemiconductor manufacturing.

Better Accuracy

The Ion PGM™ system’s accuracy hasimproved by an order of magnitudesince it was launched six months ago,as a result of the sy stem’s speed andsimplicity. Ion Torrent has capitalizedon the inherent accuracy of its simplesequencing chemistry, achieving moresignal, better signal processing and

improved base calling. Accuracyimproved quickly in part because theIon PGM™ sequencer’s two-hour runtimes enable Ion Torrent R&D teams toquickly test and rene ideas. Improve-ments in accuracy will help Ion Torrentreach 200 base pair reads in the secondhalf of this year and 400 base pair readsin 2012, making many new applicationspossible for researchers.

Scientists turned to the Ion PGM ™

sequencer to identify a deadly strainof E. coli in Europe in June because thesystem’s two-hour run time was fasterthan anything else on the market.Scientists at BGI sequenced thebacterium within three days, and ateam at the University of Münstersequenced the strain and developed aeld-deployable test in just ve days.

Scientists around the world also havereported exceptional throughputresults with their rst runs on the Ion316™ chip. Early-access users at BaylorCollege of Medicine recently broke IonTorrent’s internal record for highestoutput, after reporting 287 Mb from asingle Ion 316™chip run, and the BroadInstitute reported up to 250 Mb. Mostrecently, the Wellcome Trust SangerInstitute reported that one of the rstruns of an Ion 316™ chip on their Ion

sequencer produced a totalyield of 194 Mb.
