TNF - THE MEYPHARMA FORMULATION.

Introduction. The body is a fine-tuned Instrument with checks and balances

throughout. We are here concerned with the Auto-immune system, where

TNF (Tumor Necrosis Factor) is pivotal to do its repair and maintenance work

and fight invasive enemies. The essential features of TNF are that it triggers an

inflammatory response as well as increased cell proliferation. But as we

age, something goes wrong: once the repair work has been done, the TNF

mechanism is supposed to switch off, but doesn’t. Now the body has a

chronic problem: chronic inflammation. This becomes a chronic disease,

called Auto Immune Deficiency. It has been estimated that as many as 85%

of patients consulting Doctors in the USA suffer from Chronic Inflammation.

The standard treatment are immune suppressants, but this has serious and

severe side effects. Our formula does not suppress the immune system, but

modulates it, and in the process ensures that the Immune system can

function normally without causing side effects.

But the TNF formula goes further: it helps balance vary many bodily systems

and mechanisms for general health and vitality.

Meypharma has formulated a unique tonic containing all-natural plant

Bromelain. This is an extract from pineapples. It belongs to a group of

proteolytic enzymes and is made up of multiple enzyme species with

different proteolytic activities. However, apart from the platelet

aggregation inhibitory and anti-inflammatory action (proteolytic) it also

displays other pharmacological properties, such as inhibition of tumor cell

- Prolongs the prothrombin and partial thromboplastin time

- Prevents aggregation of blood platelets

- Reduces the blood level of plasmakinins

- Reduces the level of prostaglandin E2 and of thromboxane

A2 in exudates during acute inflammation

- Acts as anti-inflammatory agent

- Induces the secretion of interleukin and TNF-a from blood

monocytes and granulocytes

- Supports the oxidative burst and the cytotoxicity of

granulocytes against tumor cells (refer H.R. Maurer).

For the purposes of controlling excessive TNF-an activity, Bromelain

clearly performs a variety of appropriate functions, and whilst the

positive necessity of TNF- a is recognized, Bromelain was specifically

selected for its modulating (as opposed to an inhibitory) effect on

TNF-a.

No serious side effects have been detected. Bromelain is

considered to be non-toxic

Coenzyme Q10. Co Q10 is found in the membranes of many organelles.

The highest concentration is found on the inner membrane of the

mitochondrion. It is also present in endoplasmic reticulum, peroxisomes,

lysosomes and every cell of the body. Coenzyme Q10 is a highly versatile

substance which has multiple beneficial effects. The following are some

of the key benefits:

- It plays a unique role in the electron transport chain through its

involvement in ATP synthase

- It is an antioxidant

- Improve immune function

- Acts as diuretic

- Plays an important role in heart health

- Lowers blood pressure

We need to remind ourselves of the primary purpose of this formulation,

L- arginine. Aging is associated with a decline in the effective functioning

of immune cells, and one of the reasons is the decline in serum l-arginine

and nitric oxide metabolites. The most important contribution of l-arginine

in the immune function is its capacity to synthesise Nitric Oxide (NO).

However, we will list below a number of additional functions of l-arginine.

It is noted, however that an increasing number of studies refer to the

important role of NO as a modulator of a variety of acute and chronic

inflammatory disorders. It is also important to note the duality or sometimes

contradictory nature of many of these. The following is a list of the more

critical functions of NO:

- NO mediates host protection through micro biostatic and

microbicidal action

thus blocking inflammation in blood vessels

- It causes pathogen killing in parasitic diseases - It can switch from regulating apoptosis of immune cells to

blocker of apoptosis to an inducer of apoptosis

- Depending on the circumstances it can be cytotoxic, or it can

protect cells from toxic insult

- It is both apoptosis inducing and anti-apoptotic

- It can be both antioxidant or facilitator of oxygen

In summary, it appears that NO has to be considered in the context of

timing and duration and different stages of and during inflammatory

reactions. What is clear is that although NO is vital for host survival, it is

delicate, but not completely understood (Pavanelli et.al.).

In addition to the vital role of l-arginine as precursor to NO, it has also

been found to:

- Be actively involved in combating cancer

- Improving vascular function

- Reducing blood pressure

- Overcoming erectile dysfunction

Various clinical trials where l-arginine was tested reported only

gastrointestinal side effects. This is attributed to the low level of

bioavailability (± 20%). It was further found that the half-life of l-arginine

was also relatively short. Higher dosages did not improve

bioavailability, as these were discarded through urinary excretion.

The entrapment of l-arginine in the multi-liposomal base will help

obviate gastrointestinal side effects, improve bioavailability and extend

therapeutic window. The liposomal formula offers a delayed release

mechanism as well as targeted delivery to the site of therapeutic

interaction.

No 1- TOPICAL FUNGAL INFECTIONS Our antifungal formula offers the following benefits: It is known that conventional antifungal medications are cytotoxic. This

means that where the active agent is exposed to normal skin, cell damage occurs. Research has shown that Tea tree Oil has no cytotoxicity. In fact, it has no known side effects.

It is known that cells develop drug resistance to conventional antifungal medication, but it has been shown that the body’s natural defense mechanism does not respond adversely to Tea Tree Oil.

Whereas conventional medication treats fungi from the top, and has to remove fungi layer-by-layer, in this formula Tea Tree Oil is encapsulated in our Liposomal delivery system, and therefore capable of deep penetration. No 2 - SUN SPOTS SKIN BLEMISHES Our Blemish Remover serves a variety of purposes: It helps remove liver spots, certain freckles, bullous spots and small scars. But most importantly, it is an effective treatment for skin damage such as solar keratosis. It is a blend of natural ingredients that are entrapped in a liposomal base ensuring deep penetration through the skin to where the damage has been caused. If you are older than 40 years there is every reason that you will be affected. The table below shows how the testosterone levels reduce with age. Not only that, but there is another factor that makes you more vulnerable: Sex Hormone Binding Globules (SHBG). This table also shows how these Globules increase with age. This is highly significant, as SHBG binds to testosterone and once bound those testosterone particles are completely neutralised. It is only a small portion of testosterone particles that are not bound and these are called Free Testosterone. The normal portion of Free Testosterone left from total testosterone is about 2%. It will be clear from the table that with increased SHBG, there is less Free Testosterone left, and at advanced ages the eventual proportion of Free Testosterone is less that 1,5%.

Note that the above study was done in 1996. In a recent study it was found that from 1988 through to 2004, there had been a steady decline of 1.2% per year in both Total Testosterone and Free Testosterone, which means that the current values (no statistics available) should be much lower that those reflected in the above table, whereas the 1988 figures would have been much higher.

Apart from the natural decline there are also other factors that adversely affect testosterone levels. The most important of these are: Injury, infection, Chemotherapy, Some Inflammatory Diseases, Medications such as Corticosteriods, Chronic illness, Stress, High levels of Alcohol consumption and obesity. WHAT ARE THE ALTERNATE METHODS OF TREATMENT? There are mainly 4 methods of administering testosterone replacement therapy: Oral, Injectable, Topical Patch and Topical Cream. The oral method has been found to be unsuitable for clinical use for the following reasons: Research has shown that the rapid degradation of testosterone in the

liver has damaging effects on the liver, and is not suitable for clinical use.

It has also been found that natural testosterone taken orally increases risk of lowered Immune function and toxicity.

Except for the shaft and root hair is basically made up of dead, cornified cells, and its outer layer (the cuticle) consists of scales. FUNCTIONAL FEATURES First, cells need amino acids with which they manufacture certain enzymes and structural proteins. In simple terms, a cell is nothing but a set of chemical reactions, and enzymes make those reactions happen properly.

Secondly, cells need glycerol. Energy is an essential requirement, and for cells to produce energy, there are many cellular elements that need to function optimally: Cells require lipids, glucose and oxygen; then insulin is required to transport glucose through cell membranes; cells require the necessary amino acids with which to manufacture energy enzymes as mentioned above; mitochondria (the energy producers inside cells) need to function optimally. Insulin is a vital link in this process, but is one element that often causes problems. As example, research has shown a direct link between fat accumulation and insulin resistance. The basic function of insulin is to regulate carbohydrate and lipid metabolism, and in cases of type 2 diabetes and obesity insulin is either incapable of doing this or does so inadequately. With ageing there is also an increase in insulin resistance, with a consequent inadequate availability of glucose. Research findings proved that medium chain triglycerides (MCT) speed up metabolism, reduce insulin resistance, reduce fat accumulation and increase energy. The obvious consequence will be weight loss. Even more important is that MCT does not require insulin to be transported through cell membranes, and therefore bypass permanent insulin resistance. The third essential substance required by cells is Oxygen. This is burned together with Fatty Acids and glucose to produce energy. There is a specific sequence by which these nutrients are converted into ATP (the final energy molecule). The incorporation of oxygen is the last step in this process & insufficient oxygen results in waste that needs to be removed. Lactic acid in muscle cells is an example of such waste. The typical signs of shortage of oxygen are: cellular destruction, low energy, fatigue, poor digestion and lowered metabolism. Conversely, excess oxygen causes serious problems. This is called oxidative stress. This results in the production of peroxides and free radicals that damage all components of the cell. The fourth essential substance is Vitamin E. This is found in cell membranes, and its primary role is to act as antioxidant, where it is required to protect cells against intercellular free radicals. Vitamin E helps keep cells viable.

Then finally, Essential fatty acids (EFA’s) complete the parcel of essential cellular nutrients. Again, the word “essential” refers to those nutrients that the body cannot manufacture. EFA’s have various necessary functions, such as repair cell membranes, absorb nutrients, eliminate waste and produce prostaglandins.
