COS'È IL PROGRAMMA
LLP/ERASMUS

Proposition for a project for a diploma-thesis in Berlin
Background
A yeast strain mutant with a deregulated HMG-CoA Reductase produces high amounts of Squalene.
The accumulated Squalene affects composition and function of lipid particles, subcellular organelles rich in
Triacyl glycerides (TAG) and Sterylesters (STE).
Mutants lacking the enzymes responsible for TAG and STE were constructed.
Proposed title
Analysis of lipid particles in mutant strains of the yeast Saccharomyces cerevisiae.

STUDIES ON CELL POLARITY
The generation of cell polarity is critical for the control of many
biological processes such as early embriogenesis, intracellular
transport of organelles in polarized epithelial cells, directed
movement of migratory cells, neurite outhgrowth and asymmetric
cell division.
The Rho-type GTPase Cdc42 plays a key role in the
establishment and maintenabce of cell polarity.
We use the budding yeast Saccharomyces cerevisiae as a
model organism to study the biochemical and molecular
biological mechanisms underlying cell polariry.

Bacterial resistance to antibiotics is now becoming a major problem. For instance, the MRSA (Multi Resistant Staphylococcus Aureus)
bacterium, see picture above, has in several instances forced hospitals to close down due to contamination. Now, the problem gets even bigger
since the first reports of bacterial resistance against vancomycin (the antibiotic of last resort against MRSA) are emerging. Thus, new
types/classes of antibiotics are high in demand. A candidate that shows promising characteristics is the lantibiotic NISIN. This peptide (see
picture below) which is used as a preservative in the food industry (dairy mainly) has the ability to kill bacteria effectively by pore-formation in
the plasma membrane via a high affinity interaction with the membrane anchored Lipid II. More information on this subject is found at the
personal homepage of Dr. Eefjan Breukink : www.breukink.net

Aim of our work is to characterize the molecular mechanisms that control
membrane homeostasis, using yeast as model organism.
……the genetic interaction between mutations in ceramide synthesis
and those in the late part of the ergosterol biosynthetic pathway are
being analyzed to characterize the role of the two lipids in the formation of
detergent insoluble membrane domains (“rafts”).

Main subjects studied in our group are biosynthesis of lipids and their assembly
into organelle membranes of the yeast Saccharomyces cerevisiae.
……..Maintenance of organelle lipid profiles requires strict coordination
between regulation of biosynthetic and translocation activities. Specific
aspects studied are (i) assembly and homeostasis of phosphatidylethanolamine in
yeast organelle membranes, (ii) neutral lipid storage in lipid particles and
mobilization, and (iii) characterization of organelle membranes from Pichia
pastoris.
