Materials (Including the lab Manual)

• Part 1: – 2 test tubes, water, phenol red, a straw, a 2” piece of Elodea

• Part 2: – test tube+cork; mortar and pestle; leaf; acetone; spot

plate; filter paper; organic solvent

• Part 3:– A variegated leaf; Hot water bath; hot alcohol; petri dish;

iodine;

• Part 4:– 4 test tubes (A-D); P04 buffer pH 6.5; chloroplasts; DCPIP

(strong redox potential!); aluminum foil; light source

• Part 5:– Microscope; prepared slides ONE, TWO, THREE

Overview: What we’re doing

• Part 1: Watch photosynthesis happen in a closed system.

• Part 2: Separate the color molecules in a leaf.

• Part 3: Show how only the green parts of a plant make sugar.

• Part 4: Show how the ETC in the light reactions of chloroplasts has NOTHING to do with carbon fixation of the Calvin Cycle

• Part 5: look at the structural differences between C3 plants and C4 plants.

Setup

• Organize in groups of 2-3. There are 10 setups total.

• Read through the materials and make sure you have them all.

• Try to work together on one part at a time.• Be careful with some of the reagents.

They are slightly dangerous. Wear protective gloves and goggles when dealing with organic setups.

Time Considerations

• You will have the entire class period today to work on the entire lab.

• Work cautiously. Do not rush.

• You will be expected to stay the entire class period– this discourages groups to rush through

their work so they can leave early.

Time Considerations

• Part 1: Start it and come back to it in 45’ to an hour.

• Part 2: Can be done all at once.– I will determine whether we are

calculating Rf values.

• Part 3: Reread page 8, the starch test. Can then be done all at once.

Time Considerations

• Part 4: This will be the time-consuming portion.– Observations at 5 minute intervals. Use a clean piece of

paper and record 5 intervals total. Record any notes that differ from the standard protocol

.

• Part 5: Do NOT take out your microscope until you have a clean bench and hands. I will check over your lab space before I give you slides. Please do NOT use your gloves from previous sections when handling the microscopes.– This part can be worked through quietly, while reading

along in your laboratory manual.

Assessment Expectations• You and your group are expected to complete the entire laboratory. Incomplete

sections will negatively affect your laboratory grade.

• Each group will turn in ONE set of pages 63 – 72.– Answer questions on pages

• 64, 66, 68, 69

• Each STUDENT will turn in on CLEAN PAPER– Diagrams on page 67, 70, 71 (two diagrams)– A single paragraph hypothesis regarding guard cells (page 72 question #1)– A 3-5 sentence paragraph answering question #2 on page 72.

• The Group Lab Report will be stapled together. Individual Group Members work will be paperclipped to the stapled report.

• I will expect this work to be neat and typed out in 10-12 point Arial font, double spaced.

• This will be due November 20th, the last day before Thanksgiving break. Late lab reports will not be accepted.

Time Considerations

• This powerpoint will stay up during the lab; feel free to come up and look through it at any time, or ask me for clarifications.

• Please note this is my first run through this laboratory. It will go smoothly, but expect the unexpected!
