The use of primary antibodies (Prim’Ab) as research tools is, in many ways, responsible for much of the progress seen in modern biological research. These antibodies, specific of target antigens, are a first step to probes samples in various detection. They can also be used to immobilize antigens on supports. Secondary antibodies are used in a second step to detect and amplify Prim’Ab signal by colour or light emission as fluorescence or chemiluminescence.

One of the earliest use for antibodies, came in the early part of the 20th Century when Karl Lands-teiner develop a serological test for identifying different blood groups (ABO system for typing blood). Today antibodies are used as versatile reagents for an extremely wide range of assays, in technics as varied as screening phage libraries, biosensors, affinity purifications, and conventional immunoassays including ELISA, blotting, flow cytometry, agglutinations...Because of these multiple uses, a wide range of antibodies is needed (regarding antigen-specificity, binding affinity, and format/purity). Fortunately, a large number of companies now provides antibodies for researchers. This means that each individual lab no longer has to go through the hassle of generating, purifying and testing their own antibodies. However it is often difficult to find out the antibody companies, and the best supplier.

■ Chicken IgY antibodiesIgY antibodies have multiple advantages over IgG antibodies, including higher yields, lower backgrounds, higher specificity, stronger avidity, and applicability to many immunoassays.

Despite the overall similarities between IgY and conventionally used IgG antibodies, there are some profound differences in their chemical structures and biological features. IgY has a larger Fc region and a shorter hinge region than IgG. The IgY Fc region also has an additional pair of carbohydrate chains with unique sugar residues, important for highly efficient covalent coupling to solid surfaces such as microbeads or microarrays.

The affinity maturation process of IgY is via gene conversion, different from that of IgG via somatic mutation.

The phylogenetic distance between birds and mammals makes the chicken immune system often far more responsive to mammalian protein antigens thus generating highly reactive antibodies, particularly targeted to conserved proteins against which IgGs are difficult or impossible to raise.

The IgY production process is non-invasive and accumulative from collecting eggs, in contrast to that of IgG which requires bleeding and collecting sera from immunized animals.

The productivity of IgY is also superior, since one egg can generate about 100 mg of total IgY, about the yield of 50-100 mice.

» Polyclonal IgY antibodies have been shown to be suitable for all the conventional immunoassays such as Western blot, immunoprecipitation, immunohistochemistry, cell sorting, cell/tissue staining, etc. The quality of these assays using IgY antibodies is either comparable to or better than using conventional IgG antibodies.» IgY antibodies have also been shown to be very useful for cell or tissue immuno-staining (Immunohistochemistry and Immunocytochemistry). Both types of applications can implement high-throughput screening via tissue or cell array methodology. IgY antibodies are particularly more suitable for array (chip), nano-bar, or microbead applications, due to their higher stability, lower cross-reactivity, stronger binding (avidity), and greater detection sensitivity than those of IgG antibodies against the same antigens. Each of these features has been confirmed through side-by-side comparisons of the two types of antibodies under identical conditions.
