Introduction

Genevac manufacture sample holders from solid high grade aluminium to very precise specifications. Holders are mass balanced to ensure smooth running of the evaporator. Genevac evaporators are compatible with a wide variety of sample formats and are designed to combine rapid and safe evaporation with high capacity. In order to optimise evaporation performance, it is important to select the correct sample holder for your particular application. This guide will help you make the right choice.

Range of sample holders

This Brochure contains information on the range of sample holders that are intended for use in Genevac Series 3 HT, HT-SII or EZ-2 series evaporators. The compatibility of each holder is shown as follows:

Nomenclature

Tube holder : A holder which accommodates tubes with rounded ends Vial Holder: A holder which accommodates vials with flat ends

Bottle holder: A holder which accommodates bottles with flat ends Flask holder: A holder which accommodates round bottom flasks.

Holders for tubes with conical or tapered ends are identified individually.

Swing / holder type

A comprehensive range of sample holders is available to suit plates, vials, tubes, beakers, flasks, etc. They can be one piece integral holders or may consist of a separate sample holder and swing; all are designed to lift out of the rotor so samples can be loaded easily.

Heat transfer

A major factor in determining the speed of evaporation is the rate at which heat is transferred into the sample; greater heat transfer rates mean faster evaporation. Genevac HT series II and EZ-2 series evaporators optimise heat transfer through:

High quality aluminium rotors and holders: For high thermal conductivity and low weight, Genevac systems use aluminium sample holders which have particularly efficient heat transfer properties.

High quality engineering: Genevac samples holders are manufactured to very precise specifications. Tubes fit snugly into the tube holders as any gaps would slow down the heat transfer (this is why it is important to provide us with a sample of tubes when placing orders for sample holders).

Heat transfer plates for microplates

Heat transfer plates provide improved evaporation results when using mictotitre plates. Improvements will be seen in the evenness of sample drying across the plate and, for some applications, heat transfer plates may help to reduce evaporation times by up to 50%. Improvements are most notable when used with deep-well or mid-well plates. When using heat transfer plates with pliable thermal pads, the plate should be inserted into the swing with the metal base plate facing downwards (contacting the swing), the microtitre plate is loaded on top of the pliable pad.

Note: Heat transfer plates are for use with microtitre plates only.

Protective coatings

Standard aluminium sample holders are hard anodised (water sealed) for protection against all normal organic solvents including strong acids such as TFA, HCl and thionyl chloride. The colouring of the surface finish may vary slightly from batch to batch; this is normal and does not affect the performance of the sample holder. Note: HCl or thionyl chloride should only be evaporated in systems that are HCl resistant. These systems are specifically built to resists attack from HCl and thionyl chloride.

Advice on labelling glassware

There are two common methods of labelling glassware in the laboratory; both can cause problems in the evaporator. Paper or film labels: The label effectively increases the tube diameter (the

problem is exaggerated if the label overlaps itself). The tube or vial no longer fits snugly into the holder, high spots cause stress in the glass and compromise efficient heat transfer from the holder.

Ceramic etched: Ceramic labels etch the glass and weaken it. Genevac

recommend testing new tubes for breakage before buying large numbers.

Sample capacity

Genevac uses computer aided design to ensure evaporators have maximum internal capacity within a minimal instrument footprint. High throughput evaporation is possible from an instrument taking up a small amount of bench space.

As the market leader in centrifugal evaporation, Genevac have already designed holders for the majority of applications. Please examine the lists on the following pages, to find the correct part number. Should you have a special application or unusual tube size, we can design a special holder for you. Please note there is a charge to cover design time for this service. Our experienced sales team (or international distributors) will be pleased to advise you of the options available.

Sample holder compatibility

When using sample holders for the first time, always check they are compatible for use with the evaporator. Load the holder with empty tubes, vials or plates, as appropriate, and place the holder and swing assembly on the rotor. Swing the holder by hand, fully from vertical to horizontal, to make sure that no part of the assembly touches the rotor.

Only used swings and sample holders in matched pairs.

All Genevac sample holders conform to the following design constraints: Mass within prescribed maximum limit Matched sample holder mass within a set Correct centre of gravity Good thermal conductivity Correct hole form and size tolerance to reduce risk of tube or vial breakage

under centrifugal load. Third party accessories may not be designed with these constraints in mind and should not be used unless authorised by Genevac.

Minimum order quantities

A minimum order quantity of two applies to: sample holders, swings, heat transfer plates, SampleGenie assemblies, etc.

Sample Holder Maintenance

Inspect and clean the sample holders on a regular basis or following any incident that involves glass breakage or solvent spillage.

Inspection

Visually inspect the swings and sample holders to check for any spilled solvent or debris, especially in the sample holder wells. Spilled solvent can cause debris to stick to the sample holders causing high points that stress the glassware. The most likely cause of repeat glassware breakage is a fragment from a previously broken tube which has become stuck to a sample holder well. It is therefore very important that sample holders are cleaned thoroughly so that any residual solvent or sample material is removed. Superficial surface damage, such as a scratch, does not affect the performance of a sample holder. However if deep pits or crevices begin to form, or if there is any structural damage (if any part of the sample holder is bent or deformed) do not use it. Contact Genevac Service for evaluation. Repeated use of sample holders with strong acids may cause colour fading over time; this is normal and does not affect the integrity of the sample holders.

Cleaning

Clean off any adhered dirt or debris by washing the sample holders in soapy water or using a cleaning solvent such as methanol or acetone. The sample holders should then be rinsed in clean water. Alternatively, wash in a dishwasher at 90°C. Check to make sure that all adhered dirt, debris or solvent residue is removed. Always dry the sample holders thoroughly after cleaning and never place wet sample holders in a Genevac system.

Avoiding glassware breakage

The precision of Genevac sample holders contributes, not only to their excellent heat transfer properties and superior evaporation performance; it also ensures that tubes have maximum protection from breakage during evaporation. Use only borosilicate tubes with a wall thickness greater than 1.2 mm in Genevac evaporators. Do not use old, damaged or scratched glassware. Own brand glassware from catalogue companies can be highly variable in size and quality and is not recommended for use in Genevac systems. Use high quality glassware from recognised manufacturers of international repute. If glassware is to be reused, replace all glassware every two to three months and be sure to remove every item of old glassware from circulation. For additional protection against glassware breakage, some sample holders must be used with thermal cushions. Thermal cushions are made from a filled polymer which has excellent heat transfer and cushioning properties.

SampleGenie™ comprises sample tube and sample holder technology which enables large volume samples to be dried or concentrated directly into a variety of small vials. Infinity trolley

Infinity Trolley is a modular system which is designed to accommodate a range of

Genevac evaporation equipment. The trolley provides a safe and durable support

for the evaporator, whilst occupying the minimum amount of space in the laboratory.

The lower “truck” can be pulled out to provide access to items on the lower shelf.

Approximate trolley dimensions: 740 x 733 x 835 mm (w x d x h). When truck is fully extended, add 500 mm to depth. For optional side shelf, add 260 mm to width. Note: For HT-12 systems, installation onto Infinity Trolley must be performed by a Genevac authorised engineer.

Gilson and Waters fraction collection racks

Genevac offer a range of innovative options that help interface fraction collection systems directly to the Genevac HT series evaporator. Currently Gilson 215 and Waters 2767 systems are supported. To interface with the fraction collections system, Genevac provide a simple sample holder carrier which fits directly on to the fraction collection bed. Genevac sample holders fit into the carrier to make up a complete fraction collection rack. Once fractions are collected, the sample holders can be placed directly into the Genevac evaporator with no need to label, mark or manipulate individual tubes. Sample holders can be marked to indicate their location within the fraction collection rack. There are a range of options, some use standard sample holders, some use specially designed holders which keep the spacing between tubes uniform, and reduce fraction collector reprogramming. Waters 2767 users will require the adaptor plate when using 16 mm diameter tubes. Further information, including a technical article discussing these issues in greater detail, is available upon request from your Genevac representative.
