Research Achievements* and Plans Within the context of method development major achievements involved organocatalytic me–thods, which were used for the synthesis of potentially bioactives. The preparation of natural products isolated by group natural products at the NWC department was achieved, enabling the realization of field trials. However, probes for affinity/activity based protein profiling were given main attention. The work encompassed design, synthesis, evaluation and optimization by an iterative approach with the biological partners (this also applies for MLU-partners). These studies were done within the PAKT project: Proteinmuster induzierter biologischer Prozesse.

Achievements involved

Metalfree traceless triazole formation by the Sakai-approach (ACIE 2012) Design of multi-read out probes (Ugi-probes) suitable for affinity/activity based protein

profiling (OBC 2012, Synthesis 2011, Chem. Commun. 2010)

These products will be further optimized for in vitro and for in vivo applications (e. g. CNTs).

Based on the incorporation of a labile tertiary amide moiety by the Ugi-protocol, new probes suitable as ITRAQ analogues will be studied. This basic concept for expanding possibilities of MS-proteomic tools is unprecedented in the literature.

In addition, studies on method development (e. g. catalysis of MCRs) and synthesis with the focus of target- and diversity-oriented natural product synthesis will be conducted; they will be carried out in strong interactions with the natural products unit at the NWC department.
