WHAT IS TECENTRIQ?

SELECT IMPORTANT SAFETY INFORMATION

TECENTRIQ is a prescription medicine used to treat adults with:A type of breast cancer called triple-negative breast cancer (TNBC). TECENTRIQ may be used with the medicine paclitaxel protein-bound when your breast cancer:

• has spread or cannot be removed by surgery, and • your cancer tests positive for “PD-L1”

The approval of TECENTRIQ in these patients is based on a study that measured the amount of time until patients’ disease worsened. Continued approval for this use may depend on the results of an ongoing study to confirm benefit.

It is not known if TECENTRIQ is safe and effective in children.

TECENTRIQ can cause your immune system to attack normal organs and tissues and can affect the way they work. These problems can sometimes become serious or life threatening and can lead to death.

Possible serious side effects with TECENTRIQ include, but are not limited to, lung problems (pneumonitis), liver problems (hepatitis), intestinal problems (colitis), hormone gland problems (especially the thyroid, adrenal glands, pancreas, and pituitary), problems in other organs, severe infections and severe infusion reactions.

The role of a caregiver may look different for each patient and each relationship. It’s important to have an open conversation with your loved one to know how you can assist them. The type of support they need may change throughout their treatment. If you’re reading this, you stepped up to make sure your loved one gets the care and support they need.

At the start of treatment, establish your role as caregiver with your loved one, so there is no confusion about responsibilities and expectations. In addition, it is just as important to keep yourself healthy throughout this process.

This guide provides suggestions on how to help your loved one. Examples include day-to-day support, self-care, and communication strategies. You will also find resources offering support in medical, legal, and financial matters.

As a caregiver, you play an important role in your loved one’s treatment journey. You can make a significant positive impact by helping in all facets of daily life, or simply by just being there.

Here are some traditional ways a caregiver can support their loved one:

• Helping with additional medications

• Organizing and coordinating appointments

• Keeping track of any potential side effects

• Communicating any issues with the care team

• Keeping an open dialogue between other friends and family

• Making important decisions throughout treatment

There may be more ways your loved one could need support. They may not be ready to admit they need help, but assisting with any of the following tasks could be crucial:

• Shopping and meal preparation

• Household cleaning

• Helping to use the bathroom

• Laundry

• Paying the bills

• Getting bathed, groomed, and dressed

• Transportation to various appointments

• Emotional support

• Assisting with childcare

Words might not be able to describe what your loved one is feeling right now. In addition to medical care, part of your role as a caregiver is helping them get the emotional support they need.

Emotional support

• Try to keep a positive attitude—it is beneficial for your loved ones and for you

• Spend time together and do things that remind you both that you have a relationship separate from your loved one’s diagnosis

• Encourage your loved one to engage in the activities they like as much as possible

Understanding your loved one’s treatment is a helpful step towards keeping them on track.

Information gathering

• As a caregiver, you can act as an extra set of eyes and ears. Ask important questions and gather information to share with your loved one

• Offer to help your loved one note any changes in symptoms or side effects

• Keep track of medical information and paperwork

• Find out if there are any special instructions and treatment tips that the healthcare team recommends

Taking care of yourself ensures that you’re taking the best possible care of your loved one. As you step into this role as caregiver, you will be focused on their diagnosis and treatment. While you are thinking about their health, it is just as important for you to stay healthy yourself in order to provide the care your loved one may need.

Below are some tips to help you take care of yourself during this difficult time:

• Have regular doctor checkups

• Know your limits and be realistic about the time and energy you can give

• Keep a journal to organize thoughts and create memories

• Find time to relax and do what you enjoy

• Go easy on yourself, focus on positive things you are doing for your loved one

• Consider routines that can improve your mood and energy

• Don’t be afraid to ask for help; your family or friends can be your first resource

Communication with your loved one is one of the most valuable tools you have.

The more you know about your loved one and how they’re feeling, the better prepared you’ll be to help them. Your support is valuable, and sometimes just being there to talk and listen can make a big difference.

Here are some different ways to help create a healthy dialogue:

• Ask if it’s a good time to talk

• Be supportive, listen, and don’t interrupt

• Think about what you want to say in advance

• Speak from the heart

• Remember that you can have more than one talk

• Don’t always say, “It’ll be okay”

A lot of people are dedicated to your loved one’s care, not just one doctor. Your loved one will likely have access to a combination of healthcare providers or HCPs. The HCPs listed below have specialized roles and work together to provide the best care possible. Your loved one’s care team may include some or all of these HCPs and may change over their treatment journey.

ONCOLOGY TEAM

Oncologist

An oncologist is a doctor who specializes in diagnosing and treating cancers with medicines, including chemotherapy, immunotherapy, and other therapies. They manage cancer treatment and coordinate with the entire treatment team.

Infusion Nurse

These nurses work to prepare and administer infused medications.

OTHER SPECIALIZED TEAM MEMBERS

Primary Care Physician

Sometimes known as an internist, a general practitioner, or the family doctor, the primary care physician may be the initial point of contact. They may refer patients to other specialists.

Nurse Practitioners/Physician Assistants

Both of these healthcare providers treat illnesses, provide patient education, and prescribe medications. They also manage the primary care needs of patients and their families in partnership with their physician.

Nutritionist

A nutritionist helps people manage their eating and hydration needs related to a cancer diagnosis and its treatment.

Psychologist/Therapist/Counselor

These healthcare professionals can help people learn to cope more effectively with life issues.

Home Health Aide

A home health aide is a licensed professional who assists people at home with their personal care, including bathing, dressing, and other activities of daily living. This person may also assist with cooking and other household chores.

Social Worker

A social worker helps people with a range of challenges: financial, physical, mental, emotional, and more. They can also connect people to local resources.

The healthcare provider will give your loved one TECENTRIQ into their vein through an intravenous (IV) line. TECENTRIQ is dosed once every 2 weeks, as long as they continue to respond to and tolerate the treatment.

The healthcare provider will decide how many treatments they may need and how long each infusion may take.

SUGGESTIONS ON PREPARING FOR INFUSIONS

If you’re attending infusion appointments with your loved one, here are some tips to make the time pass by more quickly and comfortably for you both.

Keep yourselves entertained A personal electronic device, book, crossword puzzle, or playing cards can go a long way in making the process go by more quickly.

Keep extra layers and/or blankets handyIt is not uncommon for people receiving infusions to feel cold during their session. Bring a warm blanket, cozy scarf, or extra jacket.

Pack plenty of food and drinks You or your loved one may get hungry and/or thirsty during the infusion. Keep a light snack and bottle of water handy, just in case. You may want to first check with the healthcare provider about eating and drinking during infusions.

What is the most important information about TECENTRIQ? TECENTRIQ can cause your immune system to attack normal organs and tissues and can affect the way they work. These problems can sometimes become serious or life threatening and can lead to death.

Call or see your healthcare provider right away if you get any symptoms of the following problems or these symptoms get worse.

TECENTRIQ can cause serious side effects, including:

• Lung problems (pneumonitis)-signs and symptoms of pneumonitis may include new or worsening cough, shortness of breath, and chest pain

• Liver problems (hepatitis)-signs and symptoms of hepatitis may include yellowing of your skin or the whites of your eyes, severe nausea or vomiting, pain on the right side of your stomach area (abdomen), drowsiness, dark urine (tea colored), bleeding or bruising more easily than normal, and feeling less hungry than usual

• Intestinal problems (colitis)-signs and symptoms of colitis may include diarrhea (loose stools) or more bowel movements than usual, blood or mucus in your stools or dark, tarry, sticky stools, and severe stomach area (abdomen) pain or tenderness

• Hormone gland problems (especially the thyroid, adrenal glands, pancreas, and pituitary)– signs and symptoms that your hormone glands are not working properly may include headaches that will not go away or unusual headaches, extreme tiredness, weight gain or weight loss, dizziness or fainting, feeling more hungry or thirsty than usual, hair loss, changes in mood or behavior (such as decreased sex drive, irritability, or forgetfulness), feeling cold, constipation, your voice gets deeper, urinating more often than usual, nausea or vomiting, and stomach area (abdomen) pain

• Problems in other organs–signs and symptoms may include severe muscle weakness, numbness or tingling in hands or feet, confusion, blurry vision, double vision, or other vision problems, changes in mood or behavior, extreme sensitivity to light, neck stiffness, eye pain or redness, skin blisters or peeling, chest pain, irregular heartbeat, shortness of breath, or swelling of the ankles

• Severe infections–signs and symptoms of infection may include fever, cough, flu-like symptoms, pain when urinating, and frequent urination or back pain

• Severe infusion reactions–signs and symptoms of infusion reactions may include chills or shaking, itching or rash, flushing, shortness of breath or wheezing, swelling of your face or lips, dizziness, fever, feeling like passing out, and back or neck pain

Getting medical treatment right away may help keep these problems from becoming more serious. Your healthcare provider may treat you with corticosteroid or hormone replacement medicines. Your healthcare provider may delay or completely stop treatment with TECENTRIQ if you have severe side effects.

Before you receive TECENTRIQ, tell your healthcare provider about all of your medical conditions, including if you:

• have immune system problems (such as Crohn’s disease, ulcerative colitis, or lupus); have had an organ transplant; have lung or breathing problems; have liver problems; have a condition that affects your nervous system (such as myasthenia gravis or Guillain-Barre syndrome); or are being treated for an infection

• are pregnant or plan to become pregnant. TECENTRIQ can harm your unborn baby. Tell your healthcare provider right away if you become pregnant or think you may be pregnant during treatment with TECENTRIQ. Females who are able to become pregnant:

° your healthcare provider should do a pregnancy test before you start treatment with TECENTRIQ

° you should use an effective method of birth control during your treatment and for at least 5 months after the last dose of TECENTRIQ

• are breastfeeding or plan to breastfeed. It is not known if TECENTRIQ passes into your breast milk. Do not breastfeed during treatment and for at least 5 months after the last dose of TECENTRIQ

Tell your healthcare provider about all the medicines you take, including prescription and over-the-counter medicines, vitamins, and herbal supplements.

The most common side effects of TECENTRIQ when used in triple-negative breast cancer with paclitaxel protein-bound include:

• hair loss• tingling or numbness

in hands or feet• feeling tired• nausea

• diarrhea• low red blood cells

(anemia)• constipation• cough

• headache• low white blood cells• vomiting• decreased appetite

TECENTRIQ may cause fertility problems in females, which may affect the ability to have children. Talk to your healthcare provider if you have concerns about fertility.

These are not all the possible side effects of TECENTRIQ. Ask your healthcare provider or pharmacist for more information about the benefits and side effects of TECENTRIQ.

To be eligible for free Genentech medicine from the Genentech Patient Foundation, insured patients who have coverage for their medicine must have exhausted all other forms of patient assistance (including TECENTRIQ Co-pay Program and support from independent co-pay assistance foundations) and must meet certain financial criteria. Uninsured patients and insured patients without coverage for their medicine must meet different financial criteria.

TECENTRIQ CO-PAY PROGRAM

TECENTRIQ Access Solutions can refer you to the TECENTRIQ Co-pay Program. It can help you with the out-of-pocket costs for your TECENTRIQ, if you’re eligible.

No physical card needed; patients simply need their Member ID.

This TECENTRIQ Co-pay Program is valid ONLY for patients with commercial insurance who have a valid prescription for a Food and Drug Administration (FDA)-approved indication of a Genentech medication. Patients using Medicare, Medicaid, or any other federal or state government program to pay for their medications are not eligible.

Under the Program, the patient will pay a co-pay. After reaching the maximum Program benefit, the patient will be responsible for all out-of-pocket costs.

All participants are responsible for reporting the receipt of all Program benefits as required by any insurer or by law. No party may seek reimbursement for all or any part of the benefit received through this Program. This Program is void where prohibited by law. Gland:

An organ that makes one or more substances, such as breast milk.

HER2:

A protein involved in normal cell growth. HER2 may be made in larger than normal amounts by some types of cancer cells, including breast and bladder. This may cause cancer cells to grow more quickly and spread to other parts of the body. Also called c-erbB-2, HER2/neu, human EGF receptor 2, and human epidermal growth factor receptor 2.

Immune system:

A collection of organs, special cells, and substances that help protect you from infections and other diseases.

Infusion:

A method of putting fluids, including drugs, into the bloodstream. Also called intravenous infusion.

PD-L1 (programmed death-ligand 1):

A protein found on the surface of many cells, including cancer cells, that can affect the immune system’s ability to target and attack cells.

Protein:

A molecule that is the building block of the cells in the body. Proteins are needed for the body to function properly.

Receptor:

A molecule inside or on the surface of a cell that binds to a specific substance and causes a specific effect in the cell.

TNBC (triple-negative breast cancer):

Describes breast cancer cells that do not have estrogen receptors, progesterone receptors, or large amounts of HER2/neu protein. Also called ER-negative, PR-negative, HER2/neu-negative breast cancer.
