We begin by proposing a biosynthe0c construc0on that enables Escherichia coli to produce an an0freeze protein, (AFP), at less than 15 degrees Celsius. This protein prevents ice crystal forma0on in the cell, which in turn allows survival at very low temperatures. We develop a switch by adap0ng the coldshock E. coli operon with AFP from a genus of fishes (Zoarces sp.) using a posi0ve feedback circuit. A very important poten0al applica0on we are interested in is the use of AFP to designing systems helping crops to avoid poten0al damage from frosts. There are other possible important applica0ons in 0ssue and organ preserva0on.
