Prantoprazole is a proton pump inhibitor drug that inhibits gastric acid secretion.Pantoprazole is used for short-term treatment of erosion and ulceration of the esophagus caused by gastroesophageal reflux disease. Initial treatment is generally of eight weeks' duration, after which another eight week course of treatment may be considered if necessary It can be used as a maintenance therapy for long term use after initial response is obtained.This medication may affect the results of certain lab tests, such as drug screenings (pantoprazole can cause a false positive for THC, the psychoactive component of cannabis [1). http://www.drugs.com/pantoprazole.htmlYou should not use this medication if you are allergic to pantoprazole or to any other benzimidazole medication such as albendazole (Albenza), or mebendazole (Vermox).Pantoprazole is not for immediate relief of heartburn symptoms.Heartburn is often confused with the first symptoms of a heart attack. Seek emergency medical attention if you have chest pain or heavy feeling, pain spreading to the arm or shoulder, nausea, sweating, and a general ill feeling.

Take pantoprazole for the full prescribed length of time. Your symptoms may improve before the condition is fully treated.Pantoprazole should not be taken together with atazanavir (Reyataz) or nelfinavir (Viracept). Tell your doctor if you are taking either of these medications to treat HIV or AIDS.Some conditions must be treated long-term with pantoprazole. Chronic use has caused stomach cancer in animal studies, but it is not known if this medication would have the same effects in humans. Talk with your doctor about your specific risk of developing stomach cancer.Long-term treatment with pantoprazole may also make it harder for your body to absorb vitamin B-12, resulting in a deficiency of this vitamin. Talk with your doctor if you need long-term pantoprazole treatment and you have concerns about vitamin B-12 deficiency.

Prevention of Nausea and Vomiting Associated With Radiotherapy, Either TotalBody Irradiation, or Single High-Dose Fraction or Daily Fractions to the Abdomen• The recommended oral dosage is one 8-mg Onlit Tablet given 3 times a day.• For total body irradiation , one 8-mg Onlit Tablet should be administered 1 to 2 hoursbefore each fraction of radiotherapy administered each day.For single high-dose fraction radiotherapy to the abdomen , one 8-mg Onlit Tablet should beadministered 1 to 2 hours before radiotherapy, with subsequent doses every 8 hours afterthe first dose for 1 to 2 days after completion of radiotherapy.For daily fractionated radiotherapy to the abdomen , one 8-mg Onlit Tablet should beadministered 1 to 2 hours before radiotherapy, with subsequent doses every 8 hours afterthe first dose for each day radiotherapy is given.4.Postoperative Nausea and Vomiting PONVThe recommended dosage is 16 mg given as two 8-mg OnlitTablets 1 hour before inductionof anesthesia.For the prevention of PONV Ondansetron can be administered orally or by intravenous orintramuscular injection.Ondansetron may be administered as a single dose of 4mg given by intramuscular or slowintravenous injection at induction of anesthesia.For treatment of established PONV a single dose of 4mg given by intramuscular or slowintravenous injection is recommended.

ConaunsPantoprazoleDomeridone – Sustained ReleasePantoprazole with Domperidone is used to reduce the amount of acid produced by the stomach, to treat or prevent conditions such as heartburn, Gastritis, ulcers and GERD. For best results, it should be taken just before a meal. It can help with the greater amount of stomach acid that is always produced. The capsules are generally offered in the composition of Pantoprazole 40mg & Domperidone 30 mg sustained release. Sustained release dosage forms are designed to release a drug at a predetermined rate in order to maintain a constant drug concentration for a specific period of time with minimum side effects. This can be achieved through a variety of formulations, including liposomes and drug-polymer conjugates (an example being hydrogels). Sustained release's definition is more akin to a "controlled release" rather than "sustained".

MECHANISM OF ACTION:

Pantoprazole is a Proton Pump Inhibitor (PPI) that suppresses the final step in gastric acid production by covalently binding to the (H+, K+)-ATPase enzyme system at the secretory surface of the gastric parietal cell. This effect leads to inhibition of both basal and stimulated gastric acid secretion irrespective of the stimulus. The binding to the (H+, K+)-ATPase results in a duration of antisecretory effect that persists longer than 24 hours for all doses tested.
