2. INTRODUCTION SLIDE NO :02 Novel approach to treat, cure or ultimately prevent a disease by changing the expression of a persons gene. What is gene therapy ? 3. GENE THERAPY SLIDE NO :03 APPROCHES Researcher says : I. Replacing a mutated gene with a healthy copy of the gene. II. Inactivating or knocking out a mutated gene. III. Introducing a new gene to help fight a disease. 4. A vector delivers the therapeutic gene into a patients target cell. The target cells become infected with the viral vector. The vectors genetic material is inserted into the target cell. Transferred gene encodes & produces proteins. The Proteins encoded by Transgene corrects the disorder. PROCEDURE How its works ? SLIDE NO :04 5. Somatic cell gene therapy Therapeutic genes transferred into the somatic cells. Eg. Introduction of genes into bone marrow cells, blood cells, skin cells etc. Will not be inherited on to later generations. At present all researches directed to correct genetic defects in somatic cells. Germ line gene therapy Therapeutic genes transferred into the germ cells. Eg. Genes introduced into eggs and sperms. It is heritable and passed on to later generations. For safety, ethical and technical reasons, it is not being attempted at present day. TYPES OF GENE 6. Two way to deliver genes : 1. Ex vivo gene therapy 2. In vivo gene therapy 7. GENE TRANSFER Ex vivo approach : Target cells are removed from the body and grown in the laboratory. The gene is then introduced into the cultured cells. Returned healthy gene into the patient body. SLIDE NO :07 8. GENE TRANSFER In vivo approach: (Direct Gene Transfer) Cloned therapeutic gene is introduced directly into the affected tissue. Specially designed vector are needed. SLIDE NO :08 9.
