Single molecule analysis provides de-
tailed information on chemical char-
acteristics or biological functions, but
the detection of individual molecules
is by no means easy. The extremely
sensitive method of laser-based fluo-
rescence analysis is thus used t o
increase the signal-to-noise ratio.
Confocal microscope with singlemolecule sensitivity
PicoQuant of Berlin, Germany sup-plies the MicroTime 200 confocal,time-resolved fluorescence micro-scope for this task. “This system usesthe time-correlated single photoncount for its data acquisition and canproduce both 2D and 3D images”explains Dr. Felix Koberling, Head ofSystem Development at PicoQuant. This makes it possible to realize avariety of the methods currently usedin fluorescence microscopy such asFCS (Fluorescence Correlation Spec-troscopy) and FRET (FluorescenceResonance Energy Transfer) as wellas so-called fluorescence lifetimeimaging. Here not only the measuredintensity but also the respective fluo-rescence lifetime is used for thevisualization and quantification inorder to analyze intracellular pro-cesses even in living cells. Its modu-
lar design means the fluorescencemicroscope is also very flexible inadjusting to different applications.(www.picoquant.de)
Maximum r epeatability thanks t odynamic digital linearization
The P-733.2CD piezo stage was thescanner system of choice for themicroscope. With a travel range of 100 x 100 µm and sub-nanometerresolution, this high-accuracy nano-positioning system matches the re-quirements of fluorescence micro-scopy perfectly.

If the sample cannot be moved because it is enclosed in an environ-mental chamber, for example, thesame positioning system can be usedto the microscope objective instead. In all cases, however, the integrateddirect-measuring capacitive sensorsallow the scanner to produce an ac-curate determination of the actualposition value. The first step is typi-cally to record the image of a sampleby scanning an area quickly before in a second step individual points of interest are analyzed in detail. Inorder to return to the exact locationof these points within a few nano-meters, an advanced digital controlalgorithm (DDL) was devised. TheDynamic Digital Linearization algo-rithm improves scanning linearity, i.e. repeatability by up to three ordersof magnitude compared to conven-tional PID (proportional, integral, deri-vative control).
The third dimension: Additionalfocus adjustment
The P-721.CLQ PIFOC® Z-drives areused for three-dimensional images. They provide millisecond responsetimes and their flexure guiding and
capacitive sensors enable very accu-rate positioning, even when the travelranges are relatively large. “PI’s piezo-based nanopositioning systems make
a decisive contribution to the fact that we can achieve very high-qualityresults with our MicroTime 200”, saysKoberling in conclusion.

The P-733 piezo-based nanoposition-ing system provides a travel range of 100 x 100 µm. The optional Dynamic Digital Linearisation (DDL) features fordigital motion controllers improves the scanning linearity by a factor of to 1000. Tracking error and phase lag arereduced to almost non noticeable values. (Photo: Physik Instrumente (PI))

The high quality of positioningsystems is made apparent indaily operation by PI software.Starting with simple commis-sioning, through convenientoperation with a graphicalinterface, to quick and simpleintegration in customized pro-grams with high performance,PI software covers all aspectsimportant to an application.
Universal Command SetSimplifies Commissioningand Programming
For uniform operation of nanoand micropositioning systems, the universal PI GeneralCommand Set (GCS) is used.GCS operation is independentof the controller or drive princi-ple used, so that several posi-tioning systems can be con-trolled together, or new sys-tems can be introduced with aminimum of programmingeffort. With GCS the develop-ment of custom applicationprograms is simplified and lessprone to errors, because thecommands for all supported
devices are identical in syntaxand function. Through the useof the GCS command set withits convenient functions, theorientation phase and applica-tion development process is
significantly accelerated. TheGCS commands are availableat the controller terminal, inmacros and in the form of auniversal driver set forLabVIEW (VIs), Windows

The two-channel C-867.260 controller
PInano™ XYZ piezo scanner mounted on a M-545.2U Microscope stage. The XYZ piezo nanopositioning stage provides <1 nm resolution and 200x200x200µm scanning range. The high stability and autolocking feature of M-545 provides significant advan-tages over other microscope stage designs when using fast piezo scan-ning stages.
Preliminary Specs M-545.2U piezomotor stages are mainly designed for automated positioning applications in mi-croscopy. The form factor of the M-545 is optimized for a low profile height of 30 mm only; the mounting pattern is compatible with many PI piezo nanoposition-ing stages.
Space Saving Piezomotors Compared to conventional mo-torized translation stages, the M-545 provides a lower profile and smaller footprint. The com-pact PILine® piezoelectric linear motors and linear encoders make both, the lead screw duct and the flanged, bulky stepper motors
employed in traditional stages obsolete. In addition, the piezo-motors are self-locking at rest and hold the stage in a stable position without generating heat.
Compatibility to PI Nanoposi-tioning and Scanning Stages A number of standard PI piezo flexure stages (PInano™ family, P-736, etc) can be mounted directly on the M-545 stage. Depending on the application, these highly specialized, ul-tra-precise nanopositioning sys-tems are available as fast XYZ scanners (for fluorescence mi-croscopy), as vertical Z position-
ers (3D imaging), or with up to 6 degrees of freedom.
Limit and Reference Switches For the protection of your equip-ment, non-contact Hall-effect limit and reference switches are in-stalled. The direction-sensing reference switch supports ad-vanced automation applications with high precision.
Advantages of PILine® Micro-positioning Systems The ultrasonic piezoceramic drives used in PILine® microposi-tioners have a number of advan-tages over classical drives:
Higher Acceleration, Speed
Smooth Motion, no Vibrations
Small Form Factor
Self-Locking when Powered Down, no Dither, no Energy Consumption
No Shafts, Gears or Other Rotating Parts
Non-Magnetic , Vacuum Compatible Drive Principle
The M-545.2U microscope stage with closed-loop ultrasonic piezo motors provides 25 x 25 mm travel range and very high stability

High Stability and Linearity Optimized for High-Resolution Microscopes The new PInano™ Cap XY and XYZ capacitive sensor piezo stages are optimized for easy integration into high resolution optical microscopes. They feature a very low profile of 0.8” (20 mm), a large aperture, and extremely fast response with subnanometer closed-loop resolution–ideal for leading-edge microscopy and imaging applications. The stage design permits a full slide to be mounted at bottom allowing the turret to be rotated without mov-ing the objective in and out. Lon-gest lifetime is guaranteed by the integrated ceramic encapsulated PICMA® piezo actuators. Due to the significantly higher humidity
resistance, the patented PICMA®
design provides up to 10 times longer life than conventional piezo actuators.
High Performance, Yet Cost Effective PInano™ Cap series piezo stages provide even higher per-formance than their piezoresistive (PR) sensor equipped cousins. Systems consist of the piezo mechanics, an advanced control-ler optimized for capacitive feed-back and software. For the high-est linearity and stability, these stages are based on direct meas-uring, non-contact capacitive sensors, a principle free of John-son noise. RF excitation circuitry in the controller further reduces sensitivity to external noise sources or DC voltage drift of electronic components that can limit the long term stability of DC signal excited sensors such as film and PR strain gauges. The proprietary servo design im-proves the motion linearity com-pared to conventional piezo con-trollers.
Stage Working Principle / Re-liability Flexures optimized with Finite Element Analysis (FEA) are employed to guide the PI nano™ series stages. FEA techniques give the design the highest pos-sible stiffness in, and perpen-dicular to, the direction of motion, and to minimize linear and angu-lar runout. Flexures allow ex-tremely high-precision motion, no matter how minute, as they are completely free of play and fric-tion. The award-winning PICMA® piezo drives are more robust than conventional piezo actuators, featuring superior lifetime and performance in both dynamic and static applications. Because guidance, actuators and sensors are all maintenance-free, these nanopositioning systems achieve outstanding levels of reliability:

P-737 PIFOC® Wellplate / Specimen-Focusing Z-StageLow-Profile, Long-Range Piezo Z Nanopositioner for Microscopy Samples
PIFOC® P-737 high-speed verti-cal positioning systems aredesigned for use with XYmicroscopy stages–OEM man-ual stages as well as aftermar-ket motorized stages.
While the XY stage positionsthe sample, the piezo-actuator-based P-737 moves the samplealong the optical axis to quicklyand precisely adjust the focus.Vertical stepping with an accu-racy in the nanometer rangetakes only a few milliseconds.
The large aperture is designedto accommodate a variety ofspecimen holders includingslides or multiwell plates.
High-Speed Z Steps for FastFocus Control and Z StackAcquisition
The immediate response of thesolid-state piezo drives enablesrapid Z-steps with typically10 to 20 times faster step &settle times than classical step-per motor drives. This leads tohigher image acquisition speedand throughput.
Closed-Loop Position Controlfor High-Precision andStability
For high stability and repeata-bility, P-737 stages areequipped with position feed-back. High-resolution, fast-responding, strain gauge sen-sors (SGS) are applied toappropriate locations on thedrive train and provide a high-bandwidth, nanometer-preci-sion position feedback signal tothe controller. The sensors areconnected in a full-bridge con-figuration to eliminate thermaldrift, and assure optimal posi-tion stability in the nanometerrange.
Excellent Guiding Accuracy
Flexures optimized with FiniteElement Analysis (FEA) areused to guide the stage. FEAtechniques are used to give thedesign the highest possiblestiffness in, and perpendicularto, the direction of motion, andto minimize linear and angularrunout. Flexures allow ex-tremely high-precision motion,no matter how minute, as theyare completely free of play andfriction.
Ceramic Insulated PiezoActuators Provide LongLifetime
Highest possible reliability isassured by the use of award-winning PICMA® multilayerpiezo actuators. PICMA® actua-tors are the only actuators onthe market with ceramic-onlyinsulation, which makes themresistant to ambient humidityand leakage-current failures. They are thus far superior toconventional actuators in relia-bility and lifetime.

With a platform diameter ofonly 10 cm the M-810 Hexapodis the most compact parallel-kinematics micropositioningsystem to date. In addition topositioning all six axes withhigh speed and accuracy, itallows the user to define thecenter of rotation (pivot point)anywhere inside or outside thesystem envelope by one sim-
ple software command. Thismakes it ideal for all complexpositioning tasks with restrict-ed space.
Extremely Compact,Great Freedom of Motion
The M-810.00 with its direct-drive torque motors andActiveDrive™ system withintegrated servo ampifiers pro-vides an increased velocity ofup to 10 mm/s for loads up to5 kg. Small and compact, theHexapod allows a large strokeof up to 40 mm (linear) and 60°(angular).
Hexapod vs. Serial KinematicsSystems
The Hexapod is driven by sixhigh-resolution actuators allconnected directly to the samemoving platform. This designprovides a high system stiff-ness and a large clear aperture.
Because of the low mass of themoving platform, positioningoperations can be performedwith far lower settling timesthan with conventional,

P-721 PIFOCs® are high-speed,piezo-driven microscope objec-tive nanofocusing/scanning
devices, providing a position-ing and scanning range of100 µm with sub-nanometerresolution and very high mo-tion of linearity up to 0.03 %.For applications, such as thetwo-photon spectroscopy wichrequires a particulary high res-olution, there are versions wichallow a free aperture of up to29 mm in diameter.PIFOCs® are also available withup to 460 µm travel (P-725p. 2-28), and for exceptionaldynamic and step performance(models P-726 p. 2-32 andP-725.SDD p. 2-30).
Superior Accuracy With Direct-Metrology Capacitive Sensors
Capacitive position feedback isused in the top-of-the-line
models. PI's proprietary capac-itive sensors measure positiondirectly and without physicalcontact. They are free of fric-tion and hysteresis, a factwhich, in combination with thepositioning resolution of wellunder 1 nm, makes it possibleto achieve very high levels oflinearity. A further advantageof direct metrology with capac-itive sensors is the high phasefidelity and the high bandwidthof up to 10 kHz.Alternatively, strain gauge sen-sor (SGS) models are available. The sensors are connected in abridge configuration to eliminatethermal drift, and assure optimalposition stability in the nanome-ter range.
Open-loop models are availablefor applications where fast res-ponse and very high resolutionare essential. Here, specifying orreporting absolute position val-ues is either not required or ishandled by external sensors, such as interferometers, a visionsystem or photodiode PSD (posi-tion sensitive detector). Thesemodels retain the inherent piezoadvantages such as high resolu-tion and speed.

The P-726 PIFOC ® Nano focus -ing system was developed toachieve the fastest possiblestepping time with the heavy ,high-numerical-aperture objec-tives used in many of today’ shigh-resolution microscopyapplications. Its extremely stiffdesign offers excellent settlingtime and scanning frequencyvalues even when objectives ofseveral hundred grams aremoved. High stiffness is
achieved with the rotationallysymmetric arrangement ofmultiple piezo drives and theoptimized design of the flexureand lever elements, whichassure the excellent guidingaccuracy and dynamics.
Furthermore, like other mem-bers of the PIFOC ® family, theP-726 is equipped with directmetrology capacitive positionsensors that allow resolutionsfar below one nanometer.
Direct Metrology withCapacitive Sensors for HighestStability and Accuracy
PI's proprietary capacitive posi-tion sensors measure the actu-al motion of the moving partrelative to the stationary base(direct metrology). Errors in thedrive train, actuator , lever armor in guiding system do notinfluence the measurements. The result is exceptional
motion linearity , higher long-term stability and a stiffer ,more-responsive servo loop, because external influencesare immediately recognized bythe sensor. Due to this sensorprinciple, the P-726 features aresolution of under 0.4 nm inclosed-loop and a linearity of0.02 %.
Simple Installation withQuickLock Thread Options
The PIFOC ® is mounted be -twe en the turret and the objective with the QuickLockthread ad apter. After threadingthe adapter into the turret, theQuickLock is affixed in thedesired position. Because thePIFOC® body need not to berotated, cable wind-up is not anissue.
Ceramic Insulated PiezoActuators Provide LongLifetime
Highest possible reliability isassured by the use of award-winning PICMA ® multilayerpiezo actuators. PICMA ® actua-tors are the only actuators onthe market with ceramic-only
insulation, which makes themresistant to ambient humidityand leakage-current failures. They are thus far superior toconventional actuators in relia-bility and lifetime.

P-725 PIFOC® nanofocus sys-tems are long-travel, high-speed, piezo-driven micro-scope objective nanofocusing/scanning devices. Despite theincreased travel ranges (up to460 µm), they are 20 % shorterthan P-721 units (p. 2-25) whileproviding sub-nanometer reso-lution. The innovative, friction-less, flexure guiding systemprovides enhanced precisionfor superior focus stability withfast response for rapid settlingand scanning.

The P-725.2CL can perform a250 µm step to 1 % accuracy inonly 25 ms (E-665.CR controller,no load) and in 50 ms with aload of 150 g.
Superior AccuracyWith Direct-MetrologyCapacitive Sensors

ment in classical micropositioningtechnology, however. Piezo ultrasonicmotors are also extremely well suitedto applications where the accuracyrequirements are not quite so demand-ing – in the range between 5 and 100 micrometers for example, whichis usually sufficient for industrialautomation and handling tasks.
PI has developed the low cost M-664KCEP dispenser drive withPILine® piezo ultrasonic motors for
Stack of 8 M-664KCEP linear actuators, shown with well plate. The integrated
ceramic piezomotor provides highspeeds of more than 200 mm/sec.this kind of application. Eight or more
of these drives stacked together can move pipettes vertically and
PI is increasingly employing PILine® piezo ultrasonic drives in its positioningstages as an alternative to motor/leadscrew designs – particularly when thestages must be small and fast. They achieve positioning accuracies of up to0.1 µm and speeds of up to several 100 mm/s.

Multi-Axis Accuracy
S-334 piezo tip/tilt mirrors /scanners provide extremelylarge deflection angles in aminiaturized package. Thesefast steering mirror systemsare based on a sophisticatedparallel-kinematics design with
two coplanar, orthogonal axesand a fixed pivot point.
Large Tip/Tilt Ranges withExcellent Motion Characteristics
The novel flexure/lever designwith minimized inertia allows
for the exceptionally large tip/tilt range of 60 mrad (50 mradin closed-loop operation, whichis equivalent to 100 mrad opti-cal beam deflection) and veryfast response in the millisecondrange. These parameters makethe system unique in the mar-ket of piezo driven tip/tilt mirrorsystems.
Sub-Microradian Resolution
In addition to the large anglesand the high dynamics theS-334 provides sub-micro-radian resolution. The integrat-ed high-resolution, full-bridgestrain gauge sensors (SGS)provide absolute position con-trol, excellent repeatability andhigh linearity, typically betterthan 0.05 % over the entire trav-el range.
Differential Drive for ImprovedStability and Dynamics
The S-334 is based on a paral-lel-kinematics design with co-planar axes and a single mov-ing platform. Two pairs of dif-ferentially-driven piezo actua-tors are employed to providethe highest dynamics and posi-tion stability over a wide tem-perature range.
Compared to stacked, (two-stage), piezo or galvo scanners, the single-platform design pro-vides several advantages:smaller package size, identical
dynamic performance in bothaxes, faster response and bet-ter linearity. It also preventspolarization rotation.
High Reliability and LongLifetime
The compact S-334 systemsare equipped with preloadedPICMA® high-performance pie-zo actuators which are integrat-ed into a sophisticated, FEA-modeled, flexure guiding sys-tem. The PICMA® actuators fea-ture cofired ceramic encapsula-tion and provide better per-formance and reliability thanconventional piezo actuators.Actuators, guidance and sen-sors are maintenance-free, notsubject to wear and offerextraordinary reliability.

Cost-Effective OEM System with Low Profile
P-713 family piezo scanners andpositioners with travel ranges of15 x 15 µm feature especiallycompact designs. Ideal applica-tions for the P-713 are high-dynamic scanning or trackingtasks such as sub-steppingmethods for enhancing imageresolution. Such tasks involvemoving to specific positions in asmall area (e. g. marked cells orCCD photosites) and from therefollowing or performing motion
with an amplitude of a fewmicrons. The resonant frequen-cy of up to over 2 kHz makes forsettling times of a few millisec-onds, even after a full-rangemove, all with closed-looprepeatability of under 5 nm.A single-axis version with simi-lar footprint is available asP-712 (see p. 2-14) and XY ver-sions with longer travel rangesare available on request.
Flexibility
The basic version of the P-713nanopositioner offers a guidingaccuracy in the motion plane of50 µrad. This value is generallysufficient for dithering andinterlacing tasks in scanningpatterns of a few microns. Formore demanding applications, the P-714 offers higher accuracyof typically < 25 µrad.
Nanometer PositionServo-Control
If servo-control is required andno external position sensor is
available, the P-713.2SL version, equipped with high-resolutionstrain gauge sensors (SGS) canprovide nanometer-range reso-lution.High-resolution, broadband, strain gauge sensors (SGS) areapplied to appropriate locationson the drive train and measurethe displacement of the movingpart of the stage relative to thebase indirectly. The SGS sen-sors assure optimum positionstability in the nanometer rangeand fast response.
Ceramic Insulated PiezoActuators Provide LongLifetime
Highest possible reliability isassured by the use of award-winning PICMA® multilayerpiezo actuators. PICMA® actua-tors are the only actuators onthe market with ceramic-onlyinsulation, which makes themresistant to ambient humidity
and leakage-current failures. They are thus far superior toconventional actuators in relia-bility and lifetime.See the “Selection Guide” forcomparison with other nanopo-sitioning systems (see p. 2-4 ff ).

The flexure-guided, lever-am-plified PiezoMove™ P-601 actu-ators provide large verticaltravel ranges up to 400 µm, fastresponse and high positioningaccuracy in a very small pack-age. With settling times of only
a few milliseconds and a reso-lution in the sub-nanometerrange they are well suited forboth static and dynamic appli-cations.
P-601 PiezoMove™ lever-ampli-fied actuators cover the rangebetween direct-driven pre-loaded piezo translators, suchas the P-840 series (seep. 1-74) and single-axis nanopo-sitioning stages, like theP-611 series (see p. 2-20). Com-pared to direct-driven piezotranslators, lever-amplified ac-tuators offer larger travelranges and much higher lateralstiffness and guiding precision.Compared to single-axis nano-positioning stages, they offersignificantly smaller sizes.PiezoMove™ lever-amplifiedactuators feature a resolution to0.2 nm and a repeatabilityto 8 nm.
OEM Actuator with IntegratedGuidance
With their highly precise, fric-tionless flexure guidance, avery high stiffness and excel-lent straightness of motion areachieved. Together with theirsmall dimensions and the cost-effective design, the P-601 leveramplified actuators are espe-cially suited for OEM applica-tions. Versions withstrain-gauge sensors (SGS) areequipped with a full bridgecircuit that is insensitive to ther-mal drift. Versions without sen-sors are also available foropen-loop applications such asin high-speed switches andpumps. In addition to the stan-dard steel models, special invarand non-magnetic versions areavailable on request.
Ceramic Insulated Piezo Actua-tors Provide Long Lifetime
Highest possible reliability is as-sured by the use of award-win-ning PICMA® multilayer piezoactuators. PICMA® actuators arethe only actuators on the mar-ket with ceramic-only insula-tion, which makes themresistant to ambient humidityand leakage-current failures. They are thus far superior toconventional actuators in relia-bility and lifetime.

P-871 transducers are uniqueclosed-loop piezo bendersbased on the open-loop PL 122to PL 140 PICMA® -series multi-layer actuators p. 1-94. Equip -ped with high-resolution posi-tion feedback sensors they pro-vide better linearity , accuracyand repeatability than otherpiezo benders on the market.P-871 bender actuators achievelonger positioning ranges thantypical piezo stack actuators,
up to 1.6 mm, while still pro-viding fast response times inthe millisecond range.
Design
These multilayer piezoelectriccomponents are manufacturedfrom ceramic layers of onlyabout 50 µm thickness. Theyfeature internal silver -palladi-um electrodes and ceramicinsulation applied in a cofiringprocess. Due to the thin layersthe operating voltage is signifi-cantly lower than for classicalparallel bimorph bender ele-ments. For ease of installation, the units come complete withthe mounting hardware, cablesand connectors.
Closed-Loop Position Controlfor Higher Accuracy
P-871s are ideal devices forscanning, positioning andbeam deflection applicationsand provide much better accu-
racy, stability and repeatabilitythan conventional open-loopactuators. The special benderdesign allows the direct appli-cation of a strain gauge sensorto the surface without the needfor a polymer insulation layerin between. The advantagesare faster response, reducedphase lag and precise positioncontrol with non-linearity of<0.5 %. The settling time for asmall-signal step (up to 1 %nominal travel) to an accuracyof better than 1 % is between10 ms (P-871.112) and 30 ms(P-871.140).
Ceramic Insulated PiezoActuators Provide LongLifetime
Highest possible reliability isassured by the use of award-winning PICMA ® multilayerpiezo actuators. PICMA ® actua-tors are the only actuators onthe market with ceramic-onlyinsulation, which makes themresistant to ambient humidityand leakage-current failures. They are thus far superior toconventional actuators in relia-bility and lifetime.
Optimum UHV Compatibility -Minimum Outgassing
The lack of polymer insulationand the high Curie temperaturemake for optimal ultra-high-
vacuum compatibility (no out-gassing / high bakeout temper-atures, up to 150 °C).
Amplifiers, Drivers &Controllers
PI offers a wide range of stan-dard amplifiers and controllersfor piezo actuators. The E-651.1S and E-651.2S desktopcontrollers and the OEM boardE-614.2BS (see p. 2-123) arespecifically designed to oper -ate P-871 bender actuators.

M-228 and M-229 series linearactuators provide a travelrange of 10, resp. 25 mm, andare equipped with high-resolu-tion stepper motors. The step-per mikes can push or pullloads up to 80 N, and providespeeds up to 5 mm/s. Modelsfeaturing gearhead /steppermotor combinations offer thesame stroke in a more compactpackage.
Cost-Effective Design,Valuable Features
The cost-effective design offersmany useful features such as anon-rotating tip, limit and ref-erence switches and a mechan-ical position display.
A spherical tip and a 3 m exten-sion cable are included in thedelivery. The more compactgearhead versions include anadditional flat tip.
Non-Rotating Tip
Compared to conventionalrotating-tip micrometer drives, the non-rotating tip designoffers several advantages:
 Elimination of torque-induced positioning errors
 Elimination of sinusoidalmotion errors
Elimination of wear at thecontact point
 Elimination of tip-angle-dependent wobble
Limit and Reference SwitchesFor the protection of yourequipment, non-contact Hall-effect limit and referenceswitches are installed. Thedirection-sensing referenceswitch supports advancedautomation applications withhigh precision.
Low Cost of OwnershipThe combination of these actu-ators with the networkableC-663 Mercury Step controller(s. p. 4-112) offers high per-formance for a very competi-tive price in both single andmulti-axis configurations.

Investigations on interactions and forces at the level of individual molecules
assist in gaining a great deal of information on chemical and mechanical
properties or biological functions. Special techniques have been developed for
this purpose, e.g. atomic force microscopes or so-called optical tweezers.
The latter can grip, guide and manipulate miniscule objects by means of a
laser beam.
Force Measurement in NanoscopicDimensions
Integration of the nanopositioning stage in the NanoTracker(Photo: JPK)
The optical tweez ers consist in
principle of a powerful laser and amicroscope (Fig. 2). A “trap” whichis movable in three dimensions canbe generated by suitably split ting the laser beam. This trap mak es it possible to virtually capture objectswith dimensions in the nanometer ormicrometer range, to hold them andto move them in a predefined waywith respect to other objects, and all
this simply by using light. Since smallpolystyrene beads are easily capturedby the beam, these are attached tothe samples to be manipulated priorto the experiment. When the laserbeam is moved, the bead tries to stayin focus. If it is prevented from doingso by the sample and consequentlymoves out of focus, one can measureand analyz e the forces generatedfrom the resulting deflection of thelaser.

Optical Tweezers with Laser: Non-Contact and Three-DimensionalMeasurement
Since the tec hnique uses only theradiation pressure of the light, thereis no direct contact with the sample. The main advantage of the opticaltweezers over atomic force micro-scopes is that observations arepossible not only in the direction of the z-axis, but in all three spatialdimensions and experiments can becarried out interactively.
JPK Instruments, of Berlin, Germanyhas brought to market optical tweez -ers ready for series production whichprovide a time-resolved detec tion ofindividual molecules and directly support many of the mathematicallycomplex analytical methods thanksto their extensive software. The Nano -

data from the trap steering unit, i.e.to evaluate the laser beam deflection. “This is not suf ficient for quite a number of applications, however”explains Dr . Joost van Mameren (Fig. 4), an application scientist at JPK.“If an analysis requires motion at constant speed, for example, we inte-grate a piezo-based three-axis nano -positioning system into our Nano -Tracker. This will prevent smallinterfering signals, which could arisewith pure trap steering due to themotion caused when the optics aredisplaced. Furthermore, the position-ing system pro vides the option ofshifting the focus in the direction ofthe Z-axis, if required, in real-time.One should avoid manipulating theoptics for this purpose, if at all possi-ble, because measuring errors causedby interfering signals would then beinevitable. Moreover, the nanoposi-tioning system enables the calibra-

tion of the optical tweezers to providea higher positioning accuracy.” JPKchose the P-561.3CD nanopositioningsystem of the PIMars series from PI.“The parallel kine matics system oper-ates with repeatability in the nanome-ter range at response times belowone milli second, and the travel rangeof 100 x 100 x 100 µm matches therequirements of our application perfectly. Moreover, it proves to bevery reliable and convinces with itslong service life”, Dr. van Mamerencontinues.
Parallel Kinematics and DirectMetrology
The combined use of parallel kine-matics and parallel direct metrologyin the PIMar s system provides themulti-axis nanopositioning systemwith simultaneous determination ofall degrees of freedom with respectto a common fixed reference. An
unintended crosstalk of the motion(e.g. as a result of an external force)into a dif ferent axis can thus bedetected and actively compensatedin real-time. This can k eep the deviation from the trajectory down tounder a few nanometer s, even indynamic operation. “The capacitivesensors integrated in the nanoposi-tioning system make a decisive con-tribution to this effect. They providedirect and contact-free measurementof the position. Neither friction norhysteresis affect the measurement, and this, combined with the positionresolution of far below one nano -meter, means outstanding values forlinearity can be achieved”, explainsDr. van Mameren further.

The continuing miniaturization in medical engineering places ever in-
creasing demands on the components. Piezo drives are the solution for many
motion control applications; the piezo effect efficiently generates fast
and precise motion while requiring very little space.

Efficient Aerosol Generation with
Piezo Elements
The treatment of respiratory diseasesoften involves medications beingadministered directly with atomizers. One method of atomization is togenerate very fine droplets with theaid of ultrasonic piezo ceramics.
Specially shaped piezo disks excite a stainless steel diaphragm withseveral thousand holes to executeultrasonic oscillations at more than100 kilohertz: this produces par-ticularly homogenous aerosols, allowing the medications to be dosedaccurately and administered in amore targeted way.

Piezo elements from PI Ceramic. The broad spectrum of standard products issupplemented by custom engineered products, with the shortest-possible time-to-market
Moreover, the piezo technologyreduces the time required to atomizemedications by up to 50 % comparedto conventional systems increasingthe quality of life for patients withchronic diseases.
Piezo ceramics meet the specialhygiene requirements in medicalengineering; the aerosol generatorscan be sterilized at high tem-peratures, even in autoclaves. Theultrasonic operation is soundless for humans, and the low powerconsumption of the piezo com-ponents allows battery operation.
Piezoelectric Drives in Microfluidics
Piezo-driven microdispensers, i.e.micropumps and microvalves, candose minute volumes down to themicroliter range with very high ac-curacy. Disk-shaped piezo elementsmounted directly onto a metal micro-diaphragm provide the highly dy-namic drive for precision miniatureliquid or gas pumps. Due to theseparation of drive and mediumthrough the diaphragm, interferencewith the pumped media is completelyavoided.
Lab-on-a-chip applications are madepossible by the minute dimensions.

M-850 Hexapod Advances Research in Dental BiomechanicsFrom Christoph Bourauel and Ludger
Keilig–Department for Orthodontics at
the Rheinischen Friedrich-Wilhelms-
Universität, Bonn.
Dental biomechanics deals with theinteractions between dental materi-als, treatment instruments or den-tures and the reaction of teeth, bio-logical tissues, etc. to mechanicalstresses. A wide spectrum of forcesystems occur here with masticatoryforces exerting loads to 380 N andtorques to several Nm.
At the same time, movements of several orders of magnitude areinvolved: orthodontic equipment canchange the position of teeth by up toseveral mm, whereas—during masti-cation—teeth are deflected by lessthan 100 µm and implants by as littleas a few microns or less. These com-binations of small forces with largedeflections, on the one hand, andlarge forces and extremely smalldeflections on the other , represent achallenge with respect to the biome-chanical metrology.
To deal with this challenge, theDental Clinic of the University ofBonn designed the HexMeS (Hexa-pod Measuring System) based on theM-850.50 Hexapod. The ability tomove in 6 degrees of freedom andthe combination of small dimensions, very high stiffness and resolution ofless than 1 µm (1 arcsec) were the keyreasons for choosing the M-850 sys-tem.
HexMeS also features two 6-compo-nent force/torque sensors for theHexapod with measuring ranges of12 N (120 Nmm) and 130 N (10 Nm)respectively and an optical detectionsystem equipped with 3 CCD cam-eras.
Because of its high stiffness (100N/µm), sample deflections can usual-ly be calculated directly from theHexapod motion.
For high-load testing—simulations of mastication in the 100 N range—the optical portion of the HexMeS is used. It resolves sample deflectionsto 0.7 µm / 0.2 arcsec.
The M-850-based HexMeS currentlyrepresents one of the most flexiblemeasuring systems in the field ofdental biomechanics. Its efficiencyand the broad spectrum of its appli-cation have been demonstrated in awhole series of experimental investi-gations into dental implants, tele-scope crowns and orthodontic pros-theses.

In medical engineering, modern PILine® ultrasonic piezo motor drives are
opening up applications which were impossible using classic electric motor
leadscrew systems. Due to the piezoelectric effect and the direct creation of
linear motion, PILine® drives are not only faster, lighter and more compact
than conventional motorized drives, but they can also be made non-magnetic.
They achieve resolutions of 20 nm (0.02 µm) and v elocities of up to 1 m/s.
Their travel range is basically unlimited, and they are available in a number of
different integration levels to match the desired (OEM) application. Medical
engineering provides an up-to-date example.
OCT: Piezo Motors in Medical DesignUltrasonic piezo linear drives—new application in non-invasive medical technology
Piezo t echnologyenables fast, com-
pact, high-resolutionOCT scanner
In addition to the clas-sic, invasive, punchbiopsy technique, there
are a number of only par-tially satisfactory non-invasive
procedures in clinical and cos-metic research for properly cate-gorizing skin changes. Those basedon ultrasound do not provide goodresolution, and confocal microscopycannot penetrate sufficiently belowthe epidermis. Now, however, thereis a practical alternative: the newSkinDex scanner – developed by ISISOptronics in Mannheim, Germany –combines the advantages of ultra-sound and confocal microscopy.

The SkinDex scanner is based on the technology of optical coherencetomography (OCT) and examines thetissue on and under the skin surfacenon-invasively. The results obtainedare extraordinary. The informationcontained in the 2-D and 3-D sectionalimages is comparable to that of a histological examination.
OCT uses the basic transparency ofskin together with the interferencefringes obtainable with white light. The optical paths are made up ofoptical fibers.
Exact positioning for precise results
To enable creation of 2- and 3-dimen-sional images from interferencepatterns, the optical fibers must bemoved both axially and laterallyduring the scan. This task requirespositioners capable of the highestprecision. Ultimately, it is the perfor-mance of the drives which determinethe system resolution and hence thequality of the images.
A PILine® P-661 OEM motor is usedto position the reference-arm mirror(depth parameter). This motor waschosen primarily because of its com-pact design and, considering its size, its high force capacity of 2 newtons(0.5 lbf). Total travel is 2 mm, the po-sition resolution in this application30 nm (0.03 µm, 1.18 micro-inch).
As the images are recorded sequen-tially, the high speed and excellentdynamic response of the drive is agreat advantage. As a result, theSkinDex needs only a few seconds to generate its highly informativeimages. The lateral motions of theoptical fibers in the sensing arm executing the surface scan are alsoperformed by a PI drive.
In this case it is a PIHera® P-622.2CD,
a flexure-guided, 2-axis, piezo nano-
positioning system, which provides
a resolution of 1 nm (0.001 µm, 0.04
µ-inch) and covers an area of 250 x
250 µm. Piezo-motor drives have thus
again contributed to an innovation
from which many people will benefit
in the future.

White-light interferometry is the basis of OCT. Using optical fibers, light isdivided into a sensing and a referencebeam. After being reflected by the target(i.e., a cutaneous structure) and thereference mirror respectively, the beamsare recombined and enter the detector. 3D Structuring in Nanotechnology
A novel 3D laser lithography system based on PI nanopositioning technology is
now available from Nanoscribe GmbH (www.nanoscribe.de).The new system
allows for the first time the fully automated production of complex three-dimen-
sional microstructures and nanostructures using photosensitive materials.
Typical fields of application for thenew technology are, for example, the production of three-dimensionalmatrices for cellular biology, themanufacture of micro-optical com-ponents or photonic crystals and alsoas a rapid-prototyping instrument formicrofluidic and nanofluidic systemsand their production in small batches. The desired structures can be designedand then imported using any CAD soft-ware which supports the DXF format.
Track Widths to 150 nm and Below
The operating principle of the newlithography process, which is suitablefor all commercially available photo-resists, is easily understood: Ultra-short laser pulses are strongly focusedinto the material, which is thenexposed by means of a nonlinearoptical process in the focal point. Likea pen that is moved in three dimen-sions, the laser beam writes on thematerial following arbitrary paths. It is thus possible to achieve trackwidths from several micrometersdown to 150 nm. Two-dimensional 2D structuring or 21/2D structuringwith surface profiles are, of course, also possible and have a resolutionwhich is significantly higher than thatallowed by conventional instrumentsup to now.

4 nanometer repeatability. Its con-struction as a parallel-kinematics multi-axis system contributes to thehigh positioning accuracy. All piezoactuators act on a central platform so all axes behave with identicaldynamics. One “slower” axis, mostlyunproblematic for linear scans, would
have detrimental effects here. More-over, the high-resolution capacitivesensors can register any deviationfrom the commanded path in 3Dspace in real time. This type of position measurement directly at the moving platform against a fixedreference (parallel metrology) allowsthe immediate determination andactive compensation for axis crosstalkand lateral runout. PIFOC® Revisited: P-737 Piezo-Z Microscopy Specimen Scanning Stage
PIFOC® P-737 high-speed verticalpositioning systems are designed forintegration into motorized XY micros-copy stages of leading manufacturerssuch as Märzhäuser. While the XYstage positions the sample, the piezoflexure-based P-737 moves thesample along the optical axis to adjustthe focus quickly and precisely.Vertical stepping with nanometer pre-cision takes only a few milliseconds. The large aperture is designed toaccommodate a variety of specimenholders including slides or multiwellplates.
High-speed Z-steps for fast focuscontrol and imaging
The immediate response of the solid-state piezo drives enables rapid Z-steps with typically 10-to-20-times-faster step-and-settle than classicalstepper motor drives. This leads tosignificantly higher image acquisitionspeed and throughput. The P-737 isavailable with travel ranges of 100 or250 µm and offers a choice of feed-back sensors between strain gauge ornon-contact, capacitive positionsensors, depending on the accuracyrequirements.
The P-737 together with the E-625 piezo-controller, offers a cost-effectivesystem for high-speed, high-resolution positioning of microscopyspecimens. The controller features a choice of a 20-bit digital interface ora broadband analog interface for the target position.
The capacitive-sensor-equipped version can also be operated with thesophisticated E-753 digital servo-controller. The main advantage of thiscontroller is its adaptability to changing load conditions on the piezostage. No matter what load is applied, the system can always provide anoptimum combination of speed, settling time and resolution.

PI is the inventor of high-speedPiezo-Z objective steppers. Todaythe term PIFOC® is practicallysynonymous with all high-reso-lution vertical drives for micro-scope objectives. Depending onthe application, it can be advan-tageous to adjust the sampleinstead of the objective. While nosample stage can beat the speedof the fastest PIFOC® objectivepositioners (due to the stiffer andmore compact design), the P-737stage allows for convenient Z-stack imaging with multipleobjectives and very high through-put. Im both cases the effectremains the same: the focal planemoves through the sample. Thisis why the P-737 is also marketedunder the PIFOC® trademark.
