Copan CPA200™ Semi-Automated Card Puncher Version 2 Performance

IntroductionThe CPA200™ is a semi-automated punch systemdesigned to process human blood or saliva samples,spotted on filter paper or cards, in forensic/geneticslaboratories. CPA200™ offers punching accuracy anddelivering of each punch into micro-wells plates,flexibility to process different card shapes anddimensions, samples traceability using barcode readerand camera control and integration into laboratory SOPand LIMS.

After releasing CPA200™ in 2015, Copan NewLabEngineering developed the CPA200™ Version 2 (V2),currently available on the market. The CPA200™ V2 wasdeveloped after collecting all the requirements receivedfrom North America, Europe and Asia end-users. Newfeatures were added like extra punch of alreadyprocessed cards, extra cleaning strike, resume protocolafter interruption, customizable number of cleaningstrikes, plate layout cloning, automatic export ofreports, renewed graphic software interface, speedperformance improvement and better compatibility withdifferent brands of 96-wells plates.

ObjectiveThe objective of this study was to demonstratethat the CPA200™ V2 can generate punches fromsaliva or blood samples deposited on cards orcollector devices, to obtain human DNA profilingwhile preventing cross-contaminations and DNAcarryover between wells/samples.

MethodsIn this study, saliva and blood samples from differentdonors were spotted on Whatman® FTA® cards(FTAC), Copan NUCLEIC-CARD™ Color (NCC) for saliva,Copan NUCLEIC-CARD™ White (NCW) for blood andBode Buccal DNA Collector (BBC) for saliva. Five 96-wells microplates were filled with punches performed bythe CPA200™ following a ½ zebra + ½ checkerboardpattern, composed of negative control punches fromblank cards alternated with punches from (1) NCC withsaliva; (2) indicating FTAC with saliva; (3) NCW withblood, (4) non-indicating FTAC with blood, andtransferred into empty wells; (5) BBC with salivatransferred into wells prefilled with Prep-n-Go™ Buffer.One cleaning punch was done from blank cardsbetween consecutive samples to clean the puncher andavoid cross-contaminations. All punches were amplifiedwith GlobalFiler® Express PCR Amplification Kit onVeriti® 96-well Thermal Cycler and fragments wereanalyzed on Applied Biosystems™ 3500 GeneticAnalyzer. Data was analyzed with GeneMapper ID-Xv1.4.

Results

When performing a zebra-checkerboard plate patternwith the CPA200™ V2 instrument, 100% of completeand reproducible DNA profiles were obtained frompunches obtained from the three different sample cardtypes spotted with saliva and blood samples. No cross-contamination or foreign alleles were detected in thesamples. Additionally, no random carryovercontamination was detected in any of the negativecontrol punch.
