Summary
Lesson Plans and lab work is now available on my teacher pay teachersite, https://www.teacherspayteachers.com/Product/3D-Printable-DNA-RNA-Manipulative-Lab-1-3452084
This is a set of restriction enzymes to expand upon my DNA/RNAManipulative set. http://www.thingiverse.com/thing:1259352
These 4 restriction enzyme blocks can be used to model how they work, and how gene splicing / recumbent DNA works. The blocks also show howmany restriction enzymes are palindromes with sticky ends.
Your students will enjoy simulating genetic engineering with this expansionset.
This features EcoRI, BamHI, Hindll, and Kpnl Restriction enzymes. Thereare over 600 of these restriction enzymes used in gene splicing.
If you want to make your own restriction enzymes for gene editing, Ipunished an expansion set here http://www.thingiverse.com/thing:1321230
A partial list of restriction enzymes can be found here, https://en.wikipedia.org/wiki/List\_of\_restriction\_enzyme\_cutting\_sites
If you want other restriction enzymes made, I can make them oncommission. PM me or contact me via [email protected] for details.

Printer Brand: LulzBot
Printer: TAZ 5
Rafts: Doesn't Matter
Supports: Doesn't Matter
Resolution: .3-.4 mm
Infill: 5%
Notes:
I printed them in clear filament so they can lay over a DNA sequence andyou can find where they match.
Break all sharp edges. If you can't see the lettering, use a permanentmarker to highlight the edge of the letters.
Print 2 of each enzyme so you can model the cutting pattern and cleavedfragments.
Post-Printing
Clean up
After printing break any sharp edges. I found a emery board works greatfor rough edges. If you print clear, use a permanent marker to highlightthe edge of the lettering.
How I Designed This
Design and printing
These were designed in TinkerCAD based on the previous DNA / RNA file.
They were designed to fit as one piece so you can see how they work, andsave time building them with the other set.
If you want to expand and make your own, DL my expansion set here http://www.thingiverse.com/thing:1321230
Custom Section
Project: Gene Splicing and Restriction Enzymes
This Is to expand upon the DNA / RNA manipulative set. Your students willsee how gene splicing, and how restriction enzymes are used to splice newgenes into an organism.
You can visit https://en.wikipedia.org/wiki/List\_of\_restriction\_enzyme\_cutting\_sites for a list of restriction enzymesand their cutting sites. You can make your own with this expansion set http://www.thingiverse.com/thing:1321230
Objectives
Students will understand what genetic modification is.Students will model how restriction enzymes work.Students will model gene splicing.Students will demonstrate how to alter DNA.Students will discuss the ethics and implications of genetic modification.Students will discuss genetically modified organisms (GMO's) andagriculture.Students will show their knowledge of DNA and RNA.Students will model how mecumbant DNA works.Students will show sticky ends, fragments, and cleavage of DNA.
Audiences
This is for AP Biology, College Bio, Medical students, and home schoolstudents. This can also be used by genetic councilors, doctors, and medicalstudents.
Preparation
Students should learn about DNA and RNA.Students should understand vectors and the concepts of gene splicing.Students should know about mutations.Students should know what paladrones are, and how they appear in DNA.Students should know what an enzyme does.Students should know the base pairs, what direction (5- 3) DNA is read.

To use these restriction enzymes, you should print up 2 of them along withthe DNA / RNA set.
A activity package will soon be available, and sold on my Teacher PayTeacher page. Printed sets of restriction enzymes will be for sale on myetsy page.
Steps!
a. Build a model of DNA matching the restriction enzyme.b. Model how the restriction enzyme cuts the DNA along specific basepairs.c. Show where the sticky ends are.d. Use the enzymes to insert a new DNA sequence.e. Discuss gene splicing. Show organisms that have undergone thisprocedure.f. Discuss the ethics on gene splicing.
Results
To demonstrate knowledge of this, have the students work in small groupsto demonstrate the process or teach it to others. You can have the students make a stop-motion video of the process ofgene splicing. You can also have them show what they have learned by modeling DNAreplication, RNA transcription, protein synthesis, and gene splicing toproduce new proteins. You can tie it to pop science by having them watch sections of the filmJurassic Park / Jurassic World that discuss what they did to create thedinosaurs. With that have the students model what the scientist in the filmdid.
