Install the Image Studio SoftwareIMPORTANT: You must be logged in to Windows® Operating System using an Administra-tor type account. If necessary, log out and log back in using an Administrator account.

Windows 7, XP, or Windows® Vista Operating Systems:

Installing via download:

1) Download and double-click on the Win_Image_StudioLite-_Installer_3.1.x.exe file on

your computer

2) Click Next and follow the instructions in Setup Wizard

Mac OS X® 10.6 (Snow Leopard), 10.7 (Lion), or 10.8 (Mountain Lion):

Installing via download:

1) Go to your Mac® System Preferences and click Security & Privacy to open the Security

& Privacy window.

2) Click the lock in the bottom left corner of the window to allow editing, if necessary.

3) Under the GeneralTab, click Mac App Store and identified developers under the Allow applications downloaded from option.

4) Download, and double-click on the MacImageStudioLite-3.1.x.dmg file on your com-

puter.

a. Click Agree to accept the license agreement.

b. To install, drag the Image Studio icon into the Applications folder

c. Open the Image Studio Application by double-clicking on the Image Studio icon.

d. If a window appears that says “ImageStudio.app is an application downloaded

from the internet. Are you sure you want to open it?” click Open.

Start Image Studio Double-click the Image Studio icon on the desktop to start Image Studio Lite.

Create A New Work AreaThe first time you start the software you will need to cre-

ate the first Work Area, a folder on the hard drive or net-

work where all of the images, analyses and settings are

stored.

1) Click Create New in the Set Active Work Area dialog

box to browse to a folder on the hard drive or network

to use as the Work Area.

2) Click the New Folder icon to create a new folder for

the Work Area. This folder will now appear in the

Available Work Areas window each time the software

is opened. Select the folder and click Save to set it as

the Work Area. (To remove a Work Area from the win-dow, select it and click Remove from List. Removing the folder from the window does not delete the folder or its contents.)

NOTE: Each user should create their own Work Area, as previous settings for instru-ment, analysis, image display, etc. from the last session will be applied to the next session in the same Work Area.

Image ImportTutorial images are available at www.licor.com/ISLite. Download the tutorial files. NOTE: Image Studio Lite supports JPG, TIFF, and PNG files from many different sources.If your image does not appear to import correctly, please contact us about the imageusing the submission form found at http://www.licor.com/contact/contact_biosupport.jsp.Select SOFTWARE in the “Product Interests” box and attach a sample file for our review.

1) Click the Application Menu Button and hover over Import. Hover over the Image Stu-

dio icon and click Zip File. Select the Acquisitions_ISLite.Zip file and click Open. The tu-

torial files will import and appear listed in the Images Table at the bottom of the window.

NOTE: Images from the Odyssey® Application Software (versions 1.x, 2.x, or 3.x) can be imported into Image Studio software for analysis by clicking Import > Odyssey. Images from the Pearl Imager software can be imported by clicking Import > Pearl. Other non-LI-COR® image files (TIFF, PNG, or JPEG) can be imported by clicking Import > Other Image.

2) Double-click the ‘9999999_01’ image in the

Images table.

3) The image appears in the display window.

Image Display1) After importing an image, in the Image tab, click Choose and select 800 channel.

Click the view with the best display.

2) The Adjust Display dialog will open to allow for further adjustments as needed.

3) Click Signal at the bottom left to adjust the maximum point on the curve in the

Lookup Tables; Background to adjust the minimum point; or Midtones to adjust the

K value. Click Dimmer or Brighter to change the visual appearance of the image.

When finished, click Done.

4) Additional Brightness/Contrast enhancements may be implemented

using the adjustable sliders on the Image LUTs Tab on the right side

of the window.

Data AnalysisAdd and Manipulate Shapes Manually

1) Click the Shapes tab.

2) To use the Auto Add Rectangle or Auto Add Ellipse tools, only one channel can be

selected. Click the 700 channel’s Don’t show this channel thumbnail in the Image LUTs, leaving only the 800 channel displayed.

3) Click Auto Add Rectangle to add a rectangle to the image or Auto Add Ellipse to add

an ellipse. Click in the center of a feature to place the rectangle or ellipse around it.

After adding the desired shapes, click Select or press the Esc key on your keyboard

to return the cursor to the selection tool.

4) To move or resize a shape, click once on the shape to select. Dotted lines indicate

the shape is selected.

a. To move the shape, drag the four-pointed cursor within the selection shape.

b. To resize the shape, hover over any corner or side of the shape. Drag the yellow

dotted line to resize.

Subtract Background

1) Click the first button in the Background group and

select Median from the drop-down menu to subtract

the median value of pixels in the background segment.

2) Set the Border Width to 3 in the pop-up window and select

the segments to be used for the background calculation.

3) Click Save.

7 Export Data1) Click the Shapes tab in lower left corner (see figure at right),

then click the Table ribbon tab (see figure below).

2) Click Export File or Launch Spreadsheet in the Data group.

3) Name the file.

4) Click Save.

5) A report containing the analysis information is created. The Signal column contains

the background corrected values that should be used for further analysis.

Export Image1) In the Images tab at lower left (as shown in figure below), click on the image to be

exported. Click the Image Studio icon and hover over Export to view the Export Image and analysis data menu.

2) To save the image to a graphics file, click

Single Image View to view a choice of Current Image, Selected Images in Images Table, or

Color Bar Only. Choose the export settings

and click Save.

Create a Report1) Click the Image Studio Application Button and select

Save As Pdf.

2) In the Save As PDF dialog box, select Shapes Lab Bookand click Save to save the default report for the current

image.

3) To create a custom report, click Add Lab Book ... to open

the Print Format Builder menu. Follow the example at

right to create a custom report. Click Save.

a. Type a report name in the Lab Book Name: field.

b. Items that are available to add to the report appear

under Available in the right-hand column.

Select an item and click Add to move it to

the In Use column on the left.

c. Click Save to include the items in the In Usecolumn in the report.

d. In the Print menu, click Save to create the

custom report for the current image.
