Hon’ble Vice Chancellor Message

Most Rev. (Prof.) R.B. LalVice Chancellor, SHIATS

A life of service calls for sacrifice. Farmers, who toil day and night to grow food for the nation, are the living examples of this kind of life. Allahabad Agriculture Institute was founded to serve the servers, the unsung heroes—the farmers. Now when we are a deemed university, we take our mission to continue to serve the farmers and the nation through our education, research and extension much more seriously.

I am happy to note that our teachers, researchers and scientists are working with the same spirit, which made this institution a beacon of excellence in the state of Uttar Pradesh—this newsletter is a testimony to their hard work and spirit of service.

Inside this newsletter you will find the details of work done by our staff and students. Whether it is research on antimicrobial resistance pattern or new approaches in the field of biofertilizers, our students and staff have proved to be second to none. Creativity and innovation is seen in all our departments from Department of Agricultural Extension and Communication to Department of Home Science to departments of engineering.

Lectures, seminars, conferences constitute the life lines of a university. Various departments organised special lectures and seminars. Our staff and students attended conferences nationally as well as internationally and brought wealth of knowledge and experience to the university. In this session we were especially blessed to have Rev. Dr Edwin Higginbottom, the grand nephew of our founder Dr Sam Higginbottom, who in his unassuming manner went around and encouraged a host of people on the campus.

We had a number of distinguished guests and academician visiting us in this session. From Prof. Rebecca Reynolds of Queen's Medical Research Institute, UK to Mr Abhinn Shyam Gupta, Olympian and Arjun awardee (Badminton), we had the honour of hosting a range of eminent personalities.

Sports are an integral part of our university life. And, I am glad to see that our annual athletic meet generates a lot of interest among students.

I thank God Almighty for helping us excel in various fields and I pray that in the coming sessions we will continue to pursue excellence in personal and professional fields and bring glory to this historic institution to God, who taught us to serve and offer our lives as a sacrifice for our neighbours.

“For even the Son of Man came not to be served but to serve, and to give his

life as a ransom for many”

(Mark 10: 45)

Fusariumoxysporium and Rhizoctoniasolani were 97-100% with Evalue 0.0 and 99% sequence coverage with 1600 query length.

lA 72% fungal incidence was reported from rhizospher ic soi l of p igeon pea crop (Cajanuscajan). Fungal isolates included Penicillium spp., Helminthosporium spp., ALternaria spp., Rhizopus spp., Curvularia spp., Fusarium spp., Trichoderma spp. Aspergillus spp., Mucor spp., and Cladosporium spp. Maximum incidence was reported for Alternaria spp. and Cladosporium spp. (13.8%) followed by Fusarium spp. and Aspergillus spp. Molecular similariry 97-100% was obtained for some of the isolates Penicilliumfuniculosum, Aspergillusflavus, F u s a r i u m c a m p t o c e r a s , C l a d o s p o r i u m s p h a e r o s p e r m u m , Helminthosporium sp., Curvularialunata and Alternariaalternata.

lFungal incidence of 58% was determined from rhizospheric soil of Pisumsativum (Pea) crop. Maximum incidence was observed for Aspergillus spp. (25%), Mucor spp. (17.85%), Fusarium spp. and Trichderma spp. (14.28%), followed by Alternaria spp. (10.7%). Lowest incidence was found for Penicilliumspp, Verticillium spp. Circinella spp., Rhizopus spp. and Rhizoctonia spp. (3.57%).

lPrevalence of Trichophytonmentagrophytes, T r i c h o p h y t o n v e r r u c o s u m , Micorsporumgypseum, Microsporumcanis, Epidermophytonfloccosum was determined from suspected patients with clinical symptoms of dermatophytosis.

Antimicrobial Resistance PatternlSynergism was demonstrated between the drugs

Ampicillin and Ciprofloxacin against Methicillin sensitive Staphylococcus aureus and Methicillin resistant Staphylococcus aureusby various drug interaction studies i.e. Double Disc diffusion, Chequer board technique and Time kill studies. Fractional Inhibitory Concentration (FIC) index value was determined as 1.26, 0.257 for Methicillin resistant Staphylococcus aureus isolates, and 0.265 and 0.156 for Methicillin sensitive Staphylococcus aureus. Further a ? 2log decrease in population was indicated by the time kill curve for all the isolates.

Natural Antibacterial AgentslAntifungal activity of various extracts of and

Pongamiapinnata were demonstrated against f i v e i s o l a t e s o f d e r m a t o p h y t e s T r i c h o p h y t o n m e n t a g r o p h y t e s , T r i c h o p h y t o n v e r r u c o s u m , Micorsporumgypseum, Microsporumcanis and Epidermophytonfloccosum. Ethanolic extract of Trachyspermumammi was more effective even as compared to antifungal agent Fluconazole.

lA wide range of antibacterial activity was exhibited by various leaf extracts (Ethanol, m e t h a n o l a n d e t h y l a c e t a t e ) o f Trigonellafoenumgraecum and Murrayakoenigii against both Gram positive and gram negative bacterial pathogens. Ethanolic extractof Trigonellafoenumgraecum was most potent. In the case of Murrayakoeningii Ethyl acetate extract showed highest potency. Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) values were obtained with Trigonellafoenumgraecum for Staphylococcus aureus as 64 µl/ml and 128µl/ml. With Murrayakoenngii lowest MIC and MBC values were obtained for Staphylococcus aureus, Listeria monocytogenes, Escherichia coli, and Pseudomonas aeruginosa corresponding to 64 µl/ml.

lA nt i b a c te r i a l a c t i v i t y o f ex t ra c t s o f Boerhaaviadiffusa and Cuscutareflexa were revealed against Staphylococcus aureus, Pseudomonas aeruginosa and Bacillus subtilis. Methanolic extract of Boerhaaviadiffusa was most potent. In case of Cuscutareflexaethanolic extract was most potent. Against fungal pathogens activity of Boerhaaviadiffusa (Petroleum ether extract) and Cuscutareflxa (hot water extract) were found effective against Candida albicans and Fusarium sp. No activity was demonstrated against Alternariasolani and Penicilliumcirtrum.

lAntimicrobial potential of fruiting body of Pleurotusostreatus and Pleurotussajor-cajuwas evident against selected bacterial pathogens (Bacillus cereus, Staphylococcus aureus, Klebsiella pneumonia, Escherichia coli, Vibrio cholera and Proteus vulgaris). Bacillus cereus, Proteus vulgaris, Escherichia coli and Vibrio cholerae were found more susceptible. The most effective concentrations of methanolic and ethanolic extracts of Pleurotusostreatus and Pleurotussajorcaju were 80, 90 and 100%.

The isolate gave maximum phosphate solubilizing efficiency (90.44%) in 5 days and Indole acetic acid production was recorded in the presence of L-Tryptophan (115 µg/ml). Antagonistic activity of the isolate was demonstrated against fungal pathogen Fusariumoxysporum.

ProbioticslProduction and recovery of Mannitol was

achieved using Lactobacillus brevisthrough fermentative process. Maximum production of mannitol (4.125g/l) was achieved through HPLC analysis at temperature 30°C, pH 5.5 and glucose: fructose ratio 1.5:20 after 96 h of incubation in shaking incubator at 200 rpm.

Microbial EnzymeslCellulase producing bacteria were isolated from

various sites of Allahabad and identified as Bacillus cereus, Bacilluspantothenticum, Bacillus insolitus, Bacillus alvei, Bacillus firmus, Bacillus alvei, Bacillus alcalophilus, and Bacillus subtilis. The parameters giving maximum cellulose production were 30°C temperature, pH 7.5, 1% lactose, 1% peptone after 72h incubation period. Crude enzyme was partially purified by Ammonium sulphate precipitation, dialysis and DEAE- cellulose ion exchange chromatography. The total activity of crude enzyme and purified enzyme were 4537.2 U/60 ml and 323.52 U/8 ml, respectively. Total protein content and purified content were 1633.17mg/ml and 77.26 mg/ml respectively. Enzymatic activities were found maximum at pH 7 (5.86 U/ml) and at 50°C (40.12U/ml). Compatibility with different detergent brands showed maximum for Tide detergent (81.78U/ml).

BioremediationlPartially purified lipopetide type biosurfactant

was procured from Paenibacilluspolymyxa exhibiting a wide range of antimicrobial activity against both Gram positive and gram negative pathogens. Maximum rate of diesel degradation was 67.09% at the 15th day. Emulsification index of crude extract of biosurfactant was found

Sorghum grains (2.20 days) were found to be best highest against coconut oil at 51.75%. Biosurfactant - detergent combination was able to remove stains better than alone.

BiofertilizerslPseudomonas f luorescensand Bac i l lus

megateriumshowed Plant growth promoting (PGPR) traits i.e.Indole acetic acid production, ammonia production and phosphate solubilizing activity. Peat based bioformulation comprising PGPR isolates used for Capsicum annum in combination showed effective increase in growth parameters like plant height, total number of fruits per plant, fruit weight, fruit length, fruit width and number of seeds per plant.

lBiocontrol agents isolated from rhizosphere of Chick pea (Cicerarientum) were among fungi ( 5 2 % ) A s p e r g i l l u s s p p . ( 1 5 . 4 % ) , A l ternar iaa l ternata , Cur vu lar ia lunata , Mucorcircenelloids, Fusariumoxysporium and C h a e t o m i u m g l o b o s u m ( 3 . 8 4 6 % ) a n d Trichodermaharzianium and Rhizopusstolonifer (7.69%). Among bacteria the isolates were identified as Pseudomonas spp. Bacillus spp., and Paenibacillus spp. (0.983%), Halobacterium spp. and Planococcus spp. (0.015%), Micrococcus spp. (0.188%), Enterobacter spp. (0.462%) and Staphylococcus aureus (1.53%). Antagonistic activity of fungi Trichodermaharzianum and bacteria Pseudomonas spp. and Bacillus spp. were demonstrated against soil-borne pathogens Fusariumoxysporium, Alternaria spp., Curvularia spp., Aspergillus spp. which cause diseases in chickpea.

lP l a n t g r o w t h p r o m o t i n g Pseudomonasaeruginosa and Bacillus coagulans producing exopolysaccharide were found to promote the growth of Okara in field trials showing maximum seed germination (79.15%), maximum plant height (22.93 cm), maximum number of leaves per plant (10.66%), maximum

2leaf area graphically (181.89cm ),maximum root length (16.56cm), maximum fresh weight and dry weight of root per plant (7.83g, 1.65g respectively).

lVermiwash extracted from vermicompost composted from rice bran through Earthworms (Eisiniafoetida) significantly increased growth of Okra plant in terms of plant height, leaf surface area, number of leaves per plant, leaf surface area, and number of leaves per plant.
