A wide array of Alzheimer’s related research solutions
ß-Amyloid (Aß) is a peptide of 39 to 42 amino acids in length formed after
sequential cleavage of the transmembrane Amyloid Precursor Protein (APP) by ß and g-secretases (1-3). While only a small amount is processed by ß-Secretase, also known as BACE1 (ß-secretase APP cleaving enzyme or memapsin), APP is predominantly processed by a-Secretase, producing a 83-amino acid C-terminal fragment, C83. Subsequent cleavage of C83 by g-secretase produces a non-toxic N-terminal 3 kDa protein (1-2). Aßs are amphiphilic peptides with a hydrophilic N-terminal domain (residues 1 to 28) and a hydrophobic C-terminal domain (residues 29 to 40-42), the latter corresponding to a part of the transmembrane domain of APP (4). Aß assembly into fibrils is initiated by a conformational transition from random coil to ß-sheet (hence the name ß-Amyloid) and a nucleation-dependent aggregation process (4). Aß peptides that
are 39 to 42 residues in length with a molecular mass of approximately 4 kDa are the core components of neuritic plaques seen in Alzheimer’s disease (AD) brains. The presence of excess amount of Aß deposits and neurofibrillary tangles, NFTs (5-6), containing hyperphosphorylated Tau proteins, are the hallmarks of an AD brain (1).Harnessing over a decade’s worth of experience and a multi-level technology platform that includes peptides, antibodies, dyes, and assay kits, Eurogentec and AnaSpec joined forces to become the world’s most trusted source of integrated proteomics solutions specifically designed for AD research.

TACE (TNF-a converting enzyme), ADAM17 or a-secretase belongs to the ADAM (A Disintegrin and Metalloprotease) family of proteins, which are involved in myogenesis, neurogenesis, fertilization through the ectodomain shedding of cell surface proteins (19). TACE, the first ‘sheddase’ to be identified and the predominant protease responsible for the generation of soluble mature TNF-a (20), plays a crucial role in acute and chronic inflammation. The inhibition of TACE by a pharmacological agent may modulate the effect of TNF-a, a crucial mediator in the inflammatory process (21). TACE is also responsible for the proteolytic cleavage of APP, L-selectin and TGF-a (19, 22, 23).
The SensoLyte® 520 TACE Activity Assay Kit is a homogeneous assay that can be used to detect the activity of TACE and screen for TACE inhibitors (figures 2 and 3).
The assay is based on a QXL® 520/5-FAM FRET substrate, derived from a sequence surrounding the cleavage site of TACE (24). In the intact FRET peptide, the fluorescence of 5-FAM is quenched by QXL® 520. Active TACE cleaves the FRET substrate into two separate fragments resulting in an increase of 5-FAM fluorescence which can be monitored at Ex/Em= 490 nm/520 nm. The long wavelength fluorescence of 5-FAM is less interfered by the autofluorescence of cellular components and test compounds.

MOG protein (Myelin Oligodendrocytes Glycoprotein) is exclusively found in the Central Nervous System [CNS]. MOG-derived peptides have been used to induce experimental autoimmune encephalomyelitis (EAE) as mouse or rat models of human multiple sclerosis
(MS). MS is a chronic inflammatory demyelinating disease of the CNS. Among all MOG peptides available from AnaSpec, the 35-55 fragment is by far the most widely used, as it has been shown to induce 100 % EAE in mice. MBP (Myelin Basic Protein) induces immune reactivity in multiple sclerosis. Proteolipid protein (PLP) is the most abundant protein in CNS myelin sheath and is highly conserved among species. PLP has a 50 % hydrophobic amino acids content and a molecular mass of 30 kDa. Mouse PLP (139-151) and PLP (178-191) are able to induce autoantibody production and relapsing-remitting neurological disease causing extensive plaque-like demyelination.

Parkinson's disease is a degenerative disorder of the central nervous system. It results from the death of dopamine-containing cells in the substantia nigra, a region of the midbrain.

Scientists have made significant progress in understanding the possible causes of Alzheimer’s disease but many questions remain. One area to improve on is the early diagnosis of the disease. Thioflavin T, Congo Red and derivatives are good tools to investigate AD.
-The benzothiazole dye, Thioflavin T (ThT), is a classic amyloid stain for senile plaques containing ßA4 peptide in AD brain. ThT is a useful probe for the aggregated fibrillar state of ß-amyloid (1-40) fibrils.BTA-1 and BTA-2 are uncharged derivatives of ThT showing very good brain entry and clearance. These derivatives
label amyloid deposits up to 50-fold and 6-fold higher affinity than ThT, respectively.
-Congo Red (CR) is a dye used in biochemistry and histology. When bound to amyloid fibrils, a red-shift of CR absorption spectrum is observed and fluorescent activity is detected. This property makes CR a sensitive diagnostic tool to test the presence of amyloid in tissue sections. Chrysamine G (CG) and half chrysamine G (hCG) are carboxylic analogs of CR. Both can cross the blood-brain barrier and can be used to stain cerebrovascular amyloid in tissue sections.
