INTRODUCTION
The s a g u a ro o r g i a n t c a c tu s ( C a m e g ie a g i g a n t e a
B r i t t an d R o se ) i s th e m o s t o b v io u s an d b e s t known o f th e
S o n o ran D e s e r t p l a n t s i n th e Low er S o n o ran L i f e Z one. I t
h a s come to b e th e sym bo l o f th e w e s t and d e s e r t l i f e in
g e n e r a l , and o f A r iz o n a i n p a r t i c u l a r . I t s f lo w e r i s th e
s t a t e f lo w e r o f A r iz o n a .
P o s s i b l y th e f i r s t w r i t t e n a c c o u n t o f a s o f t
r o t t i n g d i s e a s e o f th e s a g u a ro was t h a t o f H ubbard i n 1899
( 1 1 ) ; H ubbard was p r o b a b ly th e f i r s t to i n o c u l a t e an
a p p a r e n t l y h e a l t h y p l a n t w i th t i s s u e from a r o t t i n g p l a n t .
On r e v i s i t i n g th e s i t e l a t e r , h e r e p o r t e d , " I fo u n d th e

L i g h t l e , S t a n d r in g , and Brown (1 6 ) w ere th e f i r s t

t a l i t y due to b a c t e r i a l n e c r o s i s c o n t in u e s a t i t s p r e s e n t
r a t e an d th e p ro b le m s r e l a t i n g to s e e d l i n g e s t a b l i s h m e n t
p e r s i s t , i t was p r e d i c t e d b y A lc o rn an d May ( 3 ) t h a t th e
s t a n d w i l l b e w ip ed o u t b y 2000 A. D. S in c e i n th e U . S .
s a g u a ro s n a t u r a l l y e x i s t i n A r iz o n a a n d , o n ly m a r g i n a l l y ,
i n C a l i f o r n i a , t h e r e l a t i o n s h i p o f b a c t e r i a l n e c r o s i s to

The o rg a n is m r e s p o n s i b l e f o r th e n e c r o s i s o f th e
s a g u a ro c a c t u s , E . c a m e g i a n a , was o r i g i n a l l y d e s c r ib e d
(1 6 ) a s b e in g a Gram p o s i t i v e , p e r i t r i c h o u s , e n c a p s u l a t e d ,
n o n sp o ro g e n u s r o d . I t was n o te d a s b e in g s i m i l a r to E.
c a r o to v o r a i n th e ty p e o f d i s e a s e p ro d u c e d , an d in
r e a c t i o n s to m o s t m ed ia t e s t s . H ow ever, when E . c a r o to v o r a
was i n o c u l a t e d i n t o th e s a g u a ro no d i s e a s e r e s u l t e d , and
when E . c a m e g i a n a was i n o c u l a t e d i n t o c a r r o t s and o t h e r
h o s t s o f E . c a r o t o v o r a no s o f t r o t t i n g o c c u r r e d .
A p p a r e n t ly , b e c a u s e o f th e d i f f e r e n c e i n Gram s t a i n
r e a c t i o n and p a t h o g e n i c i t y , t h e o rg a n is m was c o n s id e r e d a
new s p e c i e s .
The Gram r e a c t i o n was c o n f irm e d b y B o y le ( 4 ) who
n o te d , h o w e v e r , t h a t o ld l a b o r a t o r y s t o c k c u l t u r e s becam e
Gram n e g a t i v e . On th e o t h e r h a n d , B u rk h o ld e r fo u n d t h i s
o rg a n is m to be Gram n e g a t i v e , w i th o ld c u l t u r e s show ing
Gram p o s i t i v e g r a n u le s ( 5 ) . B o y le fo u n d E. c a m e g ia n a to
b e lo n g to th e s o f t r o t g ro u p on th e b a s i s o f p h y s i o l o g i c a l
and c u l t u r a l s i m i l a r i t i e s to E . c a r o t o v o r a . H ow ever, i t
was a l s o fo u n d to h a v e a th e r m a l d e a th p o i n t o f 59 C, 7 C

s u r f a c e and i n t e s t i n e o f th e C a c to b r o s i s f e m a l d i a l i s

l a r v a e r e s p o n s i b l e f o r th e t u n n e l s . On r e a r i n g th e s e
l a r v a e i n c a g e s , th e b a c te r iu m was i s o l a t e d from th e a d u l t
and th e s u r f a c e o f th e e g g s . When l a r v a l d e s c e n d a n ts o f
th o s e c o l l e c t e d fro m th e t u n n e l s w ere i n o c u l a t e d i n t o
c a c t i , t y p i c a l s o f t r o t c h a r a c t e r i s t i c o f th e d i s e a s e
r e s u l t e d .
B o y le b e l i e v e d t h a t C a c to b r o s i s l a r v a e h a d one l i f e
c y c le a y e a r w i th th e l a r g e s t p a r t s p e n t t u n n e l i n g i n th e
c a c t u s . She o b s e rv e d l a r v a e l e a v in g p l a n t s to p u p a te u n d e r
d e b r i s on th e g ro u n d from May th ro u g h A u g u s t. I n
l a b o r a t o r y s t u d i e s , p u p a t io n r e q u i r e d 28 -33 d a y s ; th e a d u l t
n o c t u r n a l m oth l a i d i t s eg g s an d d ie d w i t h in a t h r e e day

B u t l e r an d W ern er ( 6 ) c o l l e c t e d C a c to b r o s i s a d u l t s

From o u r own o b s e r v a t i o n s l a r v a e ch an g e c o l o r a s
th e y d e v e lo p : a t 2 mm th e y a r e p in k , from 3 to 15 mm
bro w n , an d from 18 to 24 mm b l u e . S m a ll p in k l a r v a e can
e n t e r s m a l l g re e n h o u s e r e a r e d p l a n t s a n y w h e re , h o w ev er th e y
can o n ly e n t e r l a r g e c a c t i n e a r th e a p e x ( t h e g ro w in g
p o i n t ) due t o h e a v y c u t i n i z a t i o n e ls e w h e re ( 4 ) . L a rv a e o f
a l l s i z e s , h o w e v e r , h a v e b e e n fo u n d i n s i d e th e f lo w e r s t h a t

n e c e s s a r i l y mean an o rg a n is m i s a p l a n t p a th o g e n ( 1 9 ) ;
h o w e v e r , s o f t r o t t i n g E rw in ia p ro d u c e p e c t i c enzym es m ore
r a p i d l y an d i n g r e a t e r v a r i e t y th a n n o n -p a th o g e n s (1 4 , 1 9 ) .
F o r t h i s r e a s o n a s e l e c t i v e b r o t h was d e v e lo p e d b y
m o d ify in g N asuno and S t a r r ' s medium f o r p e c t i c enzyme
p r o d u c t io n b y E. c a r o to v o r a (2 2 ) by th e a d d i t i o n o f
e ry th r o m y c in , a m a c r o l id e a n t i b i o t i c a c t i v e a g a i n s t Gram
p o s i t i v e b a c t e r i a i n v e ry low c o n c e n t r a t i o n s ( 1 2 ) . S in c e
e ry th ro m y c in i s a b a s i c m o le c u le and i t s a c t i v i t y i n c r e a s e s
f i v e f o l d f o r an i n c r e a s e o f one u n i t o f pH i n th e ra n g e
from 6 .5 to 7 .5 ( 1 2 ) , t h e medium was a d j u s t e d to pH 8 .0 .
