    It was nearly 20 years ago, in 1985, that the vision of Douglas Fordwas realized when he founded Optimize Technologies. Based on thesimple idea of offering the highest quality, innovative products forHPLC, the company has grown from simple beginnings to the presentlyworldwide recognized brand.     OPTIMIZE INNOVATION: THE DRIVING FORCEIt is innovation that started Optimize, and it is innovation that continuesto fuel its progress. The spirit of Optimize has always been to carve anew swath in the forest of HPLC products. This attitude is exemplifiedby products such as the auto-adjusting stem, OPTI-MAX check valvesystem, and OPTI-LYNX quick-connect hardware. Each of these resultedfrom not letting good enough rest, but by pushing the limits to excellence. It is shown in everything we do at Optimize.

    OPTIMIZE R&D: THE HEART OF PROGRESSResearch and Development is somewhat of an obsession at OptimizeTechnologies; when as a company you define and pride yourself onprogress, it is hard not to be. Our team of engineers and chemists isconstantly engaging in the evaluation of existing product designs andconceptualization of new ones. We realize that people look toOptimize for products that push the boundaries of conventional product design, and therefore, contribute a large part of our time andresources to the R&D process. Our many collaborative efforts with various OEMs and pharmaceutical research companies have resultedin customized components and devices for each of their unique applications, increasing our experience beyond the realm of HPLC.

    OPTIMIZE MANUFACTURING: MACHINING PRECISIONMost of our products are machine-manufactured by skilled machinistsat our facility in Oregon City, Oregon. To meet our exacting standardsfor performance and reliability, many of our component designs pushthe limits with respect to manufacturing tolerance. To ensure that thesetight dimensional tolerance controls can be maintained for every partwe produce, we individually machine each component, and avoidthe use of mold-based manufacturing techniques. And we ensure theconsistency of our manufacturing process through constant ongoingevaluation as part of a rigorous quality control program.

    WHAT IS OPTI-LYNXTM?

    When OPTI-LYNX was first introduced, it was a revolution in the HPLC industry. No longer was there the need forgetting out wrenches for stainless steel fittings, or having to crank down those finger tights to prevent leaks. OPTI-LYNX was the first to fulfill the promise of a low dispersion, quick-connect system. Since that time, OPTI-LYNXhas found its way into a number of different laboratory applications. From the large scale high-throughput labs withthe need for replacing components with the utmost efficiency, to the single instrument lab that enjoys the ease of useOPTI-LYNX offers - many have realized the advantages of the OPTI-LYNX quick-connect system.

    CONNECTIONSMaking a high-pressure connection, one that is rated to 6000 psi, cannot get much easier than a quarter-twist - and asimple quarter-twist is all that is needed to instantly make and break an OPTI-LYNX connection. An OPTI-LYNX connectionconsists of two components: a slotted side and a pinned side. Each half comes together around a specifically designedinsert that flawlessly connects via a low volume/low dispersion interface. This graceful design results in a minimal impacton your chromatographic system allowing OPTI-LYNX to be used anywhere - even the volume critical regions.

    The very nature of OPTI-LYNX is conducive to a true zero-dead-volume connection. The inserts are designed specifically for the OPTI-LYNX hardware, making the connection identical every time with no dead spaces in the flowpath. And because the connecting end caps need only be plumbed one time, there is no handling of tubing -no need to ensure that the tubing is butted against the tube stop each time a connection is made.

    VERSATILITYThe OPTI-LYNX modular design also provides the maximum in versatility for your system. OPTI-LYNX inserts can bevirtually anything - a filter, guard column, trap cartridge, analytical column - and since the interacting surfaces areidentical, the only thing you need to change is the holder tube. Install a guard column for one application and thenswitch to a pre-column filter for the next. Use an in-line filter with different porosities and diameters, just by changingthe insert. Switch out columns in an instant with the Velocity system, or retrofit current columns with the universal portadapter - we even offer columns with integrated OPTI-LYNX end fittings. You can even make and break flowpathsfor multiple detectors, or anywhere else a break in flow is frequently made. OPTI-LYNX allows all of this - without ever having to change out the basic components.

  • Set up a simple column-switching rigwithout the need for expensive high-pressure valves. Install OPTI-LYNXuniversal port adapters on yourcolumns and switch between differentcolumns quickly and easily. Whencolumns are not in use, seal themwith the OPTI-LYNX quick plug.

    One method requires a filter, whileanother calls for a guard column - noproblem. Simply choose the insertyou want (guard, filter, or straightthrough connection), couple it withthe correct holder tube, and you’reoff and running. There is no need todisconnect any tubing, and the onlytools you need are your fingers.

    If you use more than one detector, orif you need to re-route flowpaths withthe utmost efficiency, the OPTI-LYNXDirect-Connect In-Line Kit is youranswer. Simply terminate the columnoutlet line with a holder cap/tube,and get extra bayonet connectorsfor each inlet line. With this set-up,switching flowpaths becomes afacile task, not a huge production.

    FILTRATION

    Filtration is a necessary component in any HPLC system. Keeping particulates out of the flowpath prevents premature wear on vital components, wear that could lead to costly replacement or repairs. An holistic approach tofiltration, addressing potential particulates throughout the solvent flowpath, leads to a consistently and efficiently operating chromatograph. Ideally, there are three critical areas where filtration is necessary.

    SOLVENT RESERVOIRWhile the availability of HPLC-grade solvents diminishes the need for pre-filtering solvents, it is still imperative that afiltration device be installed in the reservoir filter. Placing a filter in the reservoir is an economical way to filter particles that may result from buffer salt precipitation, improperly cleaned glassware, or microbial contamination; allof which are not remedied by the use of HPLC-grade solvents.

    IN-LINE FILTRATIONThe great frictional forces generated at the piston/piston seal interface inevitably lead to particulate shedding.Placement of a filtering element between the pump and injector/autosampler removes these particulates, preventingthem from affecting downstream components, circumventing costly downtime and/or repairs.

    PRE-COLUMN FILTRATIONIncompatibilities between sample matrix and mobile phase, septa coring from the sample needle, and shedding from moving parts in the injector: these all contribute to particulates in the region post-injector. Use of a low-impact, pre-column filter removes these particulates from the flowpath, saving your analytical column and detector from deleterious effects.

    OPTI-SOLVTM RESERVOIR FILTER

    If you want to keep particulates out of your system, and we are sure you do, you need to start at the most basic level -the reservoir bottle. OPTI-SOLV Reservoir Filters employ a unique conical design inside the filter housing preventing air bubbles from getting trapped and disturbing your analysis. A solid Teflon body keeps OPTI-SOLV in the bottom of the bottle where a bottom-mounted titanium frit provides unrestricted solvent access. As always, Optimize makes installationas simple as possible - no tools or fittings required.

    We also offer Reservoir Filters with tube stems, made with Hastelloy C® for maximum corrosion resistance and inertness.

    OPTI-SOLVTM IN-LINE FILTER

    OPTI-SOLV In-Line Filters are the perfect choice for removal of particulate matter from the flowpath. Installed between the pumpand injector/autosampler, they prevent damage to downstream components - damage that can cost you in both instrumentdown time and troubleshooting labor.

    OPTI-SOLV’s unique swivel design allows frits to be changed without removing the connecting tubing. Just plumb it once andleave it. Hand-tighten the PEEK holder to make a high-pressure seal, or for those who prefer wrenches, we also have awrentch-tight stainless steel version. The user-friendly design is well suited for a number of applications, such as high temperature environments, where quick, easy changes are imperative.

    Tired of needing to be a finger contortionist to change out your in-line filter? With HPLC systems getting smaller and more compact, the areas to work with are also decreasing in size. This often makes getting at those components that need frequent changing quite a task - especially if you need wrenches!

    The OPTI-LYNX In-Line Filter System features Optimize Technologies' proprietary OPTI-LYNX hardware, allowing filter changesto be done in a quick quarter-turn. Twist the OPTI-LYNX open, slap in a new filter cartridge, and use the time saved to grab acup of joe. OPTI-LYNX In-Line Filters also offer unmatched versatility with a choice of eight different filter insert porosity/diametercombinations for every possible filtering need.

    MINI & µ-FILTER

    The OPTI-SOLV Mini Filter provides low-impact filtering in a package no larger than a finger tight fitting. Use them to prolong the life of your analytical column, or before your mass spectrometer as a last line of defense against debris.

    Perfect zero-dead-volume connections are obtained every time with our patented automatic tube stop depth adjustment. Unique depth-filtering elements allow for greater capacity with no band spreading or loss in performance.The efficient two-piece design needs no holder and threads into any 10-32 port.

    For your volume critical filtering needs, we now offer the OPTI-SOLV µ-Filter. Based on the same design that has madethe Mini Filter a huge success, the µ-Filter cuts the internal volume to less than 200 nL, but retains the Mini Filter’s easeof use and functionality.

    Often a sample bound for HPLC or LC/MS analysis requires some type of clean-up to ensure optimal results. Detergentsor salts can affect the ionization process, while overly dilute samples can be swamped out by background noise, making them undetectable. There are many methods to treat samples, but most require lengthy off-line procedures thatinvolve significant sample handling. These procedures not only consume valuable time, but also can result in appreciablesample loss.

    Optimize Technologies trap cartridges are an elegant and efficient means for sample processing. Trap cartridges aresmall packed beds that use the adsorptive properties of analytes to effectively concentrate dilute samples, desalt, removedetergents, and more. Unlike off-line procedures that require significant sample handling and potential sample loss,Optimize trap cartridges have the advantage of being able to be used in a completely on-line environment. On-line cleanup not only increases recovery, but is also amenable to automation for unattended use and higher throughput.

    Traps work on the principle of "digital chromatography." This is the most apt description because there are typically onlytwo states of the analyte: bound or unbound. This on/off behavior is exploited by either trapping the analyte and lettingsample matrix and contaminants go to waste; or by trapping the undesirable components and allowing analytes to passunimpeded. The discontinuous interaction of the analyte in a trap cartridge prevents dilution and dispersion that mayoccur in larger beds with a more distributive partitioning effect. For some ideas on the use of trap cartridges, please seethe following pages.

    Though Optimize trap cartridges can be used for a number of different applications, two of the most commonare sample concentration/desalting and detergent removal. The general procedures listed below outline apossible approach to these applications and illustrate the basic concepts of the trapping technique.

    SAMPLE CONCENTRATION/DESALTINGSome samples contain an unacceptable concentration of salts and/or are in a volume that is too great forthe analytical technique being used. In these instances, a trap cartridge may be used to trap the analyte,effectively removing salts and eluting the analyte in a smaller, more concentrated volume of eluent.

    To successfully concentrate and/or remove salts, the optimal strategy is that of trapping the analyte withinthe trap cartridge and allowing undesirable components to go to waste. This can be done most efficientlyby plumbing a switching valve as shown in Figure 1a. The chemistry of the trap cartridge should be chosensuch that there is good selectivity for the analyte of interest, with little to no affinity for the salts or other con-taminants. Loading should be done with weak solvents to prevent the analyte from dispersing in the bed.Prudent selection of bed volumes can also prevent unwanted sample dispersion. Optimize traps come in avariety of different volumes and dimensions to facilitate the best trapping environment with minimal broadening or dispersion effects.

    Once the sample has been sufficiently processed, the analyte is eluted from the trap. This can be done byrunning a gradient through the trap and onto an LC column, or it can be performed with a plug of a strongsolvent eluting the sample in one tight band. While the latter of these two options is best, it is not alwayspossible. Using the plumbing diagram from above, the elution valve position is displayed by Figure 1b.

    DETERGENT REMOVALThe second strategy for trapping is exemplified by the application of detergent removal. Detergents may bepresent as carryover from SDS PAGE analysis, or have been added to help solubilize the sample. Whateverthe reason, detergents greatly hinder MS analysis and, therefore, must be removed from the sample matrix.One of the most common detergents is the ionic detergent sodium dodecyl sulfate (SDS). Fortunately, removalof SDS is a fairly simple task when using Optimize trap cartridges.

    Ionic detergents like SDS can usually be removed using an ion exchange trap. SDS is an anionic detergent,so a strong anion exchanger (SAX) is the packing material of choice. The approach for detergent removal isexactly the opposite of desalting/concentrating outlined above: the undesired component, i.e., the detergent,is retained in the trap cartridge while the analyte is flushed directly through to the LC column or MS. For thespecific removal of SDS from a protein mixture, it is best to keep the mobile phase pH at 4.4 or lower to keepthe anionic amino acid sidechains protonated and less likely to interact with the SAX functionalities.

    One possible configuration for plumbing this set up is shown in Figures 2a and 2b. The first valve positionputs the gradient pump directly in-line with the trap, removing detergent from the sample matrix and sendingdetergent-free matrix and analyte on for further analysis. The second position takes the trap out of the mainflowpath so that it may be regenerated for subsequent runs. With a judicious choice of loading and regenerating conditions, Optimize trap cartridges can be reused numerous times.

    Most trapping applications can be performed with the myriad of silica based packings we carry. For otherapplications requiring extreme pH, there is also the choice of polymeric packing materials. We also offercustom packing to ensure that you will always have a solution available for your trapping and chromatographic needs.

Noproblem. Just unscrew and install the appropriate slotted connectortube - no need to re-plumb any solvent lines. Once you are ready to go, a simple quarter-twist engages the OPTI-LYNX connection. But in additionto its ease of use, OPTI-LYNX also provides a means for easily integrating pre-column protection to your Velocity columns.

    The Velocity Coupler allows pre-column protection devices to be coupled directly toyour Velocity analytical column without the need for additional connecting tubing or fittings.     Check valves are a crucial, yet often overlooked component of the HPLC system. While a properly functioning checkvalve is virtually unnoticeable, an improperly functioning check valve can lead to a noisy baseline, pressure fluctuations, and inconsistent flow. Because a check valve can have such a large impact on the performance of yourpump, it is imperative when choosing your replacement check valves you choose the very best. Of course, webelieve that the OPTI-MAX check valve system is the very best - we wouldn't sell it if we didn't. But don't take ourword for it; read on for the innovations Optimize Technologies has done for the check valve. And as with anyOptimize part, if you’re not satisfied, return it for a refund.

    BEST SEAT IN THE HOUSEA check valve can only function effectively if it is seating properly, reacting to changes in the piston direction. Theseating process is fairly simple - the ball lands on the seat and creates a seal - but there are many factors that contribute to optimal seating. At Optimize Technologies, we have engineered our check valves to be as responsiveas possible, scrutinizing every possible detail.

    Each ball and seat set is individually matched to ensure a lock and key fit. This guarantees 3 bubbles/minute orless in the industry standard nitrogen bubble test; common acceptable standards are 12 bubbles or less. We havealso cut down on the ball travel distance within the check valve: less distance to travel means quicker seating andresponse. OPTI-MAX check valves are also available with a ceramic ball and seat. Ceramic is the recommendedmaterial because it is denser than ruby and can result in better performance in certain solvents.

    Use PEEK cartridges for biocompatible HPLC systemswith non-metallic flowpaths, for high temperatureapplications, and with high percentages of THF.

We were the firstto introduce the concept of the free-turn housing: a housing where the thread that tightensthe housing into a pump head spins independently from the inner housing body. Thisenables you to change out a check valve without having to disconnect any tubing - a greatidea, born in efficiency.     OPTI-SEAL® OR ITBOptimize Piston Seals are the longest lasting seals you can buy. Machined from either of two proprietary polymerblends, UHMW-PE or ITB, Optimize Seals provide the ultimate in high-pressure sealing capacity. For mostly aqueous/aqueous-buffered mobile phases, the OPTI-SEAL UHMW-polyethylene is the wisest choice. Speciallydesigned to be more resistant to abrasion and wear than standard polyethylene-based seals, they can last up toseven times longer under mostly aqueous conditions. Despite the UHMW-PE’s ruggedness, it has a coefficient of friction approaching that of Teflon. The homogenous polymer nature leads to less shedding, but when shedding doesoccur, a larger, stringier particle is produced that is easily caught by a filter frit.

    PTFE-based Improved Teflon Blend (ITB) seals are a superior choice for applications with elevated temperatures, highorganic content, or when the broadest possible solvent compatibility is required. ITB seals tend to be softer than thosemade from UHMW-PE making them more forgiving at high pressures where piston side-loading or misalignment mayoccur. This softer nature also leads to more shedding, but the ITB proprietary blend is designed to shed particles thatare large enough to be caught at the surface of a standard 2 µm frit, not embed within it.

    PISTONSOptimize sapphire pistons offer optimal concentricity and resistance to wear. Each sapphire rod is specifically cut inrelation to the grain of the crystal lattice, giving maximal resistance to breakage and superior wear characteristics.Attachment of the rod to the piston ferrule, an often overlooked procedure, is performed through a proprietary processensuring concentricity, an important parameter that has a direct effect on seal lifetime.

    Seals and pistons are, by design, a consumable part of the HPLC pump and should be replaced on a regular preventative maintenance schedule. To maintain peak performance and unplanned downtime, piston seals shouldbe replaced every three to six months, while sapphire pistons require inspection every six months with yearly or as-needed replacement.

    SEAL FORM TOOL KIT

    Installing a piston seal can be a tricky undertaking. You have to fit a relatively small seal intothe seal cavity, and do it in such a way to ensure that it is pressed in evenly without distortion or damage. The Optimize Seal Forming/Insertion Tool Kit was specificallydesigned to aid in this endeavor.

    This kit contains three tools to accommodate all of your HPLC piston seals. One end has abeveled cavity for seal forming, while the other end is an insertion support for perfect sealinstallation. Also included with each kit are complete instructions for maximizing seal life,and a solvent compatibility guide to aid in prudent seal material selection.

    In-line filtration is a necessity for protecting downstream components from damage andblockages due to particulates. But what use is a filter if it is never replaced? The placementof the Agilent/HP PTFE filter is buried deep within the pump head making what is usually aquick job, into a laborious procedure. Optimize continues its tradition of innovative solutionswith the Purge Valve Upgrade Kit.

    The Purge Valve Upgrade Kit resolves this difficulty by permanently replacing the PTFE filterwith a low volume PEEK insert, and then relocating the filter frit to an easily accessibleholder on the side of the purge valve. What once was a cumbersome production of changing the filter becomes a quick and easy task.
