gke Steri-Record® Mini-Bio-Plus
self-contained biological indicators (SCBI)
and Bio-Compact Process Challenge Devices (Bio-C-PCDs®)
Application
The gke Steri-Record® Mini-Bio-Plus self-contained
biological indicators (SCBI) are used for validation
and routine monitoring of steam-, ethylene oxide-,
formaldehyde- and hydrogen peroxide/plasma-
sterilization processes. After sterilization the SCBIs
can be incubated in the CSSD by the user without
using an external microbiological laboratory.
The Mini-Bio-Plus SCBIs can be used for routine
monitoring inside a package, container, or inside a
gke Steri-Record® Bio-Compact Process Challenge
Device (Bio-C-PCD®) without losing sensitivity.
They have been designed with a minimal internal vol-
ume. The internal volume of conventional SCBIs is
too large and would dramatically decrease the sensi-
tivity of class 2 indicators and cannot be used in hol-
low load PCDs to check internal lumens of MIS in-
struments.
gke offers for the first time a special test system
worldwide consisting of a hollow process challenge
device (Bio-C-PCD®) with self-contained biological
indicators inside. The combination of the gke Steri-
Record® Bio-C-PCD® with the Mini-Bio-Plus SCBI
is the first class 2 indicator which simulates minimal
invasive surgical (MIS) hollow instruments. In addi-
tion Bio-C-PCD®s with different air removal charac-
teristics are now available. The sensitivity of these
Bio-C-PCD®s can be utilized to meet the requirement
of the load.
Bio-C-PCD®s are available with five different air re-
moval and steam penetration characteristics.
The validation of the Bio-C-PCD® according to the
load can be achieved by using the test method de-
scribed in DIN 58921.
gke Bio-C-PCD®s can be used in all four steriliza-
tion processes mentioned above, if the correct SCBIs
are inserted.
There are various hydrogen peroxide/plasma steriliza-
tion processes in the market with different penetration
characteristics of the sterilization agent. Depending on
the process used and on the load configuration an
appropriate Bio-C-PCD® should be selected.
Product description
The gke Steri-Record® Mini-Bio-Plus SCBI is de-
signed using a plastic vial with a small internal vol-
ume containing a biological indicator spore plate and
a glass ampoule with a growth medium and pH-
indicator inside. For steam-, ethylene oxide- and for-
maldehyde-sterilization processes, filter paper is used
as a carrier and as a closing filter below the cap. For
hydrogen peroxide/plasma-sterilization processes
plastic material is used. For a better recognition of the
different SCBI versions they each have different col-
oured caps (see table).
The labels of all SCBIs for steam sterilization proc-
esses contain a class 1 chemical indicator according to
EN ISO 11140-1 to check, if the SCBI has passed a
sterilization process. The self-adhesive label may be
detached from the SCBI and adhered on a documenta-
tion sheet.
An additional chemical indicator class 5 according
EN ISO 11140-1 allows that the result of steam ster-
ilization processes can be instantly read at the end of
the sterilization process. Therefore, it is not necessary
to wait for the result of the SCBI incubation since
class 5 indicators are equivalent to biological indica-
tors according to the above standard.

The specially designed and patented gke Steri-
Record® Bio-C-PCD® construction consists of a large
internal volume with a stainless steel tubing system
inside and a minimal capsule volume at the closed end.
It can be used only with the specially designed Mini-
Bio-Plus SCBIs as described before, to create a high
sensitive Hollow Load PCD which cannot be com-
bined with conventional SCBIs.
For incubation of the SCBIs gke Steri-Record® incu-
bators with two different temperatures and program-
ming versions are available (see separate data sheet).
Performance characteristics
Self-contained biological indicators:
All gke biological indicators comply with the stan-
dard EN ISO 11138 series. For hydrogen peroxide
sterilization processes the standard prEN ISO 11138-6
draft is currently under development and not yet avail-
able.
gke Mini-Bio-Plus SCBIs for hydrogen peroxide
sterilization processes are supplied with a D-Value that
is tested under the defined sterilization conditions
described in the certificate. The SCBIs for Low Tem-
perature Steam Formaldehyde (LTSF) sterilization
processes** contain in the growth medium substances,
decomposing remaining absorbed formaldehyde, so
that the pretreatment with Na2SO3 according to
EN ISO 11138-5 is not required.
The incubation time of all Mini-Bio-Plus SCBIs has
been optimized, so that steam-, formaldehyde- and
H2O2-Mini-Bio-Plus SCBIs can be fully interpreted
within 24 hours and EO-Mini-Bio-Plus SCBIs within
48 hours incubation time. The SCBIs do not contain
additional enzymes and do not require fluorescent light
for evaluation. If the incubation time exceeds the time
mentioned, the colour of the media does not change
back, this could happen with some conventional
SCBIs. If the sterilization process is unable to kill the
spores, in most cases the colour change will already
occur after 8 hours.
For steam sterilization processes an additional class 5
indicator according to EN ISO 11140-1 is inserted.
The indicator enables the user to make an interpreta-
tion of the result immediately at the end of the process
and the batch can be released or retained before the
incubation time has finished. Therefore it is not neces-
sary to wait 3 hours or longer for incubation to take
place so the batch can be released. The result of a class
5 indicator provides a much higher probability of ste-
rility instead of using the result of a SCBI incubation
after 3 hours where only a probability of > 97 % is
achieved.
The Mini-Bio-Plus SCBIs meet the performance char-
acteristics published in the current United States Phar-
macopeia (USP) and European Pharmacopeia (EP).
Operational description
The SCBIs must be placed in the most difficult pene-
tration condition of packs or containers. If hollow de-
vices are to be sterilized SCBIs cannot be placed in-
side MIS instruments. In this case Bio-C-PCD®s with
SCBIs inside are used to simulate MIS instruments.
This alternative offers for the first time the possibility
to check sterility inside hollow instruments. SCBIs in
PCDs can be directly incubated at the end of the cycle
without opening packs or containers.
After the sterilization process the SCBIs are taken out
of the load or the Bio-C-PCD® and are activated by
crushing the glass ampoule. After they are incubated
correctly according to the directions for use, the colour
of the liquid will indicate a pass or fail of the steriliza-
tion process. If the colour has not changed, this indi-
cates a pass of the process, a colour change indicates a
fail.
Special test devices (gke Steri-Record®
Bio-C-PCD®s) where SCBIs can be used as a detec-
tor inside:
Bio-C-PCD®s with SCBIs inside can simulate porous
loads and hollow devices simultaneously. A special
Bio-C-PCD® to simulate the hollow load device de-
scribed in EN 867-5 is available as well.
Additional Bio-C-PCD®s are available to simulate
different load configurations and may be validated
with the test method, described in DIN 58921.

The labels on the SCBI are self-adhesive and can
be removed for external documentation.
The combination of Mini-Bio-Plus SCBI and spe-
cially designed Bio-C-PCD®s allows correct testing
of hollow lumen instruments.

DIN EN ISO 11138-5.
