1. GENERAL INFORMATION
1.1 The Nanobaume is a compact stainless steel unit designed to pack capillary columns with a silica
bed at high pressure. Its unique sealing design permits rapid assembly, facilitating quick packing
changeover. Simple hand tightening the pressure vessel will seal up to 9000 psig with liquid media,
3000 psig with gas. No tools are required to operate the packing unit, although a set of adjustable
wrenches is useful for initial setup and column assembly. SP-400-M1 and SP-403K-M1 kits include
connections for 360um, 1/32" OD , 1/16" outside diameter (OD) columns, but the NANOBaume will
accept small OD column diameters using the TS-015 sleeve kit. LC-MS spray tips may also be packed.
Component materials include Kel-F, PEEK, 316SS, 304SS, Borosilicate glass, ETFE, and FFKM
perfluoroelastomer. The vial insert (SP-125) supplied with the SP-400-M1 will accept 2 ml (12x32mm)
glass vials. Other inserts are available, (please inquire)
2 PRODUCT SAFETY AND CARE
2.1 The NANOBaume is not for use under Supercritical Fluid conditions. Do not exceed the publishedpressure limits (9000psig liquid/3000 psig gas). Always operate with a safety shield in place. When
packing with liquid media, first flood the Nanobaume and empty column with solvent. If using the TS-
015 sleeve adapters or SP-015, SP-014 Ferrule/Nut for the capillary columns , then pressure limit is
reduced to 6000psig.

2.2 Hand tighten the cap lock (SP-400-01, FIG 1) on the packer body (SP-400-05 ,FIG 4) and do not
overtighten, as galling may occur between the stainless steel surfaces; the O-ring seals at high pressure.
Scratches, nicks or dents on the sealing surface of the Cap seal (SP-400-02, FIG 3), Nanobaume body

(SP-400-05, FIG 4) or O ring (SP-400-07, FIG 2) may impede sealing performance. As the Nanobaume
is primarily 316 SS; ethers, concentrated oxidizing acids are not recommended. (i.e. Hydrochloric acid
at greater than 10%) If small concentrations of aggressive solvents are used during packing, it is
highly recommended that the unit be flushed with a weak solvent after use.
3. ASSEMBLY AND CONNECTION TO PRESSURE SOURCE
PLEASE NOTE; THE FOLLOWING INSTRUCTIONS ARE WRITTEN TO INCLUDE COMPONENTS
FROM THE SP-403K-M1 STARTUP KIT. CUSTOMERS WHO PURCHASED THE SP-400-M1 BASE UNIT
MAY PURCHASE THESE PACKING ACCESSORIES SEPARATELY.
3.1 Remove the SP-400-M1 (FIG 5) and SP-403K-M1(FIG 6) components from the shipping container.

3.2 Fit the Gauge Tee (SP-058) with the pressure gauge (SP-087). Wrap the ¼” NPT threads 3-4 times
with teflon tape (TTB-50) Wrench tighten.

3.3 Place the SP-116 (FIG 9) nut and ferrule on the end of the SP-046(FIG 10) stainless steel tubing,
tighten into the side port of the Nanobaume until tube is secure. The SP-116 is reusable as the ferrule
does not swage permanently onto the tube. Connect the free end to the gauge tee (SP-087) and wrench
tighten to secure the tube. Note: if a torque wrench is purchased, SP-162 torque to 10 Inch-pounds. If
not, finger tighten the nut (Clockwise direction), then wrench tighten ¼” turn then check if leaks under pressure. If leaks occur, reduce pressure, ¼ turn more and repressurize , if leaks, repeat previous steps. Use this technique for tightening all Stainless steel nuts in this publication.)

3.4 Using the second piece of SS tube (SP-046), Connect one end to the Pressure gauge Tee (SP-087),
and other end to the Pressure vent valve (SP-050, FIG 11). Wrench tighten both until tubes are secure
using technique described in 3.3. The assembly should look like Fig 12.

3.5 Connect the uppermost port of the Vent valve (SP-050, ) to the pressure source. First use the
Long piece of SS tube (SP-055, FIG 13) connect using fitting provided on the vent valve (upper port, right
side of FIG 11) tighten to secure the tube. Take the SP-110, FIG 14 Pressure Regulator Adapter ,1/4"
male NPT end and apply teflon tape to threads wrapping 3-4 times around then thread into the port of
the pressure regulator(not provided). Take the free end of the tubing and place into the other end of
the pressure regulator adapter , and tighten until the tube is secure. If the HPLC pump is being used
for the high pressure line, connect directly to pump using a 10-32 female thread union (SP-095 not
provided)
4. COLUMN ASSEMBLY
4.1 If fused silica tube will be used, cut the fused silica to length using an appropriate cutting tool . Aprecision cutter will ensure a square end without fragmentation of the column end (SP-019, not provided). Secure the column end fitting CE-003 ,( FIG 15), on the exit end of the column using an
appropriate adapter sleeve,(TS-015,FIG 20) with Nut, ( SP-130, FIG 22) and Ferrule,( SP-133, FIG 23).
The ferrule –nut- sleeve fused silica combination should look like figure 21. Tighten until fingertight
and then 1/4” turn more to secure the column to the end fitting. Be sure to push the column body into
the end fitting to eliminate a void between the column end and the web of the union.

place a 0.5um screen (SP-088, not shown) into the Micro Union (SP-089, FIG 15). Ensure that the screen
is set in flat against the bottom web of the union. Place the 1/32 inch OD tube or fused silica ( adapted
to 1/32” OD ) thru the nut (SP-102, FIG 17) and ferrule (SP-092, FIG 16) These are provided with the
union. Insert the column body into the union and while exerting pressure on the tube into the union,
tighten the nut using two wrenches. Use similar procedure for the CE-006 kit for 1/16” od column
bodies, follow steps 4.2 .

4.3 The nut ferrule fused silica should look like fig 19. The sleeve on the fused silica will bring it up to
an outside diameter of .032”(1/32") to fit the column end fittings (CE-003) Note; that the use of the
fused silica will reduce the pressure rating of the column and packer to 6000 psig.

4.4 LC-MS tips are inserted into the appropriate ferrule, and/ or sleeve depending on OD, and do not
require end fittings as the tips are fritted already.
5. SLURRY PREPARATION
5.1 PLEASE NOTE: AS CAPILLARY PACKING IS AN EXPERIMENTAL SCIENCE, SOME
TESTING WITH VARIOUS SOLVENTS OR COMBINATIONS MAY BE REQUIRED FOR
OPTIMAL PACKING.
5.2 Weigh 100 mg of bulk packing material into the 2ml vial (VL-002, FIG 26) Add 1.5ml of appropriate
solvent, or buffer (Hexane for normal phase, Propanol or acetonitrile for reversed phase, weak buffer
for ion exchange) to the vial. Add the stir bar ( SP-085, FIG 27)

5.3 Place the vial on a magnetic stir plate. Allow the slurry to stir for 5 minutes or until the stationary
phase is well wetted. Once the bonded phase is wetted insert the vial into the packer body cavity using
the vial insert (SP-125, FIG 28) provided to hold the vial in place

6.1 With the correct adapter, most column or tubing with an outer diameter of 1/32inch or less can be attached
to the Nanobaume(FIG 29 OR 30) using the nut ,SP-130 ( FIG 22)And ferrule, SP-133 (FIG23). Larger 1/16” OD
columns can be attached FIG 31, using the SP-130 (FIG 32 )nut and the Ferrule SP-134 (FIG 32) ferrule
combination to the Nanobaume . In the SP-400-M1 kit, three adapter ferrules are provided. For 360 um OD fused
silica use the SP-014 nut with the SP-015 ferrule. For 1/32inch OD tube use the SP- 130 nut with the SP-133
ferrule. For 1/16 inch OD tube use the SP-130 nut with the SP-134 ferrule. These connections are a convenient
way to connect column bodies to the Nanobaume. If the fused silica tube that has a different OD than 360, use an
adapter sleeve from the TS-015 kit and the SP-130/SP-133 combination. The adapters in the TS-015 kit will adapt
fused silica columns of OD of 125 to 495 micron to a new OD of 1/32” to fit the SP-130/SP-133 ,( FIG 33)
NOTE: USE OF FUSED SILICA WILL REDUCE THE OPERATING PRESSURE LIMIT TO 6000 PSIG
6.2 Unscrew the Cap Lock (SP-400-01,FIG 1) counterclockwise and remove the Cap Seal (SP-400-02, FIG
3) if not accomplished already.

6.3. Using the appropriate ferrule sized to the column OD, insert the column body open end through
the ferrule and the nut (refer to section 6.1) Tighten the nut only finger tight into the threaded port of
the Cap seal (SP-400-02 FIG 34) column will be adjusted in the next step.
6.4 With the column in place, insert the Cap Seal onto the Nanobaume body ,( FIG 33). Insure that the
“O” ring is in place (figure FIG 2) Thread on the Cap Lock (SP-400-01, FIG 1) Hand tighten only as the O-
ring under pressure forms the seal, not the fit of the cap lock and seal. DO NOT OVERTIGHTEN Push the
column down into the slurry vial until it meets the bottom of the vial, then withdraw the column by 1/8
inch or far enough to clear the rotation of the stir bar at the bottom of the vial.
6.6 Tighten the Nut SP-130 (or the SP-014) to firmly secure the column in place ) Use two wrenches and
the wrench flats on the Cap Seal to further tighten the nut. Overtightening the nut may embed the
ferrule in the Cap seal port. If this problem occurs, take out the nut, and push the ferrule from the
bottom side. Use a straight piece of 1/6” ss tube, or 1/32 Ferrule extraction tools are available. (contact
the factory)
7.0 PACKING THE COLUMN and LC-MS TIPS
NOTE: IF PACKING WITH LIQUID MEDIA, THE PACKING SOLVENT USED SHOULD BE THE SAME AS THAT
USED TO SUSPEND THE SILICA SLURRY IN THE VIAL. IF PACKING WITH GAS , USE OF NITROGEN OR
HELIUM IS RECOMMENDED.
7.1 Place the assembled Nanobaume and column behind a safety shield and onto a magnetic stir
plate(NOT PROVIDED) .The setup should approximate the setup shown ( FIG 35) Turn on the stir plate.

With the bypass on the SP-050 (FIG 11) high pressure valve still open, slowly turn on the pressure
source. The system pressure will vent through the bypass until the bypass is closed.
7.2 Slowly close off the bypass (clockwise turn) on the SP-050 valve handle to gradually increase
pressure to the Nanobaume. If packing with liquid media, first flood the system and column with
solvent. A small amount of air will be trapped inside the Nanobaume body and will be reduced in size by
the pressure and not impact the packing process. Note: do not increase the packing pressure quickly as
this might break the glass vial with differential pressure.
7.3 Set the flow rate to reach a stable pressure by adjusting the nitrogen tank regulator valve, or the
liquid pressure source. As the slurry packs into the column, the pressure will rise. Take care to reduce
the flow rate to ensure that the pressure limits of the end fitting, column to Nanobaume connection or
the SP-400 pressure Vessel are not exceeded. . Flowrate and resulting pressure are interdependent.
(More flow, higher the back pressure generated with a given bed length).
7.4 Monitor the pressure gauge (SP-087, FIG 8), watching for a stable pressure level. Although the
column is packed once this stable pressure is reached, the silica bed is not yet stabilized. Allow the
pressure source to maintain the column pressure for at least 60 minutes, longer is some cases up to
8 hours to reach a stable pack for longer columns.
7.5 For packing of the LC_MS Tips use gas and pack at pressure not exceeding 3000 psig. Tips pack
quickly and visual confirmation is used in lieu of stable pressure reading.
8 COLUMN REMOVAL AND COMPLETION
8.1 Shut off the pressure source and relieve the system pressure by gradually opening the bypass on the
SP-050 valve (counter-clockwise turn).
8.2 Remove the column from the Cap Seal, loosening the nut counterclockwise 2 full turns. Carefully
remove the column from the cap seal. Note , removal of the column without equalizing pressure in all
parts of the NANObaume setup, might cause the packing material to extrude from the open end of the
column. Please refer to FIG 36 , top column is fused silica column with sleeves, middle is the 1/32”
PEEKSil column, and the bottom is a 1/16” SS body. These columns are not shown with the SS screens,
as they are to small to photograph.

8.3 Install a suitable column end fitting onto the open end of the column (SECTION 4) (FIG 36)
8.4 The newly packed column may be attached to a pump to flush with suitable solvent to waste. The
column is now complete and ready for use.
NOTE: WE OFFER NO GUARANTEE REGARDING COLUMN TO COLUMN PACKING REPRODUCIBILITY,
COLUMN EFFICIENCY (THEREOTICAL PLATES ACHIEVED), OR COLUMN SELECTIVITY OR TAILING FACTORS,
CAPILLARY COLUMN PACKING IS AT THIS STAGE, A HIGHLY EXPERIMENTAL TECHNIQUE.
