American Heart Association Grant #: 0755568B (PI: Pham, Si) 07/01/2007-06/31/10 EFFECTS OF AGING AND BONE MARROW STEM CELLS ON NEOINTIMAL FORMATION The central hypothesis of this application is that the aging process exhausts the pool of bone marrow vascular progenitor cells needed for vascular repair, resulting in exaggerated neointimal formation after injury in the aged. The purposes of this study are: 1) to investigate the mechanisms by which young bone marrow attenuates the exaggerated neointimal formation after vascular injury in the aged, and 2) to determine whether young bone marrow or its components attenuate neointimal formation after vascular injury in aging mice. Total support: $240,000/3 years) 2. Florida Department of Health. Team Science Project # 07KT-02 (Webster, K) 07/01/2007-06/31/09 Cellular Therapeutics and Mechanisms of Vascular Dysfunction in Heart and Kidney Disease Role: Project Leader of Project #3: MECHANISMS RESPONSIBLE FOR EXACERBATED NEOINTIMAL FORMATION BY NICOTINE Summary: The hypothesis of this project is that nicotine exaggerates neointimal formation by augmenting the recruitment of vascular progenitor cells to the site of injury, and by enhancing the proliferation of neointimal VSMC via the PDGF-receptor/ligand signaling pathway. We will test our hypothesis in the following two specific aims. In AIM I, we will determine the intracellular signaling pathways by which nicotine stimulates PDGF-BB production in vascular smooth muscle cells. In AIM II, we will determine whether nicotine increases the recruitment of vascular progenitor cells to the site of vascular injury, and increases the differentiation of these cells in the neointima by stimulating the production of PDGF-BB by VSMC. Total support for the Project: $125,000/yr for 2 years. 3. Transplantation Foundation of South Florida.
