Quantity OneVolume Analysis tutorial

Open the image you want to analyze: Select File > Open

Zoom in on the area of the image that you want to analyze

Zoom Tool to magnify regions of the gel

Full View Tool see the entire gel image

From the Volume menu, choose the type of volume tool you want to use. (See the next slide for a description of the tools.)

Volume Tools Options

Volume Contour Tool: Automatically conforms to the outer edge of the object and can be dragged to include or exclude area

Volume Free Hand Tool: Allows you to draw the edge you want

Volume Rect Tool: Draws a rectangle shaped box that can be dragged to the desired size (recommended method for most applications)

Volume Circle Tool: Draws a circle that can be dragged to the desired size. This tool is more useful for dot blots and 2-D gels

Choose the rectangle tool. click and drag to create the box. TIP: To make all of the volumes exactly the same size, copy the volume box by clicking the box while pressing the control key. Drag the copied box to the desired location and release the mouse.

Double-click on a volume to change the label names or designations of a volume. You can designate unknowns (U), Standards (S) or Background (B). Enter a concentration for each Standard. NOTE: Volumes are automatically numbered. TIP: To delete a volume, select it and press the Delete key.

Generate a Volume Regression curve if you have designated at least two standards and entered the concentration. Select Reports > Volume Regression Curve.

This is an example of a Volume Regression Curve. TIP: You can select different Regression methods.

Select the Volume Analysis Report to see the data and calculations.

Choose the information that you want to display in the Volume Report Options window by selecting options. TIP: For more information about the report options, check in the Instruction manual (Section 12.6 in PDF) and the online Help.

This is the Volume Report. Reports can have more than one page. TIP: To export the data to a spreadsheet (such as Excel), click the Export button
