It has been suggested that squamous cell carcinoma (SCC) inyoung patients is a distinct disease from that occurring in olderpatients. Retrospective samples of young and adult patientsdiagnosed with SCC of the tongue and floor of the mouth wereselected. Histopathological analysis was performed using theWorld Health Organization (WHO) grading system and histo-pathological risk assessment. Immunohistochemical staining wasused to evaluate the expression of HER-1, HER-2, and p16.Younger patients were classified as having a high risk forrecurrence. All cases were immunopositive for HER-1 and 100%of the sample was negative for HER-2. The p16 immunostainingwas greater for younger patients. HER-1 showed an importantrole in the carcinogenesis of SCC of the tongue and floor of themouth.

Cisplatin-based chemotherapy is currently the standardtreatment for head and neck squamous cell carcinoma(HNSCC). Its efficacy is influenced by the development of tu-mor resistance. Here we explore a novel mechanism by whichNFkB drives HNSCC resistance. Methods and Results: NFkBand BRCA1 were evaluated in HNSCC cell lines. HNSCC cellspresenting active NFkB showed loss of nuclear BRCA1 andincreased resistance to cisplatin along with genomic instability,gH2AX accumulation, and chromatin acetylation. Pharmaco-logical administration of HDAC inhibitors increased cisplatincytotoxicity. NFkB is a key signaling pathway involved in thecontrol of chromatin acetylation and tumor chemoresistance inHNSCC. Using NFkB interference via siRNA, we observedincreased cisplatin cytotoxicity along with increased chromatinacetylation observed by nuclear morphometry and increasedlevels of acH3. Conclusion: Our results suggest that HDACinhibitors and NFkB targeted therapies may represent viableadjuvant chemotherapy to revert HNSCC resistant to platinum-based therapies.

In the antineoplastic treatment of cancer of the head andneck the main complication is oral mucositis. This studydeveloped and characterized the use of oral mucoadhesivebuccal devices (DB), hydrogels use for the prevention and/ortreatment of mucositis. Materials and Methods: The DB in theform of films containing lidocaine/ benzidamide/N-acetylcys-teine were prepared with carboxymethylcellulose, chitosan, andhydroxipropylmethylcellulose K4M and sweeteners for betteradhesion. The hydrogels were characterized by rheology andfilms for mechanical strength, swelling, and mucoadhesion.Result: The hydrogels exhibited dilating behavior, and the DBwith 4 cm2 showed satisfactory organoleptic characteristics,excellent mechanical properties for handling, and mucoadhe-sion. Mucoadhesion was accomplished by one-way links be-tween hydrogen atoms and mucin. The controlled release ofdrugs was done by increasing the diffusion layer. Conclusion:The DO is a promising alternative for patient care in anticancertreatment that can be useful in controlling pain and inflammationof the oral mucosa.
PE-432 - ORAL FUNCTION REQUESTS IN IN-DIVIDUALS WITH A DIAGNOSIS OF BURNINGMOUTH AND XEROSTOMIA. Symptoms may bring discomfort or oral modifications tostomatognathic functions. Materials and Methods: We evaluated66 subjects age 30 to 78 years and divided them into threegroups: burning mouth group, xerostomia group, and groupwithout oral symptoms. Interviews, oral inspection, and speechrecordings were taken for all groups. Results: The identificationof general oral profile and clinical symptoms of the pathologicgroups demonstrated similarities in many aspects whencompared to the control group. The xerostomia group had morecomplaints of oral functions. Noise in speech was the prevalentsymptom in the burning mouth and xerostomia groups. Com-plaints of oral symptoms may go unnoticed and mask relevantdiseases. Multidisciplinary investigation enables early diagnosisand appropriate referral. Conclusion: Oral complaints candevelop in the presence of symptoms of burning and dryness ofthe mouth. These symptoms are evidenced by the presence ofnoises during speech.
