10(b) Eminence of nominee among peers in the profession as a person of

integrity and distinction at national/international level

The followings are the few novel contributions to the field of medical science:-

[I] Major thrust area of my research is typhoid fever and chronic typhoid carriage for last

25 years. In this field I have made a significant contribution for the first time in World

Literature of Medicine

[A] Chronic typhoid carriage and cancer gallbladder

As a major breakthrough research,

I have pioneered the work to establish the definite causal role of Salmonella Typhi

(chronic typhoid carriage) in cancer gallbladder. After Helicobacter pylori, this is the

second bacteria which could have been implicated for the first time in causation of cancer

of the organ where it has niched i.e. CaGB that too by our collaborative team. We have

delineated this role in our recently published (May 28, 2015) article entitled “Salmonella

Manipulation of Host Signaling Pathways Provokes Cellular Transformation Associated

with Gallbladder Carcinoma” in “Cell Host and Microbes”. This work has proved in

transformed experimental animal model and organoid and cell lines that S.Typhi induces

the malignancy by inducing the MAPK and AKT pathways which is also shared by human

cells. Intriguingly, these pathways are used by the bacterium for their intracellular survival.

While viruses are among the established causal factors for particular cancers, bacteria

are largely ignored as direct contributors. Accepting that bacterial infections can directly

contribute to cancer formation makes these tumors in principle preventable. Therefore, we

have concluded that if Salmonella Typhi infections are cured immediately with antibiotics

and chronic infections are prevented, or if vaccination programs to eradicate S. Typhi work,

we would expect a major reduction in the incidence of a tumor that represents the third

most common gastrointestinal tumor in India and Pakistan."

i) We could establish that S. Typhi basically stays in liver rather than gall bladder.

ii) Diagnosis of typhoid fever could be made almost in 100% of the cases by using

nested PCR for the first time by us.

iii) Widal test in the diagnosis of typhoid fever is very relevant and useful. This test is

usually does not give so called false positive results.

[B] Pioneering work in BACTERIOPHAGE THERAPY

a) Standardized and evaluated the phage therapy for acute septicaemic model

against Pseudomonas aeruginosa in burn mouse model. The phage strains have

been patented by Indian Patent Authority (Certificate of filing of patent from

Indian Patenting authorities has been received. Final application submitted on

December2, 2015 (Annexure VIII).

b) The other breakthrough research carried out by me is: For chronic as well as

acute infections caused by Staphylococcus aureus especially osteomyelitis,

phage therapy has been evaluated in rabbit osteomyelitis model (research article

has been accepted in Indian Journal of Medical Research, December, 2014 will

appear in December, 2015). Phage therapy evaluation is underway for chronic as

well as acute Salmonella Typhimurium infection in mouse model is underway as

a prototype for S. Typhi.

c) We have generated potential mutants of Salmonella Typhi as vaccine

candidates. I am deeply involved in development of candidate vaccine for

Salmonella Typhi and Paratyphi A by creating various mutant especially targeting

stress genes and their regulator (Sigma factors) are under evaluation specially

rpoE, rpoS mutants.

[C] I am in search of natural bioactive molecules (antimicrobials)-

We have explored the efficacy of the parts of fruit and flower of Mallotus phillipensis

(Kampilla in Indian Medicine) showing effect on cestodal infections.

Personal Recognition at International level

1. I was invited as member of selection committee for King Faisal Foundation Award in

Medicine, Feb, 2015 Saudi Arabia. This was as recognition of research contribution in

the areas of Gastrointestinal Infections. Feb, 2015

2. I was invited as guest faculty to Enteric Fever Diagnostics meeting on 27-28 October

2010 in London by London School of Hygiene & Tropical Medicine, UK due to my

significant contribution in the area of typhoid fever.

[D] Social service

I am regularly visiting rural schools of primary, middle inter mediate levels and

degree colleges for delivering motivational lectures for last 15 years as a part of social

service which is due on me. Frequency of my interaction with the rural youth is

approximately 10 per year. I give 10% of my quality time for this purpose. Associated

with community oriented projects and regularly visiting and interactive with the rural

people regarding health issues.
