Purpose This SOP covers the precautions and safe handling procedures for the use of Mutagens and Reproductive Toxins. For a list of Mutagens and Reproductive Toxins covered by this SOP and their use(s), see the “List of Chemicals”. Procedures described in Section 12 apply to all materials covered in this SOP. A change to the “List of Chemicals” does not constitute a change in the SOP requiring review or retraining. Physical & Chemical Properties/Definition of Chemical Group Germ Cell Mutagenicity is a hazard class that is primarily concerned with chemicals that may cause mutations in the germ cell of humans that can be transmitted to the progeny. A mutation is defined as a permanent change in the amount or structure of genetic material in a cell.

Mutagensareagentsgivingrisetoanincreasedoccurrenceofmutationsinpopulationsofcellsand/ororganisms.Reproductivetoxicityincludesadverseeffectsonsexualfunctionandfertilityinadultmalesandfemales,aswellasdevelopmentaltoxicityintheoffspring.Thesechemicalsmaypresentextremeriskpotentialtolaboratoryworkersifnothandledappropriately;therefore,thesesubstancesmayrequireadditionalcontrolmeasureswhenused.

3.PotentialHazards/Toxicity

MutagensandReproductiveToxinsarechemicalsthataffectthereproductivecapabilitiesincludingcausingchromosomaldamage(mutations)andadverseeffectsonfetaldevelopment(teratogenesis).AsdefinedbytheGloballyHarmonizedSystemofClassificationandLabelingofChemicals(GHS),reproductivetoxinsaredesignatedbyoneormoreofthefollowingHcodes:

H340MaycausegeneticdefectsH341SuspectedofcausinggeneticdefectsH360MaydamagefertilityortheunbornchildH361SuspectedofdamagingfertilityortheunbornchildH362Maycauseharmtobreast-fedchildren

Additionalrequirementsmayapply,dependingonthespecificchemical.Examplesincludeparticularlyhazardoussubstancesthatarealsohighlyflammableand/orreactive.Safeuserequiresassessingallpotentialhazards.ItisthePrincipal Investigator’s responsibility toensureactivity-specific laboratoryproceduresand/orprocessesaretakenintoaccountwhenusingthisChemicalClassSOP.Please,reviewtheSDSofanychemicalbeforeuse(seeSection11–SDSLocation).

4.EngineeringControlsUsetheengineeringcontrolslistedbelowunlessotherlab-specificinformationisincludedinSection12-Protocol/Procedure.

• Workwithmutagensandreproductivetoxins–theworkmustbeconductedinafumehoodunlessothercontrolsaredesignatedinthelab-specificProtocol/Proceduresection.Sashheightmustbekeptaslowaspossibletoavoidescapingfumesandprovideaphysicalbarrier.

• LaboratoriesandroomswhereMutagensandReproductiveToxinsareusedmusthavegeneralroomventilationthatisnegativepressurewithrespecttothecorridorsandexternalenvironment.Thelaboratory/roomdoormustbekeptclosedatalltimes.

5.PersonalProtectiveEquipmentAtaminimum,thefollowingPPEmustbewornatalltimes.EyeandFaceProtection

A. ANSIZ87.1-compliantsafetyglasseswithsideshields,orchemicalsplashgoggles.

• Ordinary prescription glasses will NOT provide adequate protection unless they alsomeetANSIstandardandhavecompliantsideshields.

GototheOccupationalHealthFacility(TangHealthCenter,oncampus); ifafterhours,gotothenearestemergencyroom(AltaBates,2450AshbyAveinBerkeley);or

Call911(fromacellphone:510-642-3333)if:

• itisalifethreateningemergency;or

• younotareconfidentinyourabilitytofullyassesstheconditionsoftheenvironmentand/orthe condition of the contaminated/injured person, or you cannot be assured of your ownsafety;or

• thecontaminated/injuredpersonisnotbreathingorisunconscious.

Please remember to provide a copy of the appropriate manufacturer SDS (if available) to theemergency responders or physician. At a minimum, be ready to provide the identity/name of anyhazardousmaterialsinvolved.

Incaseofskincontact

Ifskincontactoccurs,and/orskinorclothingareonfire,immediatelydrenchinthesafetyshowerwithcopious amounts of water for no less than 15 minutes to remove any remaining contaminants. Ifpossibletodosowithoutfurtherinjury,removeanyremainingjewelryorclothing.

Rinse thoroughlywith plenty ofwater using an eyewash station for at least 15minutes, occasionallyliftingtheupperandlowereyelids.Removecontactlensesifpossible.

● Asanalternative,unrinsedemptycontainerscanbedisposedofthroughEH&Sashazardouswaste.Theunrinsedemptycontainersmustbecapped.

● Donotmixwithincompatiblewastestreams.
