MITOSIS

Observations of mitosis • Preparation of specimens of onion root tips

• Materials: onion root tips, fixative, maceration solution, aceto-orcein stain, distilled water, forceps, microscope slides, cover slips, microscope, razor blade, petri dish, scissors, test tubes

• Cut 1 cm long root tips, trasfer them into fixative

• Transfer root to petri dish containing maceration solution

• Transfer root to another petri dish with water

• Place the root tip on a slide and cut 2 mm at the very tip of the root, cover the root tip with a few drops of stain

• Cover the root tip with a cover slip, carefully push down od the cover slide
