HighThroughput Parallel SynthesisMAGELLAN is a high-throughput parallelorganic synthesiser designed to automatethe production of large numbers ofcompounds using resin-based, solid phasechemistry. MAGELLAN automates thefollowing processes:
Addition of reagents & solvents Heating and agitation of reactions Washing of resinsCleavage and collection of compoundsDistribution of compounds prior to HTS
Maximum Productivity MAGELLAN incorporates a number ofunique features designed to maximise theefficiency of organic synthesis including:
Removeable 96-position Reaction Block4-channel Solvent Delivery ProbeIntegral Heater-AgitatorDedicated Reagent Addition Probe8-way Solvent Selector Valve96-channel Filtration ManifoldPC controlled, off-line Heater-Agitators
Robust and ReliableA key consideration in the design ofMAGELLAN was to use practical, wellproven technology throughout. This hashelped to ensure maximum reliability inoperation as well as ease of maintenanceby both user and service engineer alike. Some examples of this approach includethe use of:
Robust liquid handling systemsSimple Reaction VesselsEasily replaceable tubing and filtersChemically resistant materials
Powerful Integrated Software MAGELLAN’s Windows software controlsboth the instrument and all chemicalprocesses. A Synthesis Task List gives theuser the means to control all synthetic stepsand allows highly complex chemistries to beset up. It also provides the interface to thelaboratory’s own computer systems.
Productivity is enhanced further bynetworked controlling software which co-ordinates the operation of any number ofMAGELLAN synthesisers and Heater-Agitators. This ensures that all units worktogether for maximum efficiency.
Automated Reagent & Solvent AdditionReaction Vessels containing pre-dispensedresin are initially loaded into a 96 positionReaction Block. This is identified by its barcode and placed in MAGELLAN’s IntegralHeater-Agitator. Reagents are then addedas specified in the Synthesis Task List.
On and Off-Line Heating & AgitationThe Reaction Block is agitated and, ifrequired, heated. If reaction times are longit can be removed and placed in an off-lineHeater-Agitator to maximise the use of thesynthesiser for more intensive pipetting andwashing steps.
Each off-line Heater-Agitator is also linkedto a PC. When a Reaction Block is placedon an external unit it is first identified by itsbar code. MAGELLAN’s network softwarethen resumes control of the reaction. Whenthe reaction is complete it can then placedback on the MAGELLAN for washing.
High-Speed Resin WashingDuring a wash cycle the 96-channel filtrationmanifold aspirates excess reagents to oneof two separate waste bottles. A solvent isthen dispensed into the reaction tubes torinse the Filtration Needles and the resin.A number of wash cycles using differentsolvents can be performed before the nextset of reagents is added.
Cleavage and CollectionWhen the synthesis is complete, cleavagereagent is added to the Reaction Vessels. A 96-position Collection Block is loaded intothe Collection Bay and released compoundsare then transferred by the FiltrationManifold into Collection Tubes.
Automated Compound DistributionAfter compound collection, the CollectionBlock can be removed and the compoundsdried down. The Compound CollectionBlock can also be placed on theMAGELLAN bed and the contentsdistributed directly into bar codedmicroplates and vials. All identifiers arestored in a database and can be used laterto control downstream processes.
A separate automated CompoundDistribution system is also available. This isparticularly suitable for the higherthroughput laboratory generating largenumbers of compound libraries.
