I. Introduction
I-1. Safety
The following safety symbols are used on this product.
Warning Description
Always refer to the system manual when working near
locations at which the alert mark shown on the left is
attached. If the operation, etc., is performed without
heeding the advice in the System manual, there is a risk
of personal injury. In addition, the equipment
performance may be reduced. Moreover, this alert mark
is sometimes used with other marks and descriptions
indicating other dangers.
Warning
When supplying power to this equipment, connect the
accessory 3-pin power cord to a 3-pin grounded power
outlet. If a grounded 3-pin outlet is not available, use a
conversion adapter and ground the green wire. If power
is supplied without grounding the equipment, there is a
risk of receiving a fatal electric shock and equipment
damage.

To indicate connection to the mains, at least for mains
switches or their positions, and all those cases where
safety is involves

Repair
Warning
The user cannot repair this equipment.
DO NOT attempt to open the case or to disassemble
internal parts.
Only Thermo Scientific trained service personnel or staff
from your sales representative with knowledge of
electrical fire and shock hazard should service this
equipment.
There are high-voltage parts in this equipment
presenting a risk of severe injury or fatal electric shock
to untrained personnel.
In addition, there is a risk of damage to precision parts.
Falling Over
Caution
This equipment should be used in the correct position. If
the equipment is turned on its side, etc., it will be
unstable and may be damaged if it falls over as a result
of receiving a slight mechanical shock.
Changing Fuse
Warning
Before changing the fuses, ALWAYS remove the power
cord from the power-outlet and replace the blown fuses.
ALWAYS use new fuses of the type and rating specified
on the fuse marking on the rear panel cabinet.
T2AL indicates a time-large fuse. There is risk of
receiving a fatal electric shock if the fuses are replaced
with the power cord connected.
Cleaning
Warning
Keep the equipment free of dust.
Clean the power line regularly; if dust accumulates
around the power pins, there is a risk of fire.
Keep the equipment clean so that the ventilation holds
are not obstructed. If the ventilation is obstructed, the
system may overheat and catch fire.
Periodically clean your equipment's case using a damp
cloth.
Do not use abrasives, cleaning solvents or strong
detergents, as they may damage the finish or affect the
reliability of the structural components.
If this equipment is used in a manner not specified by
this manual, the protection provided by the equipment
may be impaired.

II. Installation
II-1. Pre-Installation
II-1-1. Instruction
Lumina fluorescence spectrometer is designed to provide a
complete analysis system for fluorescence, phosphorescence
and chemiluminescence measurements.
All instruments will be tested and proven to the performance
before the shipment
II-1-2. Pre-Installation Checklist
Preparation Done
The installation place should be prepared / checked. □
The electric power is available / stable. □
The system and accessories should be unpacked on site. □
Proper computer system should be prepared. □
Windows operating system should be prepared. □
Quartz cell for fluorescence is available. □
Operation training is required. □
II-1-3. Environmental Conditions
You are responsible for providing an acceptable operating
environment. Attention paid to the operating environment
well ensures the continued high performance of your Lumina
fluorescence instrument. The instrument warranty will be

made void if the equipment is operated in sub-standard
conditions.
Humidity
The relative humidity of the operating environment should be
lower than Maximum 75% with no condensation. Operating
the Lumina fluorescence in very low humidity may result in
the accumulation and discharge of static electricity that
shortens the life of electronic components. Operating at high
humidity will produce condensation and result in short
circuits. Thermo Scientific recommends that your laboratory
be equipped with a temperature / humidity monitor. This will
ensure that your laboratory is always in conformance with the
temperature and humidity specifications.
Temperature
Air conditioning is recommended. The room should be
temperature controlled if your analyses are particularly
sensitive. For optimum analytical performance it is
recommended that the ambient temperature of the laboratory
be between 15-35 and be held constant to within +/℃ -2%
throughout the entire working day. As laboratory temperature
increases, system reliability decreases and all electronic
components generate heat while operating. This heat must be
dissipated to the surrounding air so that the components are
operating correctly.
Vibration
Ensure that lab benches are free from vibration. If possible,
any equipment generating vibration during the operation
should be installed on the floor rather than alongside the
system on the workbench.

Work bench
The workbench must be stable and durable enough to support
the total weight of equipment. The worktops should be large
enough to permit a free circulation of air around each unit in
the system. Remember to provide space for the computer,
monitor and printer. To avoid damage through spillage of the
samples being analyzed, the worktops should be covered with a
material which is corrosion resistant and impervious to liquids.
** Lumina dimensions are 620 mm (W) x 710 mm (L) x 300 mm (H)
II-1-4. Electrical Specifications
In view of the electronic components in the Lumina system, it
should be positioned well away from any instruments
emitting an electromagnetic field and any environment that
may cause vibration.
Power Supply
All power supplies must be single phase AC voltage, three wire
system (active, neutral, ground) and should be terminated at
an appropriate power outlet and/or regulations imposed by
local authorities responsible for the supply of electrical energy
to the workplace.
A grounded AC power supply must be used. The Lumina
accepts a dual voltage of either 110 or 220V at 50㎐ or 60㎐
and requires 300 W power including computer and printer.
Good electrical grounding is essential to avoid potentially
hazardous shock hazards. 3-wire outlet with ground
connection must be provided for the instrument. Make

certain that power outlets are grounded at the grounding pin.
Avoid using power supplies from a source that may be subject
to electrical or RF interference from other services (large
electric motors, elevators, and welders for example).
Two fuses are used in the Lumina fluorescence spectrometer;
Fuse A: AC 250V, T2AL
Fuse B: AC 250V, T5AL
II-1-5. Equipment on-site
Insurance
As the carrier’s liability ceases when equipment is delivered,
Thermo Scientific recommends that the instrument owner arrange
a separate insurance to cover transportation from the delivery point
to the installation site. The delivery point will vary according to the
carrier, the shipping method and in some cases the terms of sale.
Some carriers will deliver only to their own distribution centers,
who then will deliver to the actual installation site.
Inspection for transit damage
Transit damage can be obvious or concealed and in either case will
be admitted by the carrier only if it is reported within the terms of
the carrier’s agreement. For any claims against damage in transit,
the following general rules apply;
- Before accepting delivery, you must inspect the packages for signs
of obvious damage. The nature of any obvious damage must be
noted on the carrier’s waybill, which then must be countersigned
by a representative of the carrier

- Within the time limit stated in the terms and conditions of
carriage, a further inspection must be made for concealed
damage. If any damage is found at this stage, the carrier must be
modified in writing. You must retain all packing material for
subsequent inspection by a representative of the carrier.
- A copy of any damage report must be forwarded to Thermo
Scientific or Sales representative.
- ‘SHOCK WATCH vibration indicator label that helps you to
aware of the damage level are attached on Packing Boxes. If the
indicator color is changed as red color, it means the packing box
could be impacted/damaged by the physical shock externally.
If it is happened, contact Thermo Scientific or sales
representative immediately to hear the next procedures.

III. Lumina Installation
1. Carefully remove the instrument and all of its components
from the shipping container.
2. Verify that all of the components received match the
packing list.
3. Verify that all system components required for installation
are present, this, must include:
□ Lumina Fluorescence Spectrometer Interface cable□ (RS-232) Power Cord □ □ Luminous Software CD Computer systems □
4. Place the instrument in a location that is compatible with the
required environmental conditions for operation. Connect the
power cord to the plug; the power plug must be grounded.
Warning
The electric power could be dangerous to the body. Before
connecting the power cord, verify that the main power
switch is off.

IV. Luminous Software Installation
IV-1. How to Install Luminous software
1. Insert Luminous Software installation CD into the CD-ROM
drive. InstallShield® Wizard starts automatically.
2. If InstallShield Wizard does not start automatically, click My
Computer → E Drive and then, double click Setup.exe and click
Next.
3. When the following dialog box appears, enter customer
information, if necessary, and click Next.

4. The destination folder and user information will be shown. Click
Install. Installation will begin.

5. After the set up is completed successfully, click Finish.
6. Find the newly created ‘Luminous Software’ folder on the
Desktop.

IV-2. Re-Install of Luminous software
1. To re-install the Luminous program, previously installed.
2. Luminous program must be removed. Select Start →
Control Panel Add/Remove Programs.→
3. Select the Luminous. Then click Remove and select Yes.
4. Luminous program will be removed automatically.
Thereafter, reinstall the Luminous software.

V. Operating Guide
To start the Luminous Software, follow up the procedures
below.
1. Turn on the main power switch on the rear panel of the
system.
2. Turn on the sub main power switch at the bottom right
side of the front panel of the system.
3. Push the Light Source button (Red) to turn on light firstly
and push the Control Board button (White) to turn on
the main board.
Caution!
a. It is important to keep up the order during turning
powers on:
#1 Main P/S #2 Light Source #3 Control Board
b. If the control board is powered on at first , it could cause
damage to the electric board by the high voltage/current
when the light source is on later.
c. To turn off the system, you need to follow the procedures

in reverse order against the start up. (#3 Control Board->
#2 Light Source -> #1 Main P/S)
d. Before turning off the main power switch, you need to
wait for over 10 minutes at least to protect the lifetime of
lamp after turned off the light source switch.
4. Check whether all three LED lights at the left side of the
front panel of the system are lit on. If it is ready, warm up
the system for about 20-30 minutes.
5. Double click / open the Luminous Software folder on
Desktop.
6. Double click Wave Scan in the Luminous Software
folder, then the following windows will be shown.

7. Click NEW in the toolbar.
8. Enter a file name in the New window and click Save.

9. The Create New Experiment window is displayed. Enter title,
customer and User ID if it is needed and then click OK.
10. The main window will be displayed.
11. Set up parameters in the measurement setup window
depending on the experiment conditions. Refer to the
Luminous software user guide for more details.(Chapter
IV. Measurement Mode)

12. Place the sample in the sample cell holder.
13. Click the Start button.
14. The result spectrum and data will be displayed as below.
15. Data can be printed out by clicking the Report View
icon. See the Lumonous Software User Guide for
more details.

VI. Diagnostic Software
Diagnostics software is to check if each component of
instrument works correctly.
Teflon Block is needed for diagnostics.
1. Turn on the main power switch on the rear panel of the system.
2. Turn on the sub main power switch at the bottom right side of
the front panel of the system
3. Push the Light Source button (Red) to turn on light firstly
and push the Control Board button (White) to turn on the
main board.
Caution!
a. It is important to keep up the order during turning powers
on:

b. If the control board is powered on at first , it could cause
damage to the electric board by the high voltage/current
when the light source is on later.

6. Click OK after ‘Reading serial number: OK window’ is
displayed.

7. You can check the serial number of instrument
8. Select the items which you want to inspect and enter the proper
repeat numbers in Repeat Number boxes on the Main Controller
Test panel. For the initial inspection, all items are recommended
to be checked and the proper repeat number is ‘2-5’.
9. Click Start.

10. You will see the following message.
11. Place the Teflon block into the cell holder, and then click
OK.
12. The total diagnostic progress status and the progress status of
each component will be shown.

13. If the diagnostic passes the test, then the following message will
be shown.

14. If each procedure has no problem, the test result will be
displayed as ‘pass’. If any procedure has a problem, ‘fail’
message will be displayed and the system will be stopped.
15. If the result is failed, the following message will be shown. If
you did not recalibrate the wavelength in Alignment.exe,
recalibrate both monochromators using Alignment.exe

16. Click Print to print out the result of diagnostics.
17. Click Help to search the items to be defined.

Error Cause Corrective action
Detector cannot receive the signal.
Check the cable connection or Replace PD board.
Excitation Slit Failed Start position is different every time or Slit Wheel rotates continuously.
Limit switch could be defected. Replace the limit switch.
Xenon lamp is not turned on.
Run the diagnostic software.
Excitation Filter positions are not correct.
Open the upper cover and check the Excitation Filter positions. If it is misaligned, Recalibrate the Filter positions using Alignment software.
Excitation Slit positions are not correct.
Open the upper cover and check the Excitation Slit positions. If it is misaligned, Recalibrate the slit positions using Alignment software.
Excitation Monochromator is not correct.

VII. Maintenance
VII-1. Xenon Lamp Replacement
The Xenon lamp for replacement is packed in the
manufacturer’s box and must be installed in the lamp housing
correctly. Read all the precautions before attempting to replace
the Xenon lamp.
Caution
A. Xenon arc lamp has an explosion hazard. Make sure the
system’s power is off and all AC power is disconnected from
the system. Read and follow the cautions presented below.
B. Xenon arc lamp has an explosion hazard. Wear goggles and
protective clothing when opening the lamp housing and
when handling the lamp.
C. The lamp will remain extremely hot for approximately one-
half hour after it has been turned off. Do not touch the
lamp or the metal unit until the lamp has cooled.
D. Never look directly at the xenon arc or its reflection.
Intense radiation can permanently damage eyes.
E. Do not touch the focusing lens, back scatter mirror, or the
surface of the lamp. Fingerprints will be burned onto these
surfaces when the lamp is ignited.

Xenon Lamp Replacement Flow Chart
1) Start
2) Turn off the power of the instrument.
3) Remove white screw caps (total 10) located around on the
upper case.
4) Remove Phillips Head screws using a Phillips screwdriver.
5) Remove Phillips head screws (4 ea) located on the Sample Cell
Assay part using a Phillips screwdriver.
6) Separate the Sample Cell Assay part and lift the upper case.
7) Get rid of socket screws (4 ea) using a hexa socket screwdriver.
8) Separate the lamp connector from the lamp power module
and push the hap and pull out the connector.
9) Remove the lamp mounting module from the lamp housing.
10) Remove the screws (2 ea) located on the edges of the lamp.
11) Unfasten the socket screws (2ea) located on the (+) side
of the Teflon bracket.
12) Unpack a new Xenon lamp.
13) Install the lamp in the correct polarity.
14) Fasten the socket screw (2ea) located in (+) direction
Teflon bracket. Put the changed lamp module back in the
lamp housing.
15) Finally, connect the lamp cable to the power supply cable.

reliably and predictably. If the instrument has a problem, take
measures by referring to the table below. When a symptom in
question is not found in the table, contact Thermo Scientific or sales
representative.

1. Lumina Fluorescence Spectrometer
The rapid growth & convergence of high technology has created

measurements and highly stable results. Thereby it maximizes
laboratory efficiency in various fields such as pharmaceutical,
environmental and life sciences, etc.
2. Basic Theory of Fluorescence Spectrometry
Fluorescence Spectrometry, also known as fluorometry or
spectrofluorometry, is a type of electromagnetic spectroscopy which
analyzes fluorescence from sample. It involves using a beam of light,
usually ultraviolet and visible light, which excites the electrons in
molecules of certain compounds and causes them to emit light of
lower energy, typically, but not necessarily, visible light. A
complementary technique is absorption spectroscopy.
Molecules have various states referred to as energy levels.
Fluorescence spectroscopy is primarily concerned with electronic
and vibrational states. Generally, the species being examined will
have a ground electronic state of low energy and an excited
electronic state of higher energy. Within each of these electronic
states are various vibrational states.

In fluorescence spectroscopy, the species is first excited, by
absorbing a photon, from its ground electronic state to one of the
various vibrational states in the excited electronic state. Collisions
with other molecules cause the excited molecule to lose vibrational
energy until it reaches the lowest vibrational state of the excited
electronic state.
The molecule then drops down to one of the various vibrational
levels of the ground electronic state again, emitting a photon in the
process. As molecules may drop down into any of several
vibrational levels in the ground state, the emitted photons will have
different energies, and thus different frequencies. Therefore, by
analyzing the different frequencies of light emitted in fluorescent
spectroscopy, along with their relative intensities, the structure of
the different vibrational levels can be determined.
In a typical experiment, the different frequencies of fluorescent

Lumina uses CW Xe-lamp as a source. The forward light is
collimated by Collimation lens(CL1) and the backward light is
collected/reflected by Reflection mirror(BR) and then, it will be also
collimated by Collimation lens(CL1). The collimated beam will be
focused by Focusing lens(FL1) and go through Excitation slit(SEX)
which is in Monochromator. The other light could be filtered using
Excitation Filter(FEX) except the specific wavelength if it is
necessary. After slit, the incident light will be reflected by Concave

Mirror(MEX1) which is 190mm far from slit and then, will be
dispersed by Excitation Grating. The dispersed light is reflected

exit slit by controlling the incident angle and reflective angle of
Grating (GEX). The monochromated light after passing through
the exit slit is reflected by Flat Mirror (M2) and again reflected by
Concave Mirror (M1) then, will be focused. The focused light goes
through the beamsplitter(BS) and the 2% portion of light goes to
Photodiode(PD) after reflected and other 98% pass through the
sample.
Using UV grade Window(UVW), the inner optical parts will be
protected from the outer air and dust. The sample absorbed by the
Excitation light will emit the fluorescence and the emitted
fluorescence will be collimated after passing through the collimation
lens(CL2) and focused by Focusing lens(FL2) then, goes to the
monochromator through the Emission entrance slit. The unwanted
wavelength could be filtered by the Optical filter (FEM) before
going to the monochromator per request. The incident light is
reflected by Mirror(MEM1) and monochromated by
Grating(GEM) then, the selected light will be passed through the
exit slit of Emission monochromator. This selected light goes to
PMT and the electrical signal will be read out after processing.
