The samples used were fibroblast cells of ear skin tissue of goats’ fetus of 2 - 3 cm, 7 - 8 cm, and 12 - 15 cm in size. The treatment concern with the fetal length, i.e. (1) fetus of 2 -3 cm in size, (2) fetus of 7 - 8 cm in size and (3) fetus of 12 - 15 cm in size, with three replications. Variables measured are cellular sizes (S = 3 – 4 µm; M = 5 – 6 µm; L = 7 – 8 µm); numbers, counted with hemocytometer; and viability, counted by staining with Hoechst 33342. Data were analyzed by Anova and BNT-test. Goat fetuses were obtained from a slaughterhouse and transported to the laboratory in a thermos, then in laboratory fetus ear were obtained and primary cells isolate with mechanics. Cells were cultured in TCM 199 suplemented with 10% FBS and DMEM suplemented with 10% FBS. Culture were observed at the 4th; 7th and 14th day. The research results show that the treatment to fetus of 7 - 8 cm in size with TCM 199 medium supplemented with 10% of FBS on the culture on 14th day has the most cellular numbers of 53.59 ± 2,36% among the M-sized (5 – 6 µm) cells, the highest cellular numbers of 1.6 x 106 ± 0,08 cell/ml, and the highest viability value of 85.60 ± 1,69% . ABSTRACT

Tanta & Uteng Datunsolang. 2009. The Biology of Vanda dauglas Lindl. (Orchidaceae). Novae Guinea 1(1): 17-27. – A study of flower biology of Vanda dauglas Lindl. was carried out to obtain the morphological development of flower, the characters of self incompatibility and some

visitors insect. The methods to test a character of self incompatibility were outogamy and xenogamy pollination. The result of the research showed that an inflorescence raise from its stem, racemes, the anthesis centripetally (racemosa) with hercogamy characters. The character of self incompatibility is 100 percent and potential insect in helping pollination is a garden bee (Bombus pensilvanicus).

Abstract

Research related to urban imperviousness impact became an important agenda in catchment hydrological studies, both in developed and developing countries. The main objective of this study is to understand the equilibrium of environment-development linkages – specifically to study the effects of urban imperviousness on ecosystem health. Urban imperviousness as a river ecosystem health indicator has become widely accepted by river catchment professionals, especially in the United States of America and also elsewhere. Catchment imperviousness is not solely applied in environmental planning and river ecology, but has also recently been used to formulate new urban planning strategies to enhance environmental quality. In the context of catchment hydrology, imperviousness prevent infiltration and seepage to occurred, thus affecting the streamflow characteristics and basin’s hydrological cycle. This article highlights global and local issues related to imperviousness, its research methodological development and its potential use as an ecosystem health indicator for catchment areas in Malaysia.Item Type: Journal

Abstract

The research describe about surface water ecosystem study in Merauke Papua by using multisource and multistage remote sensing data which was splitted into two parts based on the study areas. First, it is for micro scale spatial analysis focusing on the Rawa Biru - Torasi watershed, while the second is for macro scale spatial analysis for Transfly ecoregionin the floodplain areas of Merauke. Multispectral approach was adopted for Landsat image analysis, followed by field survey on the selected areas. Auxilary data used are maps, secondary documents in order to improve understanding of the areas. Interview and discussion with related institutions (Wasur National Sanctuary, Potable Water Services, Internal Affairs Government, Forestry Service) accordingly were carried out. The research result showed that remote sensing imagery are usefull for surface water resources study. Physical condition of the Rawa Biru - Torasi watershed, vegetation analysis by using multitemporal data, wetland type, hydrological process in the floodplain were presented. Rawa Biru watershed as a resource for drinking water supply environmentaly decreased considerably due to the species invasion, with successively dominated by hydrophilla, tebu rawa, rumput pisau, dan Mellaleuca and sedimentation took place in the main body of swamp. Upper part of the watershed is actually included in the Papua New Guinea, in long water resources development scheme, it need administratively belong to one recharge areas for the watershed.Item Type: Article

This research is an improvement research instrument of Elementary Science Learning that was oriented on the approach of guided discovery. It was implemented into the learning activity. The background of this research was the reality of learning activity in the classroom, which tends to be focused on the teacher and the purpose of the product. This learning activity was not match with the curriculum demands and the Science Learning purposes. The purpose of this research is to investigate the teachers’ ability in managing the learning activity of guided discovery, to identify both teacher and students’ activity in the learning activity and to know the students’ result of IPA learning activity. This research is a classroom action research by following the procedure of classroom action research. The subject of this research were elementary students on level five, Elementary School No. 59 South of Gorontalo City, year 2002/2003. Based on the data analysis and the conclusion of the research result, it was concluded: (1) the teachers’ ability in managing the learning activity of guided discovery was categorized well; (2) the result of data analysis on monitoring teachers and students activity on the learning activity was centered on the students; (3) the result of analysis Learning Result Test showed that the learning activity of guided discovery was effective to achieve the completed of learning purposes both the product and the process. By using the indicator: teachers’ ability in managing the learning activity, teacher and students’ movement in learning activity and the students’ learning result as a learning quality parameter. In addition, based on the data and the result of research data analysis, it can be concluded that the learning activity had been oriented on the approach of guided discovery can improve the quality of Elementary Science Learning.
