2. Test samples information
The following four (4) test samples for the Seized Materials test group were prepared by the Laboratory and
Scientific Section, UNODC:
Test Sample-1 (SM-1)
SM-1 was prepared from a seizure containing 13.6% (w/w) MDMA base.
Test Sample-2 (SM-2)
SM-2 was prepared from a sample of Caffeine and Lactose.
Test Sample-3 (SM-3)
SM-3 was prepared from a seizure containing 38.4% (w/w) Cocaine base. The test sample also contained traces
of benzoylecgonine, methylecgonine and tropacocaine.
Test Sample-4 (SM-4)
SM-4 was prepared from a seizure containing 21.1 % (w/w) Heroin base. The test sample also contained 6-
acetylmorphine, acetylcodeine, caffeine, narcotine, papaverine, paracetamol and trace amounts of morphine.
All test samples were prepared by preliminary grounding by mortar and pestle followed by homogenisation
using a centrifugal mill and a dry powder rotator. An aliquot (230 mg) of each test samples were weighed into
glass vials.
For each test sample, sufficient homogeneity was assured and stability studies were carried out in accordance
with methods outlined in ISO 13258:2015 “Statistical methods for use in proficiency testing by interlaboratory
comparisons” and “The international harmonized protocol for the proficiency testing of analytical chemistry
laboratories”1.

3. Introduction
An important element of the UNODC International Quality Assurance Programme (IQAP) is the
implementation of the International Collaborative Exercises (ICE). The exercises allow laboratories, from both
developing and developed countries to continuously monitor their performance in drug testing on a truly
global scale. This report provides information on analytical results of laboratories participating in the Seized
Materials (SM) test group. In order to maintain confidentiality, the participating laboratories have been
assigned random “Round Codes”, which change every round.
The analytical results returned by laboratories participating in ICE are evaluated by UNODC and a confidential
report is provided to each laboratory on its own performance. The overall analytical results are reviewed by
the UNODC’s International Panel of Forensic Experts which oversees the implementation of these exercises,
and offers guidance and support in addressing relevant quality issues. The exercises provide an overview of
the performance and capacity of participating laboratories and enable UNODC to tailor technical support in
the laboratory sector for greatest impact.
The ICE programme is a UNODC mandated activity and is implemented through regular budget funds and
through the UNODC Global Scientific and Forensic Programme – Support Project (GLOU54), which
operationalizes the forensic aspects of the UNODC Thematic Programme on Research, Trend Analysis and
Forensics". Figure 1 shows the growth of the programme in recent years for both the Seized Materials (SM)
and the Biological Specimens (BS) test groups.

4. Comments from the International Panel of Forensic Experts
4.1 Participation of Laboratories in ICE round 2019/2
In the 2019/2 round of the ICE programme, results were submitted within both the Seized Materials (SM)
and Biological Specimens (BS) test groups by 258 laboratories in 77 countries. Within the SM test group,
there were 213 participating laboratories from 67 countries and within the BS test group, results were
submitted by 104 laboratories from 48 countries.
4.2 Qualitative Analysis
Colour tests (54% of participants) were the most commonly used screening technique for test samples in
the SM test group, while GC-MS (92% of participants) was the most commonly used technique for
identification/confirmation of the components in the test samples followed by FTIR (41% of participants).
The results for the qualitative identification of the controlled substances in the SM test group, the number
of false positive/negative results and the analyses not performed are shown in the Table 1.

With regard to false negative results reported, there were a total of 5 (2 for SM-1, 1 for SM-2 and 2 for SM-
4). As for false positive results, there were a total of 19 false positive results (6 for SM-1, 3 for SM-2, 3 for
SM-3 and 7 for SM-4), which is a decrease in number compared to the previous round. Laboratories
reporting false positive or false negative results should investigate the reasons for this and corrective
actions should be taken as soon as possible in order to continuously improve performance. There were a
total of 10 analyses not performed, the majority of which were for SM-2. ICE participants should note that
test samples can contain any of the substances in the ICE menu and screening and identification carried
out should take this into account.

4.3 Quantitative Analysis
The number of participants who carried out quantitative analysis, 116 (54%) for SM-1, 137 (64%) for SM-3
and 123 (58%) for SM-4 continues to be encouraging. It is also encouraging that 141 (66%) participants in
the SM test group performed quantitation and of these, 128 (91%) quantified more than one substance
and 107 (76%) quantified all three test samples. It is understood that quantitative analysis of controlled
substances is often governed by local legislation and the requirements of prosecution. With regards to the
analytical techniques used by participants performing quantitation, 57% used GC-FID, 32% used
UPLC/HPLC-DAD and 20% used mass spectrometric methods (i.e. GC-MS, LC-MS, LC-MS-MS). z-Scores
obtained by participants in quantification are shown in Table 2 below.

According to the recommendations in ISO 13528:2015, an unacceptable z-score is considered to give an
action signal and a questionable z-score is considered to give a warning signal. A single action signal or
warning signals in two successive rounds shall be taken that an anomaly has occurred that requires
investigation. Participants with z-scores outside acceptable limits should review their quantification
procedures as soon as possible.
The following 15 laboratories obtained a single action signal or warning signal in the two successive rounds
of 2019/1 and 2019/2 and shall take this as an anomaly, which requires further investigation: 63AEKB,
64G4V7, 8IXSA9, 8SMOGU, AK3UYT, DTIACA, ELQO7L, ICNCMG, KPSSUB, MBWNRJ, MOGXMB, P8UQDW,
PKNKQN, TKLAER and V9FHUE.
Of the 22 laboratories that obtained a single action or warning signal in ICE rounds 2018/2 and 2019/1
reported in the previous summary report, 21 performed quantitation in 2019/2 and 9 of these improved
their performance in quantitation. The following 12 laboratories also obtained an action or warning signal
in 2019/2: 63AEKB, 64G4V7, 8IXSA9, AK3UYT, DTIACA, ELQO7L, ICNCMG, KPSSUB, MBWNRJ, PKNKQN,
TKLAER and V9FHUE.
Laboratories that need to perform quantitation routinely are encouraged to participate regularly in
external proficiency testing or collaborative exercises such as the ICE programme. This will enable
laboratories to assess the quality of their quantitative methods through the z-score values obtained. In
addition to the use of z-scores as a measure of performance in quantitation, participants are encouraged
to compare their results with the assigned values provided in individual quantification reports, to assess
the accuracy of their quantitative performance. Laboratories reporting false positive or false negative
results should investigate the root causes and corrective actions taken promptly in order to continuously
improve performance. Participation in the ICE programme also helps in monitoring the effect of corrective
actions.

5. New Psychoactive Substances (NPS) reported
During the 2019/2 round of ICE, participating laboratories provided information on NPS that had been
identified in their laboratories. In total there were 336 reports of 178 substances and the most
commonly reported substances were synthetic cathinones (31%) followed by synthetic cannabinoids
(26%) as shown in Figure 2.

In terms of the mode of action of the NPS reported by ICE participants, the predominant substances
reported were stimulants (35%), while 25% were synthetic cannabinoid receptor agonists (SCRA) and
19% were hallucinogens as shown in Figure 3.

The substances most often reported by ICE participants are listed in Table 3 below in descending order.
4F-MDMB-BINACA, 5F-MDMB-PICA and Etizolam were placed under international control at the 63rd
session of the Commission on Narcotic Drugs in March 2020 and these decisions will enter into force on
3rd November 2020.

Statement of findings for Test Sample 1
Code 3,4-Methylenedioxymetamfetamine (MDMA)
06JS66 + The sample was subjected to HPTLC using silicagelF254 as stationary phase and Ethylacetate:Methanol:Ammonia 85:10:5 as MP. A single spot was obtained after development. It was scanned and the UV Spectrum obtained was compared with the Library UV Spectrum and was found matching with MDMA and MDEA. The analysis was repeated using MDMA reference material. The RF and UV Spectra were found matching. Then the sample was subjected to GC-MS after subjecting to alkaline extraction using sodium hydroxide and ether. The ether layer is collected and ether evaporated at room temperature. The drug is dissolved in methanol and made upto 1ml. 100mg. of the sample is taken for extraction. GC-MS was performed using general method for basic drugs on HP5 column. MDMA reference material was also injected and the RT and Mass spectra compared and were found matching. It was also matched with library spectra and was found matching with MDMA. Therefore MDMA is confirmed in SM1. It is further subjected to Marquis test, Simons test and Galic acid test and gave positive results.

06JS66 + ANP The sample was subjected to HPTLC using silicagelF254 as stationary phase and Ethylacetate:Methanol:Ammonia 85:10:5 as MP.A single spot was obtained after development and it was scanned. The UV Spectrum was found matching with library UV spectrum of Caffeine and the analysis repeated along with Standard Caffeine and the RF and UV Spectrum were found matching. Then the sample was subjected to GC-MS analysis using HP5 column. The RT of SM2 and Standard Caffeine compared and were found matching. The Fragmentation pattern was compared with library and was found matching with Caffeine. Therefore the presence of Caffeine was confirmed.

Statement of findings for Test Sample 3

06JS66 + The sample was subjected to HPTLC using silicagelF254 as stationary phase and Ethylacetate:Methanol:Ammonia 85:10:5 as MP. A single spot was obtained after development. It was scanned and the UV Spectrum obtained was compared with the Library UV Spectrum and was found matching with Cocaine. The sample was re analysed along with Cocaine reference material using Methanol as the MP. The RF and UV spectra were matched and the presence of Cocaine is further confirmed by subjecting to GC-MS using general method for basic drugs on HP5 Column. The RT and Mas spectra of SM3 and reference material were compared and were found to be matching. The Mass spectrum was further compared with library spectrum and was found matching with cocaine. Scotts test was also performed and tested positive for Cocaine. 0GRT0S + cocaine was detected by GC-MS and quantified by GC-FID

Statement of findings for Test Sample 4
Code Caffeine Heroin Paracetamol (Acetaminophen)
06JS66 + + + The sample was subjected to HPTLC using silicagelF254 as stationary phase and Ethylacetate:Methanol:Ammonia 85:10:5 as MP. After development the chromoplate was scanned and the UV Spectrum obtained was compared with the Library UV Spectra.They were found to match with 6-MAM, DAM,Caffeine and Noscapine. There was anothter peak in the chromatogram which could not be identified which was very close to Noscapine. Therefore HPTLC was repeated with Chloroform and Ethanol 9:1 as the MP and was found to contain Paracetamol. It was repeated again along with Paracetamol and was found to be parcetamol after both RF and UV Spwxtra were found similar. For Confirmation the SM4 was subjected to GC-MS choosing general method for basic drugs on HP 5 column and Caffeine, 6-Acetylcodeine, 6-MAM,DAM, Paracetamol and Noscapine were found based on library spectra. A small peak corresponding to PAPAverine was also noticed but ignored as it is too small to confirm. Then Standard Caffeine, DAM, Paracetamol and Noscapine reference material were injected into the system and RT and Mass spectral fragmentation compared and were found to be similar. Thus the presence of DAM, Paracetamol, Caffeine and Noscapine were confirmed. 6-MAM and 6-Acetyl Codeine were also confirmed based on library spectrum. RT could not be compared for these drugs due to non availability of reference standards. Further the sample is subjected to Marquis test and tested positive for Opiates. 0GRT0S + + + Heroin was detected by GC-MS and quantified by GC-FID
