What is budget?

• A budget ensures that sufficient resources (financial, personnel, and infrastructure /equipment) are available to complete the research work that is being proposed.

• It should be based on a full analysis of research project costs, and aim to get the sponsor to pay for as many of those costs as possible, while ensuring that both the sponsoring agency and Institutional guidelines are followed.

Justification for the proposed equipment• The Department of Biochemistry, University College of Medical

Sciences, Delhi, where the proposed research project work will be executed, has all the necessary equipments as mentioned in “facilities/equipments being extended by parent institution” except the above mentioned equipment.

• A Real time PCR machine (Q-PCR), critical for quantifying gene expression patterns, is essential for achieving the objectives.

• The department is initiating cell / tissue culture facility and have all the other equipments such as Laminer flow, CO2 incubator except an inverted microscope with a digital camera attached to a computer, for detailed image analysis.

• We will also require liquid nitrogen containers that will be used for storing the cell lines.

• A dedicated deep freezer will be essential for storing tissue culture material, temperature sensitive reagents and samples for the project as the existing freezers are overloaded.

Justification for the manpower requirement

(i) Junior Research Fellow (One) : The experiments and estimations planned in the proposed research project will be carried out by the investigators themselves / their post-graduate students. However, to assist in the basic work like isolation of cells / DNA, preparation of reagents, sample collection and processing and data computation, a skilled JRF is required. Therefore, we propose to recruit one JRF with M.Sc./M.Tech degree in Biochemistry/Life Sciences/Medical Laboratory Technology/ Biotechnology (NET qualified) with at least one year work experience in a Biochemistry/Molecular biology laboratory and in-hand experience in molecular biology techniques; pay as per DST rules.

(ii) Laboratory attendant : The proposed project involves collection of large number of human samples. Transportation of samples from O.P.D. to lab, processing, storage, analysis, and disposal of samples, assisting tissue culture procedures like washing of plasticware and glassware, autoclaving and cleaning of lab accessories demands a full time helping hand. Therefore, we felt the need of a laboratory attendant with some work experience in laboratory work; consolidated pay as per DST rules.

Justification for consumables The budget for biological reagents and chemicals constitute a major

expenditure for this project. Many of the reagents are not manufactured in India and have to be imported either directly or through their distributors. We will require reagents for

(i) Gas chromatography (GC) based analysis, that include HPLC grade solvents, high quality nitrogen gas and standerds and GC vials. ECD column used for GC can cater to approximately 300 – 400 samples. As we will process more than seven hundred samples, we will require two such ECD columns

(ii) Molecular biology work with special requirements for Real time PCR (Taqman master mix and real time tubes), reagents / kits for DNA and RNA isolation and PCR based reagents such as dNTPs, restriction enzymes, Taq polymerase for gene polymorphism studies

(iii) Cell culture. For in vitro work, two cell lines will be commercially procured.

(iv) As Real time PCR and cell culture are sensitive areas of research, we will require at least two sets of dedicated autopipettes exclusively for each area.

Title of the Project : Association of Common Variants of Inflammatory Cytokine Genes with Risk of Nephropathy in Type 2 Diabetes among Asian Indians

Objective : Eight single nucleotide polymorphisms (SNPs) of

pro-inflammatory cytokine genes (CCL2, TGFB1, IL8, CCR5, and MMP9) were to be genotyped.

Sample : Two independently ascertained type 2 diabetic

cohorts with nephropathy (n=200), without nephropathy (n=200) and control ( n=200).
