PREFACE Thank you very much for your purchase of Dynamica HALO DB-20 Spectrophotometer. This instruction manual has been prepared for users of the DB-20. It describes how to handle the Spectrophotometer. The HALO DB-20 Spectrophotometer is intended for the purpose of photometric analysis. Do not use it for any other purpose. The HALO DB-20 is designed for use by persons having a basic knowledge of chemical analysis. Other persons should operate the instrument in the presence of someone who has such basic knowledge. Remember that improper use of the instrument, chemicals or samples would result not only in wrong analytical data but in consequences adverse to safety. Before using the instrument, be sure to read through this instruction manual to enable correct use. For installation/maintenance of the instrument, read the instruction manual carefully to attain a full understanding of the instructions. Be sure to observe cautionary instructions. Keep this manual handy for easy reference when necessary. Information contained in this manual is subject to change without notice for product improvement.

ABOUT THIS MANUAL

Before using the instrument, be sure to read through this instruction manual. First of all, read “SAFETY SUMMARY” included at the beginning of this manual for ensuring safety in operation of the spectrophotometer main unit.

IMPORTANT Precautions on Electromagnetic Wave Interference 1. Possible Electromagnetic Wave Interference Caused by This Instrument

When this instrument is used in a residential area or an adjacent area thereto, it may cause interference to radio and television reception. To prevent this, use the specified system connection cables in strict accordance with the instruction manual. The instrument is designed to minimize possible electromagnetic wave interference caused by it if the specified cables are connected properly. However, there is no guarantee that electromagnetic wave interference will not be caused by the instrument. If the instrument does cause interference to radio or television reception, which can be determined by turning off and on the instrument, the user is encouraged to try to correct the interference by one or more of the following measures: • Increase separation between the instrument and radio/TV

receiver. • Connect the instrument to an outlet on a circuit different from

that to which the radio/TV receiver is connected. 2. Possible Electromagnetic Wave Interference Affecting This Instrument

If this instrument is used near an intense electromagnetic source, interference noise may be given to the instrument to incur an adverse effect on its performance or functionality. To prevent this, use the specified system connection cables in strict accordance with the instruction manual. The instrument is designed to minimize possible electromagnetic wave interference affecting it if the specified cables are connected properly. However, there is no guarantee that electromagnetic wave interference will not occur in this instrument. If the instrument does incur electromagnetic wave interference, which can be determined by turning on and off possible sources of electromagnetic wave interference nearby, the user is encouraged to try to correct the interference by one or more of the following measures:

• Reorient the instrument. • Increase separation between the instrument and possible

sources of electromagnetic wave interference. • Increase separation between the power cable of the instrument

and possible sources of electromagnetic wave interference. • Connect the instrument to an outlet on a circuit different from

that to which possible sources of electromagnetic wave interference are connected.

• Confirm that any other device connected with the instrument is not affected by electromagnetic wave interference.

Warranty on Product

The Model HALO DB-20 Spectrophotometer is warranted to operate according to the specifications given in the instruction manual, provided it is used in accordance with the instructions described in the manual. (1) Scope of Warranty

(a) Any parts which prove to be defective in design or workmanship during the warranty period will be repaired without charge.

(b) A substitute part may be used for repair, or replacement

with an equivalent product may be made instead of repair. (c) Such system components as a personal computer and

printer to be updated frequently for improvement may not be available in original versions at the time of replacement.

(2) Warranty Period

One year from the date of initial installation. Consumables such as WI and D2 lamps are excluded from warranty.

(3) Availability of Technical Support Service

Technical support service for this instrument is available within regular working hours on workdays predetermined by us.

(4) Limitations and Exclusions on Warranty Note that this warranty is void in the following cases, even if they occur within the warranty period.

(a) Failure due to operation at a place not meeting the

installation requirements specified by us (b) Failure due to power supply voltage/frequency other than

specified by us or due to abnormality in power supply (c) Corrosion or deterioration of the tubing due to impurities

contained in reagent, gas, air or cooling water supplied by the user

(d) Corrosion of the electric circuits or deterioration of the

optical elements due to highly corrosive atmospheric gas (e) Failure due to use of hardware, software or spare parts

other than supplied by us (f) Failure due to improper handling or maintenance by the

user (g) Failure due to maintenance or repair by a service agent

not approved or authorized by us (h) After disposal of this instrument, or after its resale without

our approval (i) Failure due to relocation or transport after initial

installation (j) Failure due to disassembly, modification or relocation not

approved by us (k) Consumables, and failure of parts that have reached the

end of specified useful life (l) Failure of parts excluded from the warranty in the

instruction manual or other documents (m) Failure due to acts of God, including fire, earthquake,

storm, lightning, social disturbance, riot, crime, insurrection, war (declared or undeclared), radioactive pollution, contamination with harmful substances, etc.

(n) Failure of the hardware, or damage to the system software, application software, data or hard disk due to computer virus infection

(o) Failure of the personal computer connected with the

instrument, or damage to the system software, application software, data or hard disk due to power interruption or momentary power voltage drop caused by lightning or the like

(p) Failure of the personal computer connected with the

instrument, or damage to the system software, application software, data or hard disk due to disconnection of main power to the personal computer without taking the specified normal shutdown procedure

(5) Disclaimer of Warranty

(a) Any express warranties other than the explicit conditions

indicated in (1) are excluded from this warranty. Any other implied warranties of merchantability and fitness for a particular purpose are not included in this warranty. No liability is assumed for direct or indirect damages arising out of explicit or implied warranties.

(b) Oral or written information or advice given by our dealers,

distributors, agents or employees without our express permission shall not create a warranty or in any way increase the scope of this warranty.

Technical Service Installation of this instrument shall be carried out by or under supervision of qualified service personnel of Dynamica or its authorized service agent. Before installation of the instrument, the user shall make preparations for satisfying the installation requirements in accordance with the instruction manual. If relocation of the instrument becomes necessary after initial installation (delivery), please notify your local sales representative or nearest service office of Dynamica.

When you discard equipment, please check and discard a related statute etc. or ask the service section of Dynamica.

Other Precautions 1. Handling of Chemicals and Samples

(1) The user is responsible for following relevant legal standards

and regulations in handling, storage and discarding of chemicals and samples used in analytical operations of this instrument.

(2) Reagents, standard solutions and accuracy-control samples

shall be handled, stored and discarded as instructed by the respective suppliers.

Before installation, confirm that there are no missing items or standard accessories. If there is something missing or damaged, or you have noticed any problems, please contact our nearest representative.

Operating the equipment without a standard part can damage the

equipment and cause safety concerns. If that occurs, please follow the instruction of the installer.

Installation of this instrument shall be carried out by or under

supervisions of qualified service personnel of Dynamica or its authorized service agent.

When relocation of this instrument becomes necessary after initial

installation (delivery), please notify your local Dynamica sales representative or nearest Dynamica service office. Technical support service for this instrument is available from s service agent approved or authorized by Dynamica within regular working hours or workdays.

Please do not perform any other operations that are not included in

the manual. If any problem occurs with the equipment, please contact the agent from whom you purchased it or the service department of Dynamica.

The organic solvent vapor may be harmful to your health.

Please wear protective glasses when using organic solvents. If the organic solvent should get into your eye, flush your eye immediately under running water for at least 5 minutes while keeping your eyelids open. See a physician for appropriate treatment.

WARNING: Electrical Shocks from Improper Grounding Electrical Shock from Improper Grounding

When wiring the personal computer, power supply for the thermostatic cell holder and the like, please make sure to use the 3-prong wire (with ground) provided.

WARNING: Electrical Shock Electrical Shock from Contact

High voltage is used inside of the equipment. Never remove the cover except by a service person or as instructed in the section for lamp replacement.

When cleaning the inside of the cell compartment, be sure to turn off the

unit.

CAUTION: High Temperature Burns from high temperatures

The lamp will become very hot during operation, please make sure that the D2 lamp and the tungsten lamp is cooled off when replacing the lamp.

Voltage supplied to the DB-20 UV-Vis Spectrophotometer. The voltage for the Spectrophotometer system and personal computer must be a single-phase AC 100V to 200V; Variations in the voltage and noise generated in the power line will cause adverse effects on the UV-Vis spectrophotometer and may also cause accidents.

Please make sure that grounding is provided together with the power

supply wires, and make sure that it is connected with a grounding resistance of less than 10 Ω. Defective grounding may not only cause lower resistance against noise from the outside but it can also cause the Mass Spectrometer and personal computer to generate static electricity, which may involve the danger of electrical shocks.

A high voltage circuit is used inside the UV-Vis Spectrophotometer. Do

not open the covers when this circuit is operational because of the danger from electrical shock.

Fire Extinguishers

Do not smoke or use fire within 3 meters of the UV-Vis Spectrophotometer

Make sure to keep a fire extinguisher near the UV-Vis Spectrophotometer

at all times. Obtain an ABC Powder extinguisher that can be used for normal fires, oil fires, and electrical fires.

When turning off the power to the personal computer while the hard disk, floppy disk, or MO drive (optional) is still running, the computer may malfunction or the data and software can be destroyed.

Computer Viruses and Backup Data

If any program/data is damaged suddenly or an unexpected operation/screen is encountered, the personal computer may be infected by a computer virus. Computer viruses are malicious programs that sneak into the personal computer to cause misbehavior or damage to data. A program designed to offer protection against computer viruses is called a vaccine program (virus scan). Do not use any programs or storage media that are suspected of containing a virus. If there is a possibility of virus infection, check for a virus using a vaccine program. It is very important that you make a backup of your hard disk just in case.

Chapter 1 Installation

1.1 Unpacking

After unpacking the shipping crate, carefully take out the spectrophotometer and place it on a table. Since the spectrophotometer is as heavy as about 29 kg, special attention should be paid when taking it out.

1.2 Installation site

1.2.1 Installation conditions

Power requirements: Power supply voltage: AC100, 115, 220, 230 or 240V (There must be the

devices for switching voltage.) Allowable fluctuation is within ±10% of rated voltage. Frequency: 50 or 60 Hz

Allowable fluctuation is within ±4% of rated frequency. Power capacity: 200VA or more.

Prepare a capacity more than 500VA in reference with a co-use with other accessory devices.

Note: If warning labels are not readable because of stains on coming off, please notify local Dynamica sales representative or service office. New labels will be provided.

Grounding line: Grounding line resistance of 100Ω or less is required.

Place for Installation Minimum floor area: Floor area with a width of 550 mm and a depth of 710

mm or more is required. Floor strength: Floor strength 30kg or more.

Select a rigid and horizontal plane capable of withstanding the above load as an installation area.

1.2.2 Installation Environment

Operating temperature: 5~35℃. In order to perform a measurement under the most stable condition, we recommend that the instrument is used in an air conditioning room of 20℃ ~ 25℃. Operating humidity: 45% ~ 85%.

Atmosphere, gas:

Free from acid gas or alkaline gas and other gases that may corrode metals significantly. Free from gases that may dissolve paint such as from organic solvents (particularly benzene, thinner or the like).

Other general notes:

Avoid direct sunlight (otherwise optical performance might deteriorate or housing might be discolored). Avoid a window as much as possible. A strong vibration or shock that can be felt by the human body must not

be transmitted to the instrument (otherwise the fine adjustment mechanism might malfunction). Avoid the vicinity of a heat generating apparatus such as a gas burner,

electric heater or oven in order to prevent the mainframe cover being heated (beyond 70℃). Avoid the vicinity of instruments that generate a strong electric field

(such as an electric welding machine, high frequency furnace or pole transformer). Avoid a dusty environment (otherwise optical performance might

deteriorate). The line voltage must be stable and free from a rapid fluctuation

(otherwise noise might increase). Do not turn on and off frequently electric instruments (stirrer, vibrator,

etc) that are not provide with a noise-preventive device in the same power line as for the mainframe.

Note: The optical system is very delicate. And the control unit incorporates high-density electronic circuit components that function as a computer. So the above cautions should be observed strictly.

Plug the power cord securely into the connector of the mainframe. When using a plug adapter of table tap, be sure its grounding wire to the grounding terminal securely.

1.7 Connection of Printer

Connect the printer with the instrument if to use the printer with a standard parallel printer cable. The connector of printer is on the right side of the

instrument as show in Fig 1-3. DB-20 is compatible with Seiko thermal printer, Hewlett Packard printer with PCL3 setting, Epson Printer with ESC/P setting, Seiko Printer with DPU setting. For a detail list of the printer that is compatible with the DB-20, contact your local representative of Dynamica. To define the printer type connected to the DB-20, please refer to the System setting section interface under printer setting sub menu. Please refer to the relevant instruction manual of printer to install the printer.

1.8 Local-Remote switch

DB-20 can be controlled by the optional PC software, or simply via keypad control. Please put the Local-Remote switch to Local when using keypad and LCD on the DB-20. When connecting to the PC software, switch the Local-Remote switch to Remote position. The LCD will display < PC Control > when connected to PC software for complete instrumentation control and data analysis.

1.9 LCD Brightness adjustment port

Using a small minus screwdriver, the LCD brightness can be adjusted through this port by turning the screw.

1.10 Connecting Accessory to DB-20

The Acc port is for the connection and control of a variety of optional accessories available from Dynamica such as Auto Sipper, Electronic Thermostatic cell holder. Connect the optional accessories using the cable provided along with the optional accessories. Please refer to the installation manual for detail for installation of optional accessories.

1.11 Connecting DB-20 to PC Software

Optional PC software allows DB-20 to be fully controlled by the software and perform data analysis using the PC software. Connect the serial cable provided with PC software to the port marked PC. Please also set the Local-Remote switch to Remote when using PC software.

1.12 Connecting USB Flash Memory

DB-20 is equipped with a USB port to connect commercial available USB flash memory for the storage of data generated by the DB-20. The data generated in the DB-20 can save into USB flash memory with File Manager of DB-20. Refer to the relevant chapter for operation. The data can be transferred to standard computer for storage also. Simply insert the USB flash memory disk into the port. USB flash memory of Kinston Data Traveller ( Compatible to USB1.1 and FAT file system ) is tested to be compatible. Contact your Dynamica sales agent to obtain the latest list of the compatible Flash Memory.

Chapter 2 Function

2.1 Application

The HALO DB-20 Spectrophotometer is designed for absorption analyses of liquid, solid and gaseous samples in ultraviolet and visible spectral regions. In this scheme (Fig.2-1), a monochromatic beam with intensity ‘I0’ travels through a liquid phase having concentration ‘c’ and path length ‘l’ , which results in the intensity of monochromatic radiation decreasing to ‘It’. With respect to the initial intensity ‘I0’ and attenuated intensity ‘It’ of monochromatic radiation, the following equation (1) can be set up. It/I0 = 10-ε·c·l =τ……………………………………………………….(1) Where, ‘ε’ is a constant known as absorptivity, which varies depending on the sample. Equation (1) represents the statement of Lambert – Beer law, and ‘τ’ indicates transmittance which is often expressed as a percentage, i.e. ‘τ× 100 = T’ (percent transmittance or %T). Also, the common logarithm of inverse transmittance can be expressed as follows: Log (l/τ) = ε·c·l = E …………………………………………………..(2) Where ‘E’ is called absorbance (abbreviated as Abs). Absorbance ‘E’ is proportional to concentration ‘c’ and is a unit of measurement indispensable for quantitative determination.

2.2 Principle of Operation

2.2.1 Optical System

DB-20 is a double beam system, which features a half mirror to split the beam to reference beam and sample beam. The white light emitted from the light source is fed to the Seya-Namioka monochromator utilizing a concave diffraction grating (with a grating constant of 1/600 mm, a blaze wavelength of 250nm and a grating area of 20mm × 25mm), where it is transformed into a monochromatic beam. The beam sent from the monochromator is reflected by the toroidal mirror (M2), and then separated into reference beam and sample beam by the half mirror (HM). The two beams have passed through the sample compartment are focused by lenses, and are then irradiated into the detector (D1 and D2) where they are converted into electric signals. Listed below are the features of this optical system. The Seya-Namioka optical system utilizing Dynamica’s reputable concave diffraction grating realizes a monochromator featuring high-energy efficiency and low stray light level. The beam is separated by using a half mirror rather than a rotary mirror like in the conventional double beam spectrophotometer. This structure serves to eliminate the moving parts, thus realizing a highly stable and reliable monochromator. The optical systems on the sample and reference beam sides are absolutely the same to enable acquiring highly stable data.

2.2.2 Signal Processing and Control System

The electric signal converted from optical signal enters the LOG amplifier where it is LOG converted to provide absorbance data. This data is amplified, and is then A/D converted to be processed as digital value. The processed results are displayed on the LCD and recorded onto the printer.

2.3 Proper Use of Spectrophotometer

2.3.1 Solvent Selection

When selecting a solvent for sample preparation, keep the following requirements in mind. Small absorption in the spectral region of measurement wavelength. Non – interactive with solute.

The HALO DB-20 is capable of calculating the concentration of a sample after generating a standard calibration curve or by entering a factor into the system. The following are the features of this function. (1) Generation of standard calibration Curve

(a) Up to twenty samples can be measured, and the acquired values are connected to each other with segmented lines or regression lines determined by the least squares method for generating a working curve.

(b) Specific standard samples can be measured again to regenerate a working curve.

(c) A working curve can be regenerated upon deleting specific standard samples.

(d) Working curves can be stored as a part of measuring conditions. It is possible to store the curves for a maximum of twenty tests.

(e) Working curve can be graphically displayed on the LCD, and a value at a desired point can be read by moving the cursor to that point.

(f) Analytical results are displayed on the LCD together with the working curve. Thus, the results can be confirmed while observing the working curve.

(g) Working curve can be graphically plotted on the printer. (h) Working curve can be generated by entering factor values. (i) It is possible to generate a working curve by 2 - /3 – wavelength

measurement.

(2) Statistical Calculation This function enables the instrument to acquire mean value, standard deviation (SD) and coefficient of variation (CV).

(3) The upper and lower limit values of analytical data can be judged.

2.4.2 Wavelength Scan

(1) The highest wavelength scan speed is 3600nm/min. Wavelength ranging from 1100~190nm can be scanned in a little over 20 seconds even if the filter and light source are switched over halfway.

(2) Wavelengths can be scanned repeatedly for recording spectra of a sample

that changes with time. (3) Photometric values can be printed out at the specified wavelength interval.

(4) Photometric values can also be displayed on the LCD. (5) Graphic display on the LCD can be output onto the printer. It takes a little

over 10 seconds to make a hard copy for the graphic display on the LCD.

2.4.3 Time Scan

(1) A graph can be displayed with time graduated on the abscissa. (2) Photometric values can be printed out at the specified time interval. (3) Data can be calculated for right assay.

2.4.4 Secondary Processing Functions

The HALO DB-20 can store in the memory the measured spectrum and its change with time. Secondary processing can be applied variously to the stored data. Automatic Data Readout

A cursor can be moved to a desired point in the spectrum currently displayed on the LCD screen and a reading of the absorbance (transmittance) at that point.

Peak Detection

Cursor can be moved to a peak position to display absorbance (transmittance) readings at the peak. In addition, an automatic peak selection routine allows peak and valley measurements to be printed in list format on the printer.

Scale Expansion

Three methods are available for expanding the graphic display on the LCD.

(a) Simultaneous Expansion of X and Y Axes Cursor should be moved for pointing out a desired expansion range.

(b) Expansion of X Axis Numeric values should be input to specify a desired expansion range for the abscissa.

(c) Expansion of Y Axis

Numeric values should be input to specify a desired expansion range for the ordinate.

Smoothing

A spectrum with a large noise level can be corrected to form a smooth curve. Differentiation

The data of a spectrum currently displayed on the LCD are differentiated. The derivative order applicable ranges 1.

Integration

A peak area can be determined by specifying the two wavelengths or times between which you wish the integration calculated. The DB-20 then integrates the area under the spectral curve between the specified points and points out the results.

Calculations Between Spectra

You can calculate addition, subtraction, multiplication or division between two spectra or calculate a constant value with the resulting data displayed on the LCD.

Recalculation During Rate Assay (Kinetics)

Upon completion of a rate assay in the Time Scan mode, you can adjust the calculation start time and end time to allow recalculation of the data.

2.4.5 Data Storage Function

The following data are stored in the memory, and are not deleted even if the instrument power is turned off. Analytical conditions for 20 tests

The working curve prepared for each test can also be stored together with the analytical conditions.

Up to 10 measurement results in inside memory. You also can store the measurement results in flash memory which can be open via personal computer.

Three baselines (System baseline and two user-defined baseline.)

2.4.6 Automatic Start Function

When the automatic start function is set in advance, the operator need only turn on the power switch (measuring conditions are set automatically), set a sample in the instrument and then press the START key. The rest is done automatically.

Chapter 3 Operational method

3.1 Basic Operation

3.1.1 Operational Procedure

The user interface of the HALO DB-20 is convenient and easy-to-use. Screen prompts help you to select the variables that are required for method set-up and data acquisition.

3.1.2 Key-Panel Functions

The illustration below shows the operation panel of the HALO DB-20. There three groups to touch keys on the panel: Special function Numerical Control

Key Functions The function of each key on the HALO DB-20 key panel is described below:

Main Menu List the six data processing functions and retrieves the initial main menu frame. Clear Return This key acts as a backspace key to erase the last character entered or acts as a return key to the previous screen display. Left Arrow Moves the trace left or the page up.

Right Arrow Moves the trace right or the page down. Up Arrow Moves the highlight or the page up. Down Arrow Moves the highlight or the page down. 3.2 Preliminary Operation

3.2.1 Initialization

Upon turning on the power switch, the INITIALIZATION frame appears on the display and the instrument performs a self-diagnosis and automatic wavelength calibration. During self-diagnosis your HALO DB-20 spectrophotometer checks the following items.

As each item is checked, the message “OK” will display if the item’s function is normal or the message “NG” will display if the item’s function is abnormal.

CONSULT THE TROUBLESHOOTING SECTION IF YOU FIND ERROR MESSAGE DURING INITIALIZATION. Error Message: 1. If the wavelength driver is NG, the system will freeze. And you must power

off the instrument to solve the problem. 2. If the other items are error, the instrument can go on run by pressing

Clear/Return. Or else the system will stop.

3.2.2 Main Menu

After initialization, the LCD will enter into the interface which last interface before power off. Press Main Manu key and the Main Menu screen appears on the display as shown in the below. The Main Menu lists seven options plus Wavelength and photometric value displayed on the top of the screen. To guide you through each procedure, there is a prompt displayed at the bottom of each screen page. The function of each option item refers to chapter 2.

In which 1st order means linear; 2nd order means quadratic regression curve; segment means polygonal line; K-factor means k-factor input line. If select “1st order”, the standards number should be more than 2. If select “Second order”, the standards number should be more than 3. If select “K-factor”, then you can input the value of k0, k1 and k2.

Note: If the standards number is not fit the curve type, the system will automatically changed the numbers to fit the curve type.

Curve Data: Input the standards concentration.

Press <3> key on the keypad to input the Curve Data. The data list will appear on the right side of the screen as follows. Left and right arrow key are used to select the ID of Standards (STDs).

Lamp Change Mode:

The followings three Lamp Change Mode are available. Auto: Changeover is automatically made between D2 Lamp and WI Lamp. D2 Lamp Only: Using D2 lamp over the entire wavelength region. WI Lamp Only: Using WI lamp over the entire wavelength region.

Lamp Change WL (nm):

This parameter is used for selection of light source switching wavelength. This spectrophotometer is provided with WI lamp for visible range measurement and D2 lamp for ultraviolet range measurement. The wavelength is selectable with a range of 325.0 ~ 370.0 nm in increments of 0.1 nm.

WI Lamp: Turn on or turn off the WI lamp.

D2 Lamp: Turn on or turn off the D2 lamp.

Note: If turn on the lamp but display “OFF”, perhaps the lamp is error.

If the memory location that you have selected is available, the instrument stores entire variable set under test name you have specified. If the catalog number you requested is already full, the following message appears: Overwrite this File? To delete the previous test variables and store the new variable set, press ENTER. To leave the old variables set intact and store new variables in a different location, press RETURN.

3.3.2 Performing Photometric Measurements

Once the appropriate operating conditions are selected, the system is ready for measurement. 1. “%T” or “ABS” selected for Data Mode Press <0> on the keyboard and the variable selection screen will change to the measurement screen and the following message will appear on the screen: Insert SAMP and Press Start to Measure: Press Autozero on the keyboard to begin an autozero first, and then insert the sample and press START key to begin a photometric measurement. Printer Type There are three kinds of printer to select: EPSON, Seiko, HP.

Auto Print If the autoprint function is “ON”, the DB-20 will print automatically the setting parameter and the graph or data after completion of a measurement. But if the autoprint function is “OFF”, the instrument will not print automatically unless the Print key is pressed.

Graph Print Using this parameter to determine whether to output the on-screen spectral graph onto the printer automatically at the end of wavelength scanning.

Data Print If it is desired to print out photometric values at a fixed wavelength interval during spectral scanning, set the Text Print parameter to Peak, Valley or All Data. Otherwise, set this parameter to Not Print.

List Interval (nm) If the Text Print parameter is set to ON, specify a wavelength interval at which photometric values are printed out. This parameter is specifiable within a range of 0.1 to 100.0 nm in increments of 0.1 nm.( Please refer to section 3.4.4 )

Lamp Change Mode: The followings three Lamp Change Mode are available. Auto: Changeover is automatically made between D2 Lamp and WI Lamp. D2 Lamp Only: Using D2 lamp over the entire wavelength region. WI Lamp Only: Using WI lamp over the entire wavelength region.

Lamp Change WL (nm): This parameter is used for selection of light source switching wavelength. This spectrophotometer is provided with WI lamp for visible range measurement and D2 lamp for ultraviolet range measurement. The wavelength is selectable with a range of 325.0 ~ 370.0 nm in increments of 0.1 nm.

WI Lamp: Turn on or turn off the WI lamp. D2 Lamp: Turn on or turn off the D2 lamp.

Peak Threshold:

Threshold function is used to determine the level of signal changes required to be identified as peak or valley. The range of threshold is 0.1 ~ 100.0 %T(E) or 0.001 ~ 1.000 ABS.

Baseline: Choose the kind of baseline to use for measurement. The following four kinds of baselines are selectable: a) System (auto calibration from 190 to 1100 nm)

Response: There are three response speeds to select: Fast, Medium and Slow. In the ordinary measurement, select “Medium”. If it is desired to reduce variation, choose the slow response parameter. If it is required carry out high-resolution measurement, select “Fast” response parameter.

Init Delay: Specify an initial delay time where it is required to wait until the temperature becomes stable for sample measurement or the sample sipper is employed. In an ordinary measurement application, set an initial delay time to ‘0’ (zero). This parameter is specifiable within a range of 0 to 9999 seconds in increments of one second.

The wavelength range of the User 1 and User 2 is specified under the parameter Setup. The user baseline measurement is conducted under the currently defined conditions. Therefore, efficient correction can be made using the identical conditions (wavelength range, scan speed, data read interval, etc.) to those of actual measurement. Note: If the baseline is out of baseline range you selected or there is not

baseline stored in memory, such hint will appear on the bottom of screen.

3.4.2 Performing a Wavelength Measurement

Upon completion of measurement conditioning setting, press the <0> key and the measurement screen appears on the LCD. When the conditioning screen is switched over to the measurement screen, the instrument checks the user-specified parameters. If any conditional inconsistency is found, the relevant error message is displayed at the bottom of the screen. Also, if the currently wavelength does not match the start wavelength, it is forced to shift to the level of start wavelength. Press Autozero key on the keyboard to execute an auto zero. Then set a sample and press the START key to perform wavelength scanning and the spectrum will be displayed on the screen. To abort a measurement during operation, press the STOP key. Upon completion of measurement, the following spectral plot appears on the LCD. If the Auto Print and Graph parameter is preset to “On”, the on-screen spectral plot is output to the printer if connected. If the printer does not print the graph correctly, check the printer setup menu to make sure that the correct printer type is selected.

3.4.3 Secondary Processing

The measured spectrum is stored in internal memory. Several kinds of computational processing can be performed for the stored spectrum. The following explains these secondary-processing operations. Note: The second processing is effective only for the spectral data measured last. For the overlaid spectral attained through repetitive scanning, only the spectral data measured in last cycle can be handled in the secondary processing. (a) Trace The Trace processing function allows the user to read out values of the on-screen spectrum by means of the crosshair “+” cursor. On completion of measurement, press <1> on the keyboard and then the trace screen appears as shown below.

When the crosshair (+) cursor appears on the spectral graph, it can be moved left/right using the arrow keys. The wavelength range that the cursor moved each time is according to Wavelength Range on Interval. Holding the arrow key down causes this cursor to move quickly. The wavelength and photometric value pointed by the cursor are read out and indicated at the bottom of screen.

The Peaks processing function allows the user to detect spectral peaks. On completion of measurement, press <2> and then the peaks screen is presented as shown below.

The Deriv function allows the user to obtain a derivative spectral graph. DB-20 can displays 1st through 4th order derivate spectrum.

Area

This function is used to carry out area calculation. Specify the wavelength range for area calculation, and this function will perform integration. When the spectral wavelength Start wavelength and End wavelength are specified, the Area function calculates the following area values.

There are three kinds of printer to select: EPSON, Seiko, HP. Auto Print

If the Auto print function is “ON”, the DB-20 will print automatically the setting parameter and the graph or data after completion of a measurement. But if the Auto print function is “OFF”, the instrument will not print automatically.

Graph Print Using this parameter to determine whether to output the on-screen spectral graph onto the printer automatically at the end of wavelength scanning.

Data Print If it is desired to print out photometric values at a fixed wavelength interval during spectral scanning, set the Text Print parameter to Peak, Valley or All Data. Otherwise, set this parameter to Not Print.

List Interval (nm) If the Text Print parameter is set to ON, specify a wavelength interval at which photometric values are printed out. This parameter is specifiable within a range of 0.1 to 100.0 nm in increments of 0.1 nm.

If any conditional inconsistency is found, the relevant error message is displayed at the bottom of the screen. Also, if the currently specified wavelength does not match the measurement wavelength, it is forced to shift to the level of measurement wavelength. Press Autozero key on the keyboard to execute an auto zero. Then set a sample and press the START key. Thus, time scanning is carried out and the spectrum is displayed on the screen. To abort measurement prematurely, press the STOP key. On completion of measurement, the following spectral plot appears on the LCD. If the Auto Print and Graph Print parameter is preset to “ON”, the on-screen spectral plot is output onto the printer also.

3.5.3 Secondary Processing

The measured spectrum is stored in internal memory. Several kinds of computational processing can be performed for the stored spectrum. The following explains these secondary-processing operations. 3.6.2 Performing Multiple WL Measurement

Creating a Standard Curve After setting the appropriate operating conditions, use the following steps to perform multiple wavelength measurements. 1. Press <0> on the keyboard. The condition-setting screen will switch to the measurement screen. 2. Press <Autozero> to start an autozero measurement. 3. Insert the first standard sample in the sample compartment. Then press Start to start measurement. Repeat the step, as many times as are required to analyze the number of standards after completing analysis of standard samples, the measurement screen will change to following screen.

DB-20 will perform wavelength scan from 660.0 to 650.0nm as in the WL Accuracy test in the first stage. The maximum value of wavelength will be located, then the motor moves from 650.0 to 190.0nm, and then from 190.0 to 660.0nm. In second stage, DB-20 will scan from 660.0-650.0nm again; find another wavelength position at the same time. In the third stage, DB-20 will moves from 650.0-1100.0nm, from 1100.0-660.0nm and perform a wavelength scan the third time and mark the third wavelength peak position. From the three values recorded in the 3 stages, DB-20 will obtain the maximum wavelength value and the minimum wavelength value, taking the mean of the two values i.e. (max-min)/2. If the result is within 0.1nm, the WL Reproducibility item will display <OK>.

3. <Bandpass> DB-20 perform a wavelength scan from 660.0 to 650.0nm as in WL Accuracy test. From the measured data, DB-20 identified the maximum value and the minimum value and taking the mean of the absorbance value i.e. A/2= (Max-Min)/2. At A/2 Abs value, two wavelength values that are close to A/2 can be identified. The difference between these two wavelengths is the value of band pass. If the bandpass value is within 1.5nm, the bandpass item will display <OK>.

4. <Noise Level>

After the time scan of 300S at 500.0nm, DB-20 will identify the maximum value and the minimum value of the measured data. The noise level value is the difference between the maximum value and the minimum value (peak to peak). If the noise level value is within the range of 0.001ABS, the item will display <OK>.

5. <Baseline Stability> After the time scans of 3600S at 500.0nm, the peak-to-peak value of the wavelength scan is defined as baseline stability value. If this value is within the range of 0.001ABS, this item will display <OK>.

6. <Baseline Flatness> DB-20 will perform a wavelength scan from 950.0nm to 200.0nm twice. The first time the measured data is recorded as the baseline, and the second group of data is defined as absorbance. The maximum value and the minimum value will be identified in this scan. The result (Max-Min)/2 is defined as baseline flatness. If the value is within the range of (±0.004ABS), the baseline flatness item will display as <OK>. When one function is completed, the system display <OK> or <NG> message as described in the above.

Press <7> key on the keyboard to check hardware. The hardware of the instrument will be listed on the right side on the screen as shown below.

3.9.1 Retrieving a Test Stored in Inside Memory

You can retrieve a test method that you have stored in Inside memory by following steps: Press <1> on the keyboard to select the Load Test, and a file list will appear on the side of the screen. Select the desired test number using the UP and DOWN arrow key and then press ENTER. The operating parameters for the selected test are initiated automatically and the screen then advances immediately to the START position.

3.9.2 Deleting Test From Inside Memory

Press [2] on the keyboard to select the Delete Test, and a file list will appear on the side of the screen.

Use the UP and DOWN arrow keys to select the desired file number and then press [ENTER]. Then the test method has been deleted from the inside memory. Press [RETURN] key at any time if you wish to cancel the method-set deletion request. Note: It is a good laboratory practice routinely to remove test method sets

that you no longer use. This ensures that there always available file numbers to allow the instrument to remember your method for you automatically.

3.9.3 Autostart Function

The AUTOSTART function allows you to select the operating parameters for a particular test, so that the test will be initiated automatically at instrument power-up. Press [3] on the keyboard to select AutoStart function.

Chapter 4 Maintenance

4.1 Self Diagnosis and Automatic Adjustment

The HALO DB-20 spectrophotometer will execute self-diagnosis and automatic adjustment immediately after the power switch is turned on. For details, refer to chapter 3.

4.2 Confirmation of Specifications

4.2.1 Wavelength Accuracy

1. Verify that the instrument is powered up and wait for 30 minutes at least. 2. Select Wavelength Scan from the Main Menu. 3. Set the operating conditions as shown below:

normal. If the peak is not in the range then: a) Change the existing scale to provide an easier to read graph and if

the peak is still not within range, perform the WL Calibration in Chapter 3.

b) Once you have performed the wavelength calibration, repeat steps necessary to confirm that the peak is within the specified normal value range.

4.3.2 Cleaning the Sample Compartment Windows

If the transparent window of the sample compartment is contaminated, remove the window plate, and then clean it completely. Be sure to wash your hands thoroughly before cleaning the window, and then wipe off the contaminant with a soft paper or cloth soaked with alcohol and ether solution (mixed in a ratio of 1 to 1).

CAUTION: Pinch both ends of the window plate with your fingers to set it back in the sample compartment. Do not leave fingerprints on the window plate.

4.4 Troubleshooting

5.2 Part Replacement

5.2.1 Replacement of Light Source Lamp

Cautions on Replacement of Lamp Do not replace the lamp with the power switch on. Note that high

voltage is applied to the lamp. High voltage is discharged while the power is turned off.

Wait for at least 10 minutes after the power switch is turned on. Wear clean gloves for mounting a new lamp. Do not leave fingerprints

on the lamp tube wall. For D2 lamp, care should be taken to prevent leaving fingerprints on the protruded sections.

The following shows how to replace the existing lamp with a new one.

(1) Turn off the power switch. Pull off the power cord. Then, wait for at least 20 minutes until the lamp cools down.

(2) Remove the fastening screw from the side face of the light source cover, and then remove the cover. (See Fig. 5-1)

(3) Replacement of WI Lamp Loosen the retainer spring, and then pull it out. Push in a new lamp to the extreme end of the lamp holder.

(5) Replacement of D2 Lamp

a) Pull out the D2 Lamp connector.

b) Hold the top end of the D2 lamp, and turn it counterclockwise until it is separated from the holder. (See Fig. 5-3)
