Phage purification

• Harvest phage infected cells. Phage in SUPERNATANT!

• Concentrate by precipitation (+PEG/salt)

• Pellet may not be visible…have faith

Plaque assay

• Serially dilute phage in water

• Mix with bacteria

• Add warm “top agar” and plate on LB--> 37° overnight

• Give remaining phage to Luis
