BioMaterials: Let’s start with the biology
1. Infection
2. Amplification
3. Morphogenesis + release of progeny phage
• new M13 phage particles within 10’ post-infection
• 1000 phage/cell within the first hour

Parts 2 + 3 of your experiment today
Phage purification
• Harvest phage infected cells. Phage in SUPERNATANT!
• Concentrate by precipitation (+PEG/salt)
• Pellet may not be visible…have faith
Plaque assay
• Serially dilute phage in water
• Mix with bacteria
• Add warm “top agar” and plate on LB--> 37° overnight
• Give remaining phage to Luis
BioMaterials: now for materials
Oxidized (purple) Reduced (clear)
Part 1 of your experiment today
Hydrolyze Iridium with citrate at 90° 4 hrs
Generates Ir colloidal particles
Next time will oxidize to form IrOx colloidal particles which
can react with phage p8
You must wear safety glasses, gloves, lab coat to do this part
of today’s experiment
