ANALYSIS

The question samples (EX01-1 and EX01-2) were compared against the reference samples (EX02-1 and EX03-1) by deoxyribonucleic acid (DNA) analysis of short tandem repeats (STRs) using polymerase chain reaction (PCR) technology. The amplification of DNA was performed using Promega’s PowerPlex 16 HS kit, which includes the thirteen core CODIS loci as specified by the FBI (D3S1358, TH01, D21S11, D18S51, D5S818, D13S317, D7S820, D16S539, CSF1PO, vWA, D8S1179, TPOX, and FGA), as well as Penta E, Penta D, and Amelogenin.

The analysis was performed on an ABI Prism 310 Genetic Analyzer. The raw analytical data is available upon request.

The sample identified as EX03-1 (reference blood sample of Macy Beth Queen) produced a DNA profile.

Item 1 – Dagger EX01-1 (blood stain from blade of dagger)

Summary: DNA profile consistent with Duncan King The sample identified as EX01-1 produced a DNA profile at sixteen of the sixteen loci analyzed. This DNA profile is consistent with a single contributor. The DNA profile is consistent with that of EX02-1. Hence, Duncan King cannot be excluded as a source of this sample. Statistical Interpretation: Using the genotypic frequencies published in Applied Biosystems’ AmpFlSTR Identifiler User’s Manual, the statistical probability of another unrelated individual with the same alleles as EX01-1 is:

This DNA profile is consistent with a single contributor. The DNA profile is consistent with that of EX03-1. Hence, Macy Beth Queen cannot be excluded as a source of this sample.

CLOSING REMARKS

The submitted items and associated exhibits will be retained until further disposition. Sufficient quantities of reference and question samples remain for reanalysis, depending upon the type of DNA analysis technology used. This analysis was performed under chain of custody. The chain of custody documentation is on file at Guardian Forensic Sciences. Positive and negative controls in each stage of analysis of this case performed as expected.
