EXECUTIVE SUMMARY

The BREAK BIOFILMS Training Network “Breaking Bad Biofilms: Innovative Analysis and

Design Rules for Next-Generation Antifouling Interfaces” started in January 2019. This report

presents the progress during the first year of the project, providing an overview of the work to

be carried out and introducing the 15 ESRs recruited.

INTRODUCTION – THIS IS US!

BREAK BIOFILMS is a consortium of world leaders in sensing, cell imaging, interfacial

engineering, microbiology and nanoformulation from 6 universities, 8 companies, a consortium

of food industries, a research centre, and a business and innovation centre (Table 1). A key

element of BREAK BIOFILMS is multidisciplinarity, which represents a unique strength and

opportunity to bring together 15 early stage researchers (ESRs) with diverse backgrounds.

THE PROJECT

Why biofilms?

Biofilms are communities of microorganisms which can be formed at all kinds of surfaces:

plastic, metals, natural materials, etc. This means that biofilms are all around us!

How do we break biofilms?

BREAK BIOFILMS aims to deal with the harmful impact they have in our daily lives

following an integrated strategy from biofilm detection to destruction.

These objectives are challenging and exciting. The ESR (or “the Breakers”) have access to

state-of-the-art equipment and will obtain a unique technical, industrial, and entrepreneurial

training to achieve these goals.

Structure of BREAK BIOFILMS

This Project is structured into eight different workpackages that are interrelated and

focused on the training of the next generation leaders. Our Breakers will spread the science as

well, since they will be intimately involved in the communication activities of the network.

OUR ACTIVITIES

Scientific progress

WE UNDERSTAND BIOFILMS

Develop multifunctional probes to measure simultaneously pH, reactive oxygen species or

membrane potential at bacterial colonies. Some model bacteria have been selected for

mapping topography/surface charge of samples.

Selection of antibacterial agents to deliver locally to biofilms using nanopipettes.

Investigate the swelling behaviour of AgNPs-fluoropolymers (AgCFx) composite films

with AFM, and mapping the release of Ag ions via SECM in combination with square wave

voltammetry.

WE DETECT BIOFILMS

Initial selection of signaling molecules as potential targets for biofilm detection in order

to develop a paper-based sensor.

Preparation and characterisation of gold electrodes and novel screen printed electrodes

for low cost point-of-need sensing.

Development of novel polymer based method for the immobilisation of high coverages of

active antibodies for bacteria and their characterisation using super resolution

microscopy.

Selection of strains for biofilms and biosensor studies.

Electrochemical production of copper-benzalkonium chloride superNAMs.

First experiments on preparation of different vesicle types for encapsulation of antibiotics

and endolysins.

Evaluation of anti-biofilm mixtures composed of endolysins and bacteriophages.

WE INHIBIT BIOFILMS

Electrodecoration of iron oxide magnetic nanoparticles by copper nanoparticles with

antibacterial activity.

First experiments to characterize the real time swelling behaviour of AgNP-fluoropolymer

composite films using atomic force microscopy.
