WHAT IS ÄKTA PURE? ÄKTA pure is an easily customizable liquid chromatography system designed for protein purification. The UNICORN software comes with a library containing a wide variety of column parameters and predefined method templates to increase productivity and efficiency. High pressure and air bubble sensors are incorporated in the system to allow unattended runs. A programmable email feature is available to notify operators when an alarm is triggered.

1. Column packing
2. Buffer prep (molarity, buffer degassing)
3. Column validation (asymmetry & HETP)
4. Instrument preparation (system purge, ethanol check for pumps)
5. Sample preparation, choice of loop, manual injection
6. Fraction collection system
7. Run samples
8. Process results
9. System maintenance and trouble- shooting
DAY 1
1. Brief introduction to liquid chromatography
2. Introduction to the ÄKTA pure systems (hardware and software)
3. Introduction to column resins and chromatography separation methods
4. Hands on: setting up instruments, priming lines and purging pumps
5. Quick column packing and prep of buffers for demos
6. Familiarization with software
7. Running the instrument
DAY 3
1. Trainee to repeat tasks from day two with assistance if needed
2. Trainee given a protein sample with instruction for purification
3. Hands-on, shut down and regular maintenance
