AMLODIPINE BESYLATE
Process Description
Stage-1
Phthalic anhydride reacted with Mono ethanol amine in Toluene media forms
phthalimido ethanol
Stage-2
Phthalimido ethanol reacted with Ethyl-4-chloro aceto acetate in presence of sodium
hydride, Toluene forms ethyl phthalimido ethoxy aceto acetate.
Stage-3
Ethyl phthalimido ethoxy aceto acetate condensate with orthochloro Benzaldehyde to
give Stage-3 as product.
Stage-4
Ethyl phthalimido ethoxy aceto acetate and orthochloro Benzaldehyde condensate
reacted methyl β-amino crotonate in Acetic acid media and further purified in Ethyl
acetate and forms phthalimido Amlodipine
Stage-5
Phthalimido Amlodipine reacted with Mono methyl amine forms Amlodipine Base.
Stage-6
Amlodipine Base reacted with Benzene sulphonic acid in water media forms Amlodipine
Besylate
Stage-7 (Purification)
Amlodipine Besylate further purified in Methanol and Ethyl Acetate to get Pure

ATORVASTAIN CALCIUM
Process Description:
Stage-1
Tert-Butyl-2-[(4R,6S)-6-(cyano methyl)-2,2-dimethyl-1,3-dioxan-4-yl] acetate
reduction with Hydrogen on Palladium Carbon in presence of Methanol gives tert-
Butyl-2-[(4R,6S)-6-(amino ethyl)-2,2-dimethyl-1,3-dioxan-4-yl] acetate.
Stage-2
Aniline on condensed with Dimethyl Carbonate and 2-Methyl-2-butanone in presence of
Methanol gives 4-Methyl-3-oxo-N-Phenyl pentanamide.
Stage-3
4-Methyl-3-oxo-N-phenyl pentanamide upon reaction with Benzaldehyde in presence of
Potassium Carbonate, Acetone and Toluene media gives 2-Benzylidene-4-methyl-3-oxo
pentanoic acid phenyl amide.
Stage-4
2-Benzylidene-4-methyl-3-oxo pentanoic acid phenyl amide on reaction with tert-Butyl-
2-[(4R, 6S)-6(amino ethyl)-2,2-dimethyl-1,3-dioxan-4-yl] acetate and fluoro benzaldehye
in presence of Para toluene sulfonic acid, Isopropyl alcohol and toluene media to give
Atorvastatin ester.
Stage-5
Atorvastatin ester undergoes hydrolysis in presence of sulfuric acid and methanol gives
Atorvastatin.
Stage-6
Atorvastatin upon salt formation with calcium acetate in presence of Isopropyl alcohol
gives Atorvastatin Calcium.

Process Description:
Stage-1
N-carbethoxy-4-piperidone is reacted with Hydrogen and Ammonia in the presence of
Raney nickel as catalyst and Methanol as solvent media to give stage-1 as product.
Stage-2
Stage-1 reacts with 2, 5-Dichloro nitrobenzene & Sodium Carbonate in presence of
Methanol as a solvent media to give Stage-2 as product.
Stage-3
Stage-2 reacts with Hydrogen in presence of Raney Nickel as catalyst and Toluene as
solvent media to give Stage-3 as product.
Stage-4
Stage-3 reacts with Urea in the presence of Sodium Hydroxide as a solvent media to
give Stage-4 as product.
Stage-5
Stage-4 undergoes hydrolysis with Sodium Hydroxide to give Stage-5 as product.
Stage-6
Stage-5 reacts with 1-(3-chloropropyl)-1, 3-dihydro-2H-benzimedazole-2-one and
Sodium Carbonate in the presence of Methyl Isobutyl Ketone as solvent media to give
Domperidone as product.

subsequently on Nitration with Nitric Acid, Sulfuric Acid to produces stage-1.
Stage-2
Stage-1 Product reacts with 2, 2, 2- tri fluoro ethanol and Sodium hydroxide to give
Stage-2A reaction mass. Reaction Mass (Stage-2A) reacts with Acetic Anhydride in
presence of Acetic Acid, an intermediate is formed. Further it is treated with dry
hydrochloric acid and Sodium hydroxide to give stage-2 Compound.

Above compound Chlorinated with Thionyl chloride in presence of Toluene solvent
media to give stage-3 Chloro compound
Stage-4
Stage-3 Condensed with 2-Mercapto benzimidazole in Presence of Sodium Hydroxide

Stage-5
Stage-4 Compound reacts with Hydrogen peroxide in presence of IPA produces

LOSARTAN POTASSIUM
Process Description:
Stage-1
OTBN and Sodium nitrate are made to react together in Toluene solvent medium
using TEA HCl as catalyst. Product is isolated by using Hydrochloric acid.
Stage-2
The Above stage-1 material is treated with Trityl chloride. Methylene dichloride is
used as solvent. Product is washed with water and crystallized in Methanol
Stage-3
Stage-2 is dissolved in Methylene dichloride and treated with a mixture of AIBN and
DDH. Methylene dichloride is distilled-off to obtain the product. Finally the product is
treated with Sodium Metabisulhite solution. The product is washed with Ethyl acetate
while centrifuging.
Stage-4
Stage-3 mass is dissolved in sodium borohidride solution. Butyl chloro formyl
imidazole(BCFI) , TBAB are made to react with stage-III solution. Finally stage-III
product is purified with Carbon and water
Stage-5
Stage-4 mass is dissolved in IPA and HCL mixture. The mass is isolated by
precipitated with water. The reaction mass pH is adjusted with Sodium hydroxide to
get the Losartan Base.
Stage-6
The stage-5 mass is treated with Potassium hydroxide in Methanol solvent medium.
Product is purified with Carbon. Pharma is isolated by distilling of Methanol.

PANTOPRAZOLE SODIUM
Process Description
Stage-1
Maltol and Dimethyl sulphate condense in Acetone solvent medium. Excess Dimethyl sulphate
is removed by treating the reaction mass with Potassium Carbonate.
Stage-2
Stage-1 compound on treating with Liquor Ammonia solution to give stage-2 as product.
Stage-3
Stage-2 compound on reaction with Phosphorous oxy chloride produces stage-3 compound.
Further reaction is stopped by treating the reaction mass with water. Final product is extracted
with MDC and after distilling out MDC compound is obtained.
Stage-4
Stage-3 compound reacts with Hydrogen peroxide in presence of Acetic acid & Methanol,
stage-4 compound is obtained.
Stage-5
Stage-4 compound reacts with sodium hydroxide in presence of Methanol to produces stage-5
compound.
Stage-6
Above stage-5 compound on reaction with Acetic anhydride produces an intermediate.MDC
used as solvent. Product is precipitated with C.S. Flakes.
Stage-7
Stage-6 compound when reacts with Thionyl chloride an intermediate is formed.MDC is used
as solvent. Product is crystallized in Methanol. Finally product is purified with Acetone.
Stage-8
When 2-Chloromethyl-3, 4-dimethoxy-pyridine Hydrochloride reacts with 5-diFluoro methoxy-2-
mercapto-benzimidazole, in presence of Sodium hydroxide, product is formed. Mythylene
dichloride is the solvent.
Stage-9
Stage-8 is treated with Sodium hypochlorite in presence of Sodium hydroxide, to produce the
Pantoprazole sodium .This is purified in Acetone. Methylene dichloride is used as solvent.

Stage-3 Compound is reacted with Thionyl Chloride in presence of MDC as a solvent media to

Process Description
Stage – 1
β-Thymidine is reacted with Trityl chloride and Tri ethyl amine in the presence of 1, 4-
dioxane to give 5’-O-Trityl Thymidine.
Stage –2
5’-O-Trityl Thymidine is reacted with methane sulphonyl chloride and tri ethylamine in
the presence of Toluene to give mesyl thymidine intermediate, which is reacted with
tri ethylamine in the presence of methanol to give 5’-O- Trityl -2,3’-Anhydro Thymidine.
Stage – 3
5’-O-Trityl -2, 3’-Anhydro Thymidine is reacted with sodium azide and ammonium
chloride in the presence of di methyl sulphoxide to give 5’-O-Trityl -Zidovudine.
Stage – 4
5’-O-Trityl –Zidovudine is reacted with p-Toluene sulphonic acid monohydrate in the
presence of Methanol to give Zidovudine.
