Areas of research activity

Characterisation and exploitation of microbial molecular diversity (i) Microbial biodiversity, particularly of extreme environments (hydrothermal, including deep

sea hydrothermal vents) and cold (Antarctic). Techniques include in situ ATP analysis, 16S rRNA gene phylogenetics and DGGE. These studies are used to identify the scope of unculturable microbial diversity and as a basis for analysis of physiological properties such as trophic interactions, gene transfer and molecular adaptation. The major molecular ecology program in the IMBM laboratories is a large research project in collaboration with Professor Craig Cary (U. Waikato, NZ) on the diversity, trophic structure and molecular function of microbial communities in Antarctic Dry Valley soils.

(ii) Novel enzymes for biotransformations, including hydrolyses (esterases/lipases), chiral

reductions (dehydrogenases), oxidations (peroxidases, dioxygenases) and nitrile conversions (nitrile hydratases and nitrilases). Techniques include the screening and isolation of new extremophilic enzymes, enzymic purification and characterisation, cloning and expression of the enzyme genes and biotransformation studies. An extension of this field is the investigation of the behaviour of enzymes in the immobilised state and in organic-aqueous solvent systems. We focus particularly on the nitrile-modifying enzyme group, and have an extensive collaboration with local structural biologists (M Sayed, UWC; T Sewell, UCT) and international enzymologists (MJ Danson, U. Bath; RM Daniel, U. Waikato).

(iii) Metagenomic Gene Discovery: methods for direct exploitation of microbial genomic

diversity. Metagenomic Cloning is a means of accessing the ‘unculturable’ fraction of natural microbial populations and provides a route to the identification of new catalytic activities and bioactive peptides. This technology has very wide applicability and, in conjunction with appropriate assay methods (e.g., reporter assays, receptor binding, bioactive responses), may be adapted for the identification of novel peptide therapeutics. We have successfully used plasmid and fosmid expression libraries as a source for identifying novel enzymes.

(iv) IMBM has embarked on a major biofuels research program. In collaboration with TMO

Renewables (UK) Ltd, researchers have used gene discovery techniques to search for genes which can enhance bacterial ethanol synthesis. In parallel, the Institute has successfully raised funding from PlantBio/TIA for a 4-year research program on the development of enzymes for lignocellulosic digestion. This program, which includes research groups from the University of Stellenbosch and CSIR, Modderfontein, is coordinated by DAC.
