RESEARCH INTERESTS: The research interests of my laboratory focus on studies of the implication of the natural fibroblastic 3D microenvironment progression due to tumor development in a bi-directional exchange of information at cell-matrix contacts such as “three-dimensional matrix adhesions” also known as ‘3D-adhesions.’ For this, I have developed a primary fibroblast-derived and in vivo-like 3D system that mimics stroma human progression (i.e., desmoplasia) in assorted epithelial cancers such as pancreas, kidney, inflammatory and regular breast, as well as ovary and lung. We use this physiologically relevant 3D system to investigate the mechanisms of matrix induced myofibroblastic differentiation (e.g., desmoplastic activation) and the tumorigenic effects imparted by tumor-associated extracellular matrices (i.e., tumor cell invasion, growth, etc) as well as its effects in neo-neurogenesis and perineural cancer cell invasion. The physiological/pathological relevant progressive stroma 3D system also serves as basis for a platform investigating tumor-associated matrix induced drug responsiveness and discovery of activated stromal (including neural) and cancer biomarkers. Tissue engineering methodologies are used to question the topographical/architectural vs. biochemical aspects of tumor-associated stromal microenvironments. Novel pre-clinical therapeutic approaches are assessed using human implanted surgical tissues in immune-compromised mice as well as via orthotopic and isogenic murine xenograft and spontaneous genetic models. Validations are conducted using simultaneous multi-channel spectral immunofluorescence using human surgical tissue microarrays that include well-annotated clinical information and patient outcomes; we developed special analysis software needed for this approach https://github.com/cukie/SMIA-CUKIE. Studies incorporate molecular biology, biochemical and cell-based assays, as well as laser scanning confocal immunofluorescence, real time multi-channel microscopy, digital imaging analyses, genetic manipulations, tissue patterning, mathematical modeling and more.
