It is unlikely that these precast gels could be used with visible fluorescence automated DNA sequencers such as the ABI PRISMR 377 since the system sensitivity would be lower due to significant emission of background fluorescence by the soda-lime plates.

With the LI-COR system, by using infrared radiation, the background fluorescence due to the soda lime glass of the precast gel is greatly reduced.

Multiplexed PCR reactions were run on a CastAway 8% gel and analyzed using Hitachi’s FMBioII Scanner. The pattern of banding, corresponding to family specific CDR3 length polymorphisms allows comparison of T cell receptor utilization by peripheral blood lymphocytes with that of breast tumor-infiltrating lymphocytes.
