This User Manual includes special details regarding to DASware migrate and the handling of 3rd partybioprocess controllers with DASGIP Control. Refer to the DASGIP Control User Manual for complementaryinformation.

1.1 Using this Manual

1 DASware® Solutions

Increasing process complexity coupled with rising cost pressures and rapidly evolving regulatoryrequirements make today’s process development efforts a special challenge. The pressure of achievingfaster time to market for new and innovative biopharmaceuticals has led to the need to optimize everyelement of the total development workflow.

DASGIP‘s DASware was designed as a suite of smart and flexible software solutions to acceleratebioprocess development. It enables interconnectivity of bioreactors with external lab devices,comprehensive data- and information management, factorial design of experiment and remote control ofbioprocesses. DASware can be used with the DASGIP Parallel Bioreactor System for benchtop applicationsin microbiology and cell culture as well as with legacy benchtop bioreactor controllers.

In the past, bioreactor processing, recipe management, process and product analysis, data generation, datamining and storage were poorly interconnected thus requiring time-intensive manual work. DASwaresupports interconnectivity combined with sophisticated bioprocess information management. Theutilization of DASware enables streamlined process development at benchtop scale in accordance with QbDstandards.

DASware opens up additional dimensions for bioprocesses and their information management. Usersbenefit from accelerated up workflows, which e.g. are also DoE capable, optimal interconnectivity betweenbioprocess-controller and analyser as well as almost unlimited access to bioprocesses.

1.1 DASware® access

DASware access provides an unprecedented level of freedom and flexibility in the management ofbioprocesses. Each DASGIP Control System on-site is accessible remotely by one or more remote clientssimultaneously. Depending on the user-defined configuration and associated authentication either monitoror monitor and control access can be enabled for any network or mobile client.

Wi-Fi, Intranet, VPN and 4G connections can be used to provide web based access with almost everybrowser to one or more bioreactor systems via PC, Notebook or Netbook. The unique DASGIP iAppsupports access from iPhone, iPod touch and iPad, optionally with webcam support.

1.2 DASware® analyze

DASware analyze was designed for seamless integration of sampling and analytical laboratory devices tothe bioreactor system. A broad and growing range of analyzers can be integrated, among them nutrientanalyzers, cell counter (YSI Life Science, Nova Biomedical), automation platform and autosampler (BayerTechnology Services, BaychroMAT®) biomass monitors (Aber Instruments), HPLC (Waters, Shimadzu) andmass spectrometers (Thermo Scientific).

The OPC network protocol allows for interconnectivity between the bioreactor system and the analyzer,including the possibility of direct feedback from the bioreactor system according to online measuredanalytical data. This facilitates feedback control loops for e.g. nutrients, biomass or product concentrations.Online calculations as well as event and data-driven decisions are supported. The unique bidirectional OPCcommunication, available for supporting devices enables sampling on demand and process dependentanalyzer panel selection.

1.4 DASware® design

DASGIP’s Parallel Bioreactor Systems serve as an ideal platform to carry out the Design of Experiment(DoE) principle on bioreactors in parallel. DASware design automatically compiles DoE information from3rd party DoE tools into recipes and feeds-back response information into DoE and multivariate analysisand reporting tools.

DASware design comes with a built-in Full Factorial DoE builder. Alternatively, a large variety of DoEdesigns for screening, process development and optimization can be automatically imported from mostpowerful 3rd party DoE tools like JMP and others. Parallel recipes incorporating the DoE factor variations(i.e. pH, DO, T set-points or feed rates) are automatically populated. Following DASGIP’s Point-Click-Growconcept, they can be carried out on a set of bioreactors with a single mouse-click. DoE responseinformation is collected and prepared for an automated export.

DASware discover helps you to structure efficiently the assortment of process information like

Use this intuitive database query tool to initiate real-time retrievals and run-to-run comparisons. All criticalprocess information is clearly listed as table and can be configured. In addition, you can easily generatecharts in Microsoft Excel® using DASGIP Documentation Wizard.

1.6 DASware® migrate

A significant installed base of ageing bioreactor controllers offers rudimentary cultivation controlcapabilities. However, recent advances such as bioprocess data management, analytical device integration,DoE support and remote control, available from DASware solutions, are often missing. Now users of 3rdparty bioreactor controllers can also benefit from DASGIP’s intelligent software solutions.

DASware migrate integrates most common 3rd party bioreactor control units and systems (e.g. SartoriusStedim Biotech & Applikon) as well as DASGIP modules into legacy bioreactor controllers (e.g. gas analyzerGA4, gassing station MX4/4, mulitipump MP8).

• Seamless integration with DASGIP Control (version 4.5 and higher)

• Display, logging and charting of controller variables in DASGIP Control

• Control options for set-points using DASGIP Control

• On and off switching of the bioprocess controller using DASGIP Control

• Seamless integration of DASGIP Advanced Trigger Automation and DASGIP Advanced Scripting

3 Safety

3.1 Intended Use

3.2 Warning for Intended Use

Read the User Manual first and observe the following general safety instructions before using DASwaremigrate.

3.2.1 Information on Product Liability

In the following cases, the protection provided by the device may be impaired. the liability for the functionof the device passes to the operator if:

• The device is not used in accordance with the User Manual.

• The device is used outside of the range of application described in the preceding chapters.

• The owner has made unauthorized modifications to the device.

Lethal voltages inside the device.

Ensure that the housing is always closed and undamaged so that no parts inside the device can be contacted by accident.

Do not remove the housing of the device.

Do not allow any liquids to penetrate the inside of the housing.

Electric shock due to damages to device or mains cable.

Only switch on the device if the device and mains cable are undamaged.

Only use devices that have been properly installed or repaired.

In case of danger, disconnect the device from the mains supply.

4 Installation

Each DASGIP process computer comes pre-configured with all necessary software installed. The license ofDASware migrate requires no further installation steps. All relevant services (gateway) will startautomatically within the program start of DASGIP Control.

4.1 Interfaces

4.1.1 New Brunswick

Connect the serial port of a New Brunswick bioprocess controller to the serial port of the device server (i.e.Moxa) according to Fig. 4-1. The device server is connected to the DASGIP process computer via Ehernet.

You can not use DASware migrate and New Brunswick BioCommand at the same time.

4.1.2 Sartorius DCU

Sartorius bioprocess controllers can be directly connected to the DASGIP process computer via Ethernetaccording to Fig. 4-2.

4.1.3 Applikon ADI

Connect the serial port of an Applikon bioprocess controller to the serial port of the device server (i.e.Moxa). The device server is connected to the DASGIP process computer via Ethernet.

It may be necessary to involve your IT administrator to ensure proper configuration, IPaddresses and port settings.

You can not use DASware migrate and Sartorius MFCS/win at the same time.

4.2 IT Network Integration

In a local set-up the DASGIP Process Computer is directly connected to one or more Controllers usingnetwork cable or serial device servers. If the Process Computer is integrated in the local IT infrastructurethe set-up can be different to the installations described before.

You can not use DASware migrate and Applikon BioXPert at the same time.

It may be necessary to involve your IT administrator to ensure proper configuration, IPaddresses and port settings.

5 Operation

DASware migrate displays all relevant process and controller values as well as set-points of the integratedbioprocess controller in a Summary and Process View of DASGIP Control (s. 5.3.2).

5.1 First Steps

Switch on the process computer.

5.2 Login

The program symbol of DASGIP Control is displayed on the desktop of the process computer.

Double click on the program symbol to start DASGIP Control.

Enter your user Name and the corresponding Password.

Click Login to continue.

You can operate the bioprocess controller using the operation/touch panel of the controller orDASGIP Control. For more details regarding the process start refer to 6.1.4.

Please enable the remote function of the bioprocess controller (according to the manufacturer‘smanual) to ensure the full access to all available operations for DASGIP Control.

5.3.1 Settings for Bioprocess Controllers

Since the DASware migrate license is activated, the integrated bioprocess controller is displayed in DASGIPControl. Use the following procedure to check the settings:

Choose the Overview tab on the left to display the navigation tree. Click on Plant > DASGIP Plant … > Modules to show the list of all integrated bioprocess controllers and other connected modules.

Right click on the controller name to open the context menu.

Choose Info to display the corresponding information on the right.

When you have started a process from the bioprocess controller, DASGIP Control displays the processname on the Overview tab under the branch Running in the Navigation Tree.

5.3.2.1 Summary View

Use the Navigation tab for a simple and intuitive operation within DASGIP Control. You can easily switchbetween Summary and Process View of different setups (vessels) of a process at any time.

Click on the Xxx.Control button to display a Summary View, which gives a summary of all process values like pH, DO value(s), gas mixture, gassing rate and feeding volumes for a set of vessels selected to be operated in parallel (Fig. 5-2, Fig. 5-3, Fig. 5-4). Large numerical display elements facilitate an easy reading of all relevant process values.

The displayed views vary depending on the integrated bioprocess controller. The following shows thedifferent Summary Views of DASGIP Control with an integrated BioFlo/CelliGen 115 from New Brunswick, aDCU from Sartorius, and an ADI 1010 from Applikon illustratively.

The operating status of different sensors and actors (feeding pumps, gasmixer, impellers, etc.) is displayedby color coded symbols and monitored. The following table (Tab 5-1) shows the color definitions for thesymbols.

Additionally, you can click on each symbol to display the current process values of all affected reactors onthe right side in the Summary View.

5.3.2.1 Process View

When you have started a process within the bioprocess controller, Process View displays all color codedtransferred controller states.

The following shows the different Process Views of DASGIP Control with an integrated BioFlo/CelliGen 115-from New Brunswick, a DCU from Sartorius, and an ADI 1010 from Applikon illustratively.

Please check and activate / deactivate each function according to your needs. DASGIP Control does notforce the controllers into a default state to avoid conflicts with local operations. For this reason the on/offsymbols (x/o) may not reflect the controllers state/s through the color does.

Synchronized display (color matches x/o symbol). Click on the cross or the white point of asymbol to activate or deactivate the corresponding action.

Unsynchronized display (color does not match x/o symbol).

Different symbols are available for vessels, sensors and controllers as well as actors likepump, gassing and agitation.

Controllers like pH and DO (i.e. DO Control) can operate actors like pumps, gas mixing or agitation.

In this case orange symbols indicate actors which are controlled interdependently.

The color indicates that direct access it is not possible. You have to inactivate the controller itselffirst by clicking on the white point of the symbol e.g. of DO Control to get access to theinterdependent gas mixing. After a short delay the gas mixing symbol will turn from orange to grayand can be operated manually.

Click on the vessel symbol to display the vessels parameters at the right side of the working area.

Click on a controllers symbol to display the corresponding controller parameters.

The parameter dialogs are subdivided in three tabs:

5.3.2.1 Define or Adapt Set-point Profiles

DASGIP Control provides a very easy to use and flexible way to enter and edit controller profiles.

Click on a controller symbol in Process View to open the corresponding values in Details at the right side.

Ti Reset time The reset time (Ti) influences the integrating part and therefore thesensitivity of the control. The larger the value, the smaller the effect onthe control. The control becomes smoother, but slower at the sametime.

Td Derivation time Differentiator

Min Lower limit Minimum value of the control variable (Anti-Reset-Windup).

Max Upper limit Maximum value of the control variable (Anti-Reset-Windup).

Deadband Deadband Control deadband for a two-sided control.

Slope Slope Sensor slope calibration parameter.

Offset Offset Axis shift as calibration parameter.

Calibration Calibration Flow rate at 100 % output.

Please note that not each 3rd party bioprocess controller supports all parameters. Therefore theDetails tab in DASGIP Control displays differing parameters depending on the integratedcontroller.

Click on the Profile tab to enter and edit the corresponding profile options.

The drop down menu of Profile Source gives the opportunity to choose between different time or valuebased profiles (a detailed list of all profile sources is given in chapter 3.3.2 of the User Manual DASGIPControl).

Each profile is sectioned into the three distinct presentation modes Type, Data and Graph:

Type comprises three different profile types. Choose Constant Value, Datatable or Function as profile type.

Use the Data tab to enter profile values and Graph to show the corresponding profiles graphically (pleaserefer to chapter 3.3.2 and 3.7 of the User Manual DASGIP Control for more details).

5.3.2.2 Alarm Option

If the alarm option is activated, DASGIP Control displays alarm options on an Alarm tab within the specifiedsensor values separately.

Click on the title bar or a process value (e.g. DO.PV) to display the corresponding sensor values. There is an additional Alarm tab, if the alarm option is activated.

Click on the Alarm tab to adjust the alarm settings.

Please note that all alarms are inactive until the inoculation clock(Inoculation) is started.

5.3.2.3 Offline Values

You can supplement the process documentation with offline values in DASGIP Control. These valuesdetermined by external analyzers can be entered together with the time stamp of determination directly orlater on. They will be stored as an event in the database and can be displayed together with all otherprocess values e.g. in a diagram.

Click on the Offl. symbol in the Process View to display the dialog at the right side. You can enter up to four Offline values and user events by default. Additionally, you can specify the name as well as the engineering unit. Please refer to DASGIP Control User Manual chapter 5.1 for details.

5.3.2.4 Chart Control Panel

During a process run all values are visualized in online charts in the Summary or Process View (Fig. 5-8) atthe bottom of the window.

Click on the corresponding curve button to hide or display a curve on the diagram.

Click on the chart, hold the mouse button and drag a box to zoom in an area.

Click on the x- or y-scale and drag down the appearing double line to display a legend with corresponding values.

Click on the Tools button on the upper left edge of the Chart Control Panel to display further options. You can display all available data (Show all curves) or just data of determined time intervals (Zoom to last hour‘s data).

You can configure and save chart settings individually (Save User chart settings). Please refer to the UserManual DASGIP Control chapter 6.1.1. for more details.

5.3.2.1 Event Monitor

The Event Monitor at the bottom of the DASGIP Control window displays all events, interventions andpossible error messages of the connected bioprocess controllers during the process runtime.

5.3.2.1 Plant Overview

The Plant Overview gives a complete tabular overview of the current process status and eases the access tomain procedures’ process values. Additionally, the already logged process values of the running processcan be viewed graphically and displayed according to individual points of view. Therefore you can combineany variables even historical ones for trend analysis.

Click on the Navigation tab or on the plant name (XXX Plant) in the Navigation Tree to start the Plant Overview.

Click on single values or select all values of a column by clicking on the column header. Your selection is marked by color and you can add values by pressing the <Ctrl> key and clicking simultaneously.

6 Quick Start

6.1 Short Instructions

Start DASGIP Control.

6.1.1 Calibration and Cleaning

Perform Calibrations and Cleaning with the bioprocess controller.

6.1.2 Start a New Workflow

Define a New Workflow for a process.

Click on File in the menu bar and choose New Workflow or click on the corresponding symbol.

There are two ways to start a workflow:

1. Select an already pre-defined template from the Default Template tab, which suites the workflow of the bioprocess controller best.

– The template names, which can vary from system to system, are listed right aside Selection. (Fordetailed information regarding the creation of templates refer to DASGIP Control User Manualchapter 3).

– Find a short description of the template aside Description.

Select the number of Reactors from the drop-down menu.

Please perform calibrations and cleanup using the bioprocess controller panel as usual. Due tolimited network protocols DASGIP Control cannot provide support for these procedures yet.

2. The User Template tab gives a list off all user defined workflows. Alternatively, you can select one of these workflows.

Select a workflow from the list.

You can adjust the workflow name aside Identifier.

Select the number of Reactors from the drop-down menu.– If the list consists of a lot of workflows, you can enter Search text and start searching with a click on

the Start Search button.– Additionally, you can select workflows from the Last modified drop down list. Click on New Search to

start the search with the new criteria.

DASGIP Control displays the Workflow Builder, which provides further options to adapt the workflow.

You can edit the names of each setup in the middle individually.

Click beneath Setup Identifier to enter the name.

You can change the mapping and the status of each setup by the drop down list of each edit box. The StateFree indicates available units and occupied indicates a process in progress. You can not start a new processfor an occupied unit.

At the right you can adjust more details.

DASGIP Control accepts only well-defined workflow Identifiers with limited special characters. Especially, ifyou want to use a workflow as template, we recommend using self-evident identifier.

DASGIP Control maintains the State automatically. Usually there is no need to change the statemanually.

Choose the appropriate vessel type from the drop down list next to Reactor or choose the universal vessel type UNI 1000.

Optionally, you can edit the displayed Comment.

6.1.2.1 Save as Recipe Template

You can use each created workflow as template for further processes.

Click on the checkbox next to Save as Recipe Template to use the current workflow as template.

Click the Save button to continue.

Click the Yes button in the following dialog to confirm the changes.

DASGIP Control displays each recipe template beneath Workflow > Template in the Navigation Tree and onthe User Template tab when you start a new workflow (Fig. 6.1.2) or run a workflow (Fig. 6.1.4).

Choose a template and click the Run button.

6.1.3 Recipe Editor

DASGIP Control opens the Recipe Editor Fig. 6-1. You can complete and adjust all recipe settings e.g.pre-defined set-points for control loops and actors at this point.

The Recipe Editor comprises several columns and one row for each setup (vessel). Corresponding to thepre-selected workflow, the profile cells are already allocated and values entered.

Choose Run Workflow from the File menu or click on the corresponding symbol to start each already set up workflow.

Already created and saved workflows are listed on the Workflows waiting to be started tab.

You can also adjust settings during process later on, but please note, that these changes will notbe stored for documentation. White recipe settings are part of the batch report in MicrosoftExcel®.

The User Templates tab lists all created and saved workflow templates.

Select a workflow and click the Run button to continue.

Subsequently, you can change the Workflow name, the number of Units, and Setup names.

Click the Run button to continue.

The process starts and DASGIP Control displays all relevant functions in a Summary View.(5.3.2.1).

6.2 Documentation

DAGIP Control logs all data and events in one central database (SQL). Further features such as export intocsv files via one mouse click and strong charting capabilities ease plotting and data comparison in anunrivalled manner.

6.2.1 Data Export

You can export all process data from completed processes.

Click on the Overview tab and open the Finished branch of the Navigation Tree.

Open the context menu by right mouse click.

Click on Load to load data.

A right mouse click on single procedures opens the next context menu. Use Export (CSV) to export data as csv file.

DASGIP Documentation Wizard eases the editing of csv files in Microsoft Excel®.

6.2.2 DASGIP Documentation Wizard

DASGIP Documentation Wizard is a Microsoft Excel® Add-In and provides an easy and convenientevaluation of exported files including a graphical presentation. Automatically generated online cultivationreports of current and historical runs can be compared to each other. This allows the chart creator to createtrends populated with the selected tracks i.e. pH and dissolved oxygen (DO) from a subset of runsautomatically. DASGIP Documentation Wizard helps you to select the necessary columns in the experiment data tablesheet and accomplishes also the following features:

• The time axis has the suitable format and scale for the chosen range.

• The axis of the process values is labeled.

• The chart and the corresponding sheet in the working folder are labeled according to the chosen options.

• The markers for data points (graphs), which are usually shown in Microsoft Excel® x-y-charts, areremoved. Due to the amount of data generated by DASGIP Control they are optically distracting andslow down processing speed. If needed, they can be added.

6.2.2.1 Installation

Each DASGIP Control PC includes an already installed DASGIP Documentation Wizard. The installation onan additional computer requires Microsoft Excel® in the version XP or higher.

Please refer to the User Manual DASGIP Documentation Wizard for more details.

6.2.2.2 Create a Chart

The DASGIP Documentation Wizard window is subdivided in three parts:

• Time axis displays a list of available units for the x-axis at the left.

• Value axis displays a list of available process values and setups (Channels) for the 1. y-Axis in the middle.

• Additionally, chart Options can be selected at the right.

Define the time axis for the chart. Timestamp is the default selection. Duration displays the process time and Inoculation Time the time since inoculation.

Select the process values from the list of 1.Axis for the chart. Click on the value names for selection and press the <Ctrl> key while highlighting with the mouse to select additional entries (selection).

If you require an additional y-axis, click on the Options tab the check box next to 2. Y-Axis. You will get an additional column of process values for the 2. axis.

Select the setups from Channels for the chart. Press the <Shift> key while highlighting with the mouse to select the range of entries between the existing highlighted entry and the next selected entry (selecting a range).

Use the corresponding check boxes of the Options and 1. Axis tabs to select certain features or trend linesfor the chart.

Check the option 2. Y-Axis to display an additional 2. Axis tab on the right.

Click the New Chart button to create a new data sheet with the chart.

Highlight at least one entry in each of the shortlists to create a chart.

6.2.2.3 Process Documentation

In addition to the charts, the Microsoft Excel® working folder contains tables with information regardingWorkflow, Procedures and the individual Setups, the recorded Data and Events. This information is shown ondifferent sheets.

The Procedure sheet contains all configuration data, which has been defined as starting values for this procedure during the workflow configuration:

• Controller settings

• Cascade settings

• Descriptions

• Alarm settings (optional)

• Data mining properties (optional)

The upper part of the table shows general information regarding the procedure. The lower one showsfeeding and control parameters from the profile headers of the Recipe Editor.

The sheets Setup 1, 2, ... contain the configuration data of the individual setups. This contains:

• Profile definitions

• Calibration values

• Trigger settings (optional)

The sheets Data1, Data2, … include all process values, which where recorded during the runtime of aprocedure

To reduce the data amount, values are just stored after significant changes or after a time span ofapproximately 5 – 60 minutes.

6.2.2.4 Fill Empty Data Cells

The data log interval and the data reduction feature is the reason for empty data cells on the data sheet.Empty cells result in errors when the data columns are used for calculations in Microsoft Excel®. To avoidthis DASGIP Documentation Wizard can fill cells automatically.

Click on the Add-Ins tab and select Dasgip > Data Column Manager.

Click the radio button next to fill with last value to fill the empty cells with the previous value or the one next to interpolate liear to fill the cells with an interpolated value.

Click on the Fill Button to start.

The new values of the chosen Setup (1) are displayed in blue color on the data sheet Data (1).

You can undo the cell filling.

Open the Daten Column Manager again and click the Undo button pm the Fill tab.

6.2.2.5 Add Data Column, Low Pass and/or Derivation

You can enter additional data columns, low pass filter and/or derivations by the Data Column Manager.

Start the Data Column Manager as described before.

Click on the Add Column tab and add the Name, the Unit and the number of Digits for the column.

Select the radio button next to Procedure to add a column regarding to a procedure, which will be displayed on the Data sheet.

Select the radio button next to Setup to add a column to each setup, which will be displayed on the Data1-x sheets at the end of the existing columns.

Click on the Low Pass tab and select a value from Value axis that will be displayed as Name.

Define the unit of the low pass (hh:mm:ss) in the edit box of T:

Switch between the tabs by clicking on the arrows in the upper right corner.

Define a derivation with respect to time on the d/dt tab.

Click on the d/dt tab and select a value from Value axis that will be displayed as Name.

The Events data sheet shows a list of all events that were recorded during the runtime of a process.

Events are sorted according to the timestamp. The different colors of the rows reflect the type andimportance of the even s. 5.3.2.1.

After the data export in Microsoft Excel® you can print charts and/or the complete process documentation.

Check, if the process computer is connected to the network (Ethernet).

Check, if the controller is connected to the device server (i.e. Moxa) for New Brunswick and Applikon or directly to the process computer for Sartorius.
