The basic screening and characterization functionality of Biacore Insight Evaluation Software ensures faster, data-driven decisions in SPR screening and characterization. The Extended Screening and Characterization package is available as an add-on extension package, with dedicated software tools for screening of LMW fragments. The extension package speeds up assay setup and streamlines evaluation, providing greater flexibility and tools to analyze complex biomolecular interactions.

Efficient evaluation of affinity and kinetic dataBiacore Insight Evaluation Software enables kinetic and affinity evaluation with a few simple clicks. The software is equally suited for the rapid analysis of large screening campaigns as well as deep kinetic characterization of a single interaction. Generic tools scale with the size of your experiment rendering fast results you can trust, irrespective of the number of samples analyzed.

The flexible evaluation interface (Fig 2) is configurable to maximize the space for your most important tasks at any time:

Simultaneous visualization of results from thousands of samples

Thousands of samples from multiple runs can be visualized simultaneously and the results presented in a Result plot. The Result plot provides efficient, flexible, easy-to-use tools to process the data of interest. Absolute or referenced data can be plotted and double-referenced by applying Blank subtraction. Sample responses can easily be harmonized with respect to the molecular weight of the analyte or the level of captured ligand respectively, making the entire data set comparable.

Adjustment for controls normalizes sample responses to the selected positive (and optional negative) controls, to correct for drift in surface activity during the course of the assay. Cutoff/Ranking is manually set. Alternatively, control-based automatic cutoff allows a cutoff to be set at the selected number of standard deviations in relation to the control.

Evaluation methods to maximize speed

The data evaluation process may be standardized and speed up by applying predefined evaluation methods, shortening time to decision. The software comes with application-specific Evaluation methods that may be applied as is or used as template for optimized user-defined evaluation methods. Relevant evaluation parameters may be saved in Evaluation methods, avoiding repetitive tasks. When opening a run file with the selected Evaluation method, the first results are obtained in minutes.

Fast export of selected or comprehensive data

The flexible result export feature in Biacore Insight Evaluation Software provides the means to export selected or comprehensive data for continued data processing, result reporting, or storage in the company database. Data is exported in a Microsoft® Excel® format. Evaluation items included in the export may easily be selected and detailed settings for each item defined. Sensorgrams or plots may be included in the export as raster images in three different sizes. Images may also be exported as vector graphics. Result tables are exported either using a comprehensive format or using the same format as defined in the evaluation data. Results are therefore always presented in a preferred format to facilitate reporting or adoption into other software.

Data may be easily transferred into Microsoft PowerPoint® format (Fig 4). Simply switch to the Presentation generator to select evaluation items and export them as “native” PowerPoint charts and tables.

Data may be further modified using PowerPoint extensive tool set and layouts. This new way of working with Biacore data sets will make it even easier to share data with your colleagues and peers.

Bring greater flexibility to the software with extensionsBuilding on the framework of the Basic Screening and Characterization package, the Extended Screening and Characterization package may be selected upon startup and unlocked for more advanced features or dedicated application support for screening of LMW fragments to speed up assay setup and streamline evaluation (Fig 5).

With a growing number of extension packages, you will be able to use Biacore Insight Evaluation Software as your research expands and evolves.

Efficiently analyze challenging interactions

• Characterize a wider range of compound-target interactions using additional kinetic fitting models and evaluation settings.

• Evaluate challenging interactions such as LMW fragment binding to transmembrane G protein-coupled receptors (GPCRs) and bivalent protein interactions with relative ease.

• Dedicated software tools for screening of LMW fragments to speed up assay setup and streamline evaluation.

The Extended Screening and Characterization package enables a wider variety of fitting models (Fig 6) using maximum response (Rmax) control and Blank subtraction, and gives access to more customization options (grouping of data channel vs channel, color by curve markers). The extension package also has three kinetic and/or evaluation templates—Clean screen, Binding level screen, and Affinity screen. These evaluation methods guide the user and shorten time to results for the entire SPR workflow in fragment-based drug discovery, FBDD (Fig 7).

Clean screen (Fig 8) enables efficient LMW fragment library cleanup by identifying sticky, residual binding to the sensor chip surface that masks subsequent cycles. Sticky, residual binding is evaluated by assessing the difference in absolute baseline response between adjacent cycles in a run.

Binding level screen (Fig 9) identifies primary hits to the target to find and exclude fragments with atypical binding behavior. Fragment hits at this stage are identified by both binding response and by curve shape, which gives much deeper insight into the binding characteristics of the fragments.

Affinity screen (Fig 10 and 11) verifies target binding and enables an estimate of steady-state affinities of fragments selected in previous screens. Affinities for fragments are often in the millimolar to high micromolar range. Figure 11 shows Affinity screen sensorgram evaluations.

While providing guidance, the templates are fully flexible, may be easily adjusted to fit specific assay needs, and may be saved upon customization to speed up future analyses.

Confident concentration and potency analysis

• Determine active protein concentration with confidence, short time to results with automation and minimum hands-on time.

• Increase assay precision and reproducibility over a wide dynamic range.

• Reliably determine drug product amount and activity using surrogate potency assays, reducing costs and saving time

• Eliminate errors in data transfer with built-in Parallel Line Analysis tool

The Concentration and Potency Extension package enables measurement of the biologically active concentration, not only the total protein amount that would be obtained from an A280 determination. The precision and automation of the system reduces hands-on time and generates highly reproducible data over a wide dynamic range with CV typically below 5%. The software supports both parallel and serial run modes allowing you to select the optimal mode based on your needs.

Calibration curves may be generated using four-parameter or linear-fitting models. The software supports evaluation of the samples using single and average calibration curves. For long runs in serial mode, interpolation of calibration curves to compensate for drifts in the assay may be performed to generate highly reliable data. The possibility to include control samples allows you to ensure a rigorous quality control of your assay.

Drug product amount and activity are reliably determined using surrogate potency assays, reducing costs and saving time. Potency assays can be easily evaluated using the built-in software tools of the Concentration and Potency Extension package. This allow you to determine the potency of your sample relative to a reference sample. This streamlines the analysis workflow and lowers the risk for user mistakes and data manipulation when transferring data between different software. Potency analyses are performed using Parallel Line Analysis (PLA) or half-maximal response (EC50) determinations. The PLA functionality makes the assessment of potency based on a linear fit to the linear part of the response vs logarithmic concentration assuming a common slope (Fig 12). The EC50 determination is based on a four-parameter equation being fitted to the response vs concentration.

Multisystem support and easy data handlingBiacore Insight Evaluation Software analyzes data derived from Biacore 8K+, Biacore 8K, Biacore T200, and Biacore S200 systems for the supported applications (Table 1). Users analyze data from different sources using the same familiar interfaces, features, workflows, and graphical presentation of data. This reduces training needs if multiple and different Biacore instruments are used.

A detailed list of basic features as well as the content of the advanced Extended Screening and Characterization extension are listed in Table 2.

Work in GxP-regulated environments - optional software for GxP compliance

An optional GxP Extension package allows Biacore 8K and Biacore 8K+ to integrate seamlessly into GxP-regulated workflows. The package provides validated software supporting GLP/GCP/GMP and 21 CFR Part 11 compliance, and includes validation support. For full validation support of the system, the package can be supplemented with GE Qualification Service.

Features in the GxP Extension package include:

• Data integrity – access control and enforced version handling

• User authorization levels – administrator, developer and user levels set access rights to software functions

• Procedures for operational control – enables assay run and evaluation settings to be locked together in routine assays

• Audit trail – tracks record modifications and maintains complete version histories for published procedures

e-LicensingBiacore Insight Evaluation Software introduces floating licenses that allow sharing of access rights between users and computers and removes restrictions on the number of simultaneously installed copies of the software. Users are able to electronically secure a machine license for one computer, or a concurrent license for networks. In addition, time-limited licenses are available, allowing you to scale the number of licenses needed for more flexibility.
