1) INTRODUCTION
Lipid peroxidation is a natural process essential for cell growth. However, when the oxidative stress overwhelms the
antioxidative cell defense, the balance is disturbed and enhanced formation of lipid peroxidation products occurs. At
present, lipid peroxidation is considered to be one of the basic mechanisms involved in the initiation and progression ofmany diseases.Various studies have provided evidence that oxidative stress resulting in lipid peroxidation and protein
modification is involved in the pathogenesis of atherosclerosis and coronary heart disease. Lipid peroxidation products are
formed during normal cell metabolism via producing an excess of free radicals that can react with unsaturated fatty acids, in
particularly low-density lipoprotein (LDL), the major carrier of plasma cholesterol. LDL is eliminated by macrophages.
Normally, receptor mediated uptake of LDL is suppressed through down-regulation of LDL receptor expression in response
to increased cholesterol levels. Once LDL is oxidized, it is still internalized by macrophages but through scavenger
receptors whose expression is not controlled by cholesterol loading. The binding of oxidized LDL (oxLDL) is the step by
which cholesterol accumulation in macrophages is induced transforming them into lipidloaded foam cells. This process is
accompanied by extensive cell proliferation and elaboration of extracellular matrix components and contributes to the
genesis and progression of atherosclerosis by promoting endothelial damage and amplifying the inflammatory response
within the vessel wall. Cholesterol-loaded macrophage foam cells are present in the earliest detectable atheroscleroticlesions, the precursor of more complex atherosclerosis that cause stenosis and limited blood flow. These advanced lesions
ultimately represent the sites of thrombosis leading to myocardial infarction.

5) REAGENTS AND SAMPLE PREPARATION
Samples should be stored at -20C, for long term storage store at -70C. Lipemic or haemolysed samples may giveerroneous results. Samples should be mixed well before assaying. We recommend duplicates for all values.
Handling / Reconstitution:
STD (standards) and CTRL (control) are supplied frozen. Immediately put them at or below -20C after receipt.
5 freeze-thaw cylces can be performed without loss of activity. They are stable until expiry date stated on the labelwhen stored at or below -20C
WASHBUF (Wash buffer): Dilute the concentrate 1:20 (e.g. 50 ml WASHBUF + 950 ml distilled water). Crystals in thebuffer concentrate will dissolve at room temperature. Diluted buffer is stable at 2-8C until expiry date stated on label. Use only diluted WASHBUF (Wash buffer) for the assay performance.
The EDTA-plasma and serum samples should be diluted 1+9 (e.g. 50 l + 450 l) with ASYBUF (assay buffer) prior toanalyses. Standards and control are after thawing ready to use andMUST NOT be diluted.

7) ASSAY PROTOCOL
All reagents and samples must be at room temperature (18-26C) before use in the assay.
Mark position for BLANK/STD (Standards)/SAMPLE/CTRL (Control) on the supplied protocol sheet.
Take microtiter strips out of the alubag, take a minimum of one well as Blank. Store unused strips with desiccant at
2-8C in the alubag. Strips are stable until expiry date stated on the label.
Add 100 l STD/CTRL/diluted SAMPLE (Standard,/Control, /diluted Sample) in duplicate into respective wells.
Cover the strips tightly and incubate at 37C for 2 hours.
Aspirate and wash wells 5x with 300 l diluted WASHBUF (Wash buffer). Remove remaining WASHBUF by hitting
plate against paper towel after the latest wash.
Add 100 l CONJ (Conjugate, anti oxLDL antibody - HRPO) into each well, except blank.
Cover the strips tightly and incubate at 37C for 1 hour.
Aspirate and wash wells 5x with 300 l diluted WASHBUF (Wash buffer). Remove remaining WASHBUF by hitting
plate against paper towel after the last wash.
Add 100 l SUB (Substrate) into each well. Incubate at room temperature (18-26C) for 30 minutes in the dark.
Add 50 l STOP (Stop solution) into each well, shake well.
Measure absorbance immediately at 450 nm with reference 620 nm, if available.
8) CALCULATION OF RESULTSSubtract the blank extinction from all other values. Construct standard curve from standard values. Use commercially
available software or graph paper. Obtain sample concentration from this calibration curve.
The results of the diluted samples (ng/ml) must be multiplied by 10.
The assay has been evaluated using a 4PL algorithm. Different curve fitting methods need to be evaluated by the user.Respective dilution factors have to be considered.
The quality control protocol supplied with the kit shows the results of the final release QC for each kit. Data for optical
density obtained by customers may differ due to various influences and/or due to the normal decrease of signal intensity
during shelf life. However, this does not affect validity of results as long as an optical density of 1.00 is obtained for the
standard with the highest concentration.

11) TECHNICAL HINTS
Do not mix or substitute reagents with those from other lots or sources.
Do not mix stoppers and caps from different reagents or use reagents between lots.
Do not use reagents beyond expiration date. Protect reagents from direct sunlight.
Substrate solution should remain colourless until added to the plate. To ensure accurate results, proper adhesion of plate sealers during incubation steps is necessary.
12) PRECAUTIONS All test components of human source were tested with 3rd generation tests against HIV-Ab and HBsAg and were found
negative. Nevertheless, they should be handled and disposed as if they were infectious.
All liquid reagents contain 0.01%Proclin 300 as preservative. Proclin 300 is not toxic in concentrations used in this kit. It
may cause allergic skin reactions avoid contact with skin or eyes.
Do not pipette by mouth.
Do not eat, drink, smoke or apply cosmetics where reagents are used.
Avoid all contact with reagents by using gloves. Sulfuric acid is irritating to eyes and skin. Flush with water if contact occurs. Avoid contact with skin and mucous.
Irritations are possible. Flush with water after contact!

1) EINLEITUNG
Oxidation von Lipoproteinen ist ein fr das Zellwachstum natrlicher und notwendiger Prozess. Wenn der oxidative Stress
jedoch die anti-oxidativen Abwehrmechanismen der Zelle berwindet, wird die Balance gestrt und eine erhhte
Peroxidation von Lipoproteinen findet statt. Die Peroxidation von Lipoproteinen wird als grundlegender Mechanismus beider Entstehung und Entwicklung von vielen Krankheiten diskutiert.
Verschiedene Studien haben gezeigt, dass oxidativer Stress in der Peroxidation von Lipoproteinen und der Modifikation von
Protein resultiert und damit in die Pathogenese von Artherosklerose und koronaren Herzerkrankungen involviert ist.
Produkte der Lipid-Peroxidation entstehen im normalen Zellmetabolismus durch Bildung eines berschusses an freien
Radikalen. Diese reagieren mit ungesttigten Fettsuren, insbesondere mit solchen am low density lipoprotein (LDL),
welches das Haupttransportmolekl fr Cholesterin darstellt. LDL wird von Makrophagen aus der Zirkulation eliminiert. Die
rezeptorvermittelte Elimination von LDL wird durch die Down-Regulierung des LDL Rezeptors auf den Makrophagen in
Antwort auf erhhte Cholesterinspiegel unterdrckt.
Wenn LDL oxidiert worden ist, kann es noch immer von Makrophagen aufgenommen werden, allerdings nur durch einen
Scavenger Rezeptor, welcher nicht ber den Cholesterinspiegel kontrolliert wird. Diese Aufnahme von oxidiertem LDL
(oxLDL) ist jener Schritt, der zur Anreicherung von Cholesterin in den Makrophagen fhrt und sie in lipidgeladeneSchaumzellen (foam cells) transformiert. Dieser Prozess wird von extensiver Zellproliferation und Bildung extrazellulrer
Matrixkomponenten begleitet und trgt somit zur Entstehung und zum Fortschreiten der Atherosklerose bei, indem das
Endothel beschdigt und entzndliche Prozesse innerhalb der Gefwnde verstrkt werden. Cholesterinbeladene
Makrophagen finden sich schon in den frhesten, gerade einmal nachweisbaren atherosklerotische Lsionen, welche die
Vorlufer komplexerer Atherosklerosen darstellen und dann Stenosen und verringerte Durchblutung verursachen. Diese
fortgeschrittenen Lsionen reprsentieren die Stellen, an denen Thrombosen entstehen, die letztlich zum Herzinfarktfhren.

5) REAGENZIEN UND PROBENVORBEREITUNG
Proben sind bei -20C zu lagern, Langzeitlagerung bei -70C.
Lipmische oder hmolysierte Proben knnen zu fehlerhaften Ergebnissen fhren. Vor dem Einsatz im Test Proben gut
mischen. Wir empfehlen, alle Werte in Doppelbestimmungen durchzufhren.
Handhabung / Rekonstitution:
STD (Standards) und CTRL (Kontrolle) werden in gefrorenem Zustand geliefert.
Unmittelbar nach Erhalt bei -20C oder darunter lagern. 5 Frier/Tau-Zyklen knnen ohne Aktivittsverlsut durchgefht
werden. Standards und Kontrollen sind bei -20C oder darunter bis zum Ablaufdatum haltbar.
WASHBUF (Wasch Puffer):
Das mitgelieferte Konzentrat wird 1:20 verdnnt (zB. 50 ml WASHBUF + 950 ml destilliertes Wasser). Kristalle im
Pufferkonzentrat lsen sich bei Raumtemperatur auf. Der verdnnte Puffer ist bei 2-8C bis zum Ablaufdatum haltbar.
Im Testsystem darf nur verdnnter WASHBUF (Waschpuffer) verwendet werden.
Sowohl EDTA Plasma als auch Serum-Proben mssen vor dem Einsatz im Test 1:10 (1+9, zB. 50 l + 450 l) mit ASYBUF
(Assay buffer) verdnnt werden. STD und CTRL sind nach dem Auftauen gebrauchsfertig und drfen nicht verdnnt werden.

7) TESTPROTOKOLL
Im Test drfen nur Reagenzien und Proben verwendet werden, welche Raumtemperatur (18-26C) aufweisen.
Markieren Sie die Positionen fr BLANK/STD/PROBE/CTRL (Leerwert/Standard/Probe/Kontrolle) am Protokollblatt.
Nehmen Sie die bentigten Mikrotiterstreifen aus dem Alu Sckchen. Mindestens 1 Well fr den Leerwert reservieren.
Nicht verwendete Mikrotiterstreifen knnen mit Trockenmittel im Alu Sckchen bis zum angegebenen Ablaufdatumgelagert werden.
Pipettieren Sie 100 l STD /CTRL(Standard /Kontrolle ) oder SAMPLE (verdnnte Probe) in Doppelbestimmung in die
Mikrotiterstreifen.
Streifen abdecken und 2 Stunden bei 37C inkubieren.
Inhalt der Wells verwerfen und 5x mit 300 l verdnnten WASHBUF (Waschpuffer) waschen. Nach dem letzten
Waschschritt Reste von Waschpuffer durch Ausklopfen auf saugfhigem Papier entfernen.
Pipettieren Sie 100 l CONJ (Konjugat) in alle Wells, mit Ausnahme des Leerwerts.
Streifen abdecken und 1 Stunde bei 37C inkubieren.
Inhalt der Wells verwerfen und 5x mit 300 l verdnnten WASHBUF (Waschpuffer) waschen. Nach dem letzten
Waschschritt Reste von Waschpuffer durch Ausklopfen auf saugfhigem Papier entfernen.
Pipettieren Sie 100 l SUB (Substrat) in alle Wells.
30 Minuten bei Raumtemperatur (18-26C) im Dunkeln inkubieren.
Pipettieren Sie 50 l STOP (Stopplsung) in alle Wells, mischen.
Extinktion unmittelbar bei 450 nm messen, mit 620 nm als Referenz, wenn mglich.
8) BERECHNUNG DER ERGEBNISSE
Die OD des Leerwertes ist von allen Wells abzuziehen. Konstruieren Sie eine Standardkurve aus den Werten der
Standards unter Verwendung von kommerziell erhltlichem Millimeterpapier oder einer geeigneten Software. DieKonzentration der Proben wird aus der Standardkurve abgelesen.
Die Ergebnisse der verdnnten Proben (ng/ml) mssen mit dem Verdnnungsfaktor 10 multipliziert werden.
Der Test wurde mit einem 4PL Algorithmus Auswerteprogramm evaluiert. Andere Auswerteprogramme mssen vom
Verwender evaluiert werden.
Eventuelle weitere Verdnnungen mssen bercksichtigt werden.
Auf dem beigepackten QC Protokoll sind die Resultate bei der QC Freigabe des Kits vermerkt. Vom Verwender erhaltene
Daten der optischen Dichte (OD) knnen abweichend sein, bedingt durch verschiedene Einflsse und/oder dem normalen
Signalverlust des Kits whrend der Laufzeit. Dieser mgliche Signalverlust hat keinen Einfluss auf die Gltigkeit derResultate, so lange die OD des hchsten Standards den Wert 1,00 erreicht.

11) TECHNISCHE MERKMALE
Reagenzien verschiedener Lots oder aus anderen Quellen drfen nicht mit Kitreagenzien gemischt werden.
Stpsel oder Verschlsse von verschiedenen Reagenzien drfen nicht vertauscht werden.
Abgelaufene Reagenzien drfen nicht verwendet werden. Reagenzien sind vor direktem Sonnenlicht zu schtzen.
Substratlsung muss vor Verwendung farblos sein.
Mikrotiterstreifen mssen bei den Inkubationen mit Abdeckfolie abgedeckt sein.
12) VORSICHTSMASSNAHMEN
Alle Bestandteile humanen Ursprunges wurden mit Testen der 3.Generation auf HIV Ak und HBsAg getestet und negativ
gefunden. Trotzdem sollten die Reagenzien als potentiell infektis behandelt werden.
Alle flssigen Reagenzien enthalten 0,01% Proclin 300 als Konservierungsmittel.
Proclin 300 ist in der verwendeten Konzentration nicht toxisch. Allergische Reaktionen sind mglich.
Nicht mit dem Mund pipettieren.
Nicht rauchen, essen, trinken oder Kosmetika benutzen whrend der Verwendung der Testreagenzien.
Verwenden Sie Handschuhe zur Vermeidung jedes Kontaktes mit Reagenzien.
Schwefelsure reizt die Augen und die Haut. Bei Berhrung grndlich mit Wasser splen.

ASSAY PROTOCOL AND CHECKLIST
Bring all reagents to room temperature (18-26C).
Prepare reagents and samples as instructed.
Take microtiter strips out of the alubag and mark positions on the protocol sheet.
Add 100 l STD/CTRL/diluted SAMPLE/ (standard/control/diluted sample) into respective
wells.
Cover tightly and incubate at 37C for 2 hours.
Aspirate and wash wells with 300 l WASHBUF (Wash buffer) five times. Remove
remaining buffer by hitting plate against paper towel.
Add 100 l CONJ (Conjugate) into each well, except blank.
Cover tightly and incubate at 37C for 1 hour.
Aspirate and wash wells with 300 l WASHBUF (Wash buffer) five t imes. Remove
remaining buffer by hitting plate against paper towel.
Add 100 l SUB (Substrate) into each well.
Incubate at room temperature (18-26C) for 30 minutes, in the dark.
Add 50 l STOP (Stop solution) into each well.
Read Optical Density at 450 nm with reference 620 nm, if available.
