Water, the life origin
The life has first grown in the ocean. Then,the watery species left their medium little bylittle to conquer the terrestrial part of ourEarth. By doing this, they set up completelyinteresting adaptive strategies in order to pre-serve their hydrous capital and to survive intheir new environment.
In the animal world, the periophtalm of themangroves is an illustration of this passage ofthe watery life to the terrestrial life: this smalleasily recognizable jumping fish with largeprotruding eyes is perfectly adapted to these2 environments. It eats insects like the mos-quitoes essentially and thus passes the majo-rity of its existence out of water.
In the vegetable world, one finds particularproperties related to the adaptation of thespecies. One thus discovers aerial roots in theficus, and of the fruit trees, like pomegranatetree, amazing properties to bloom in comple-tely unexpected wet places!
Each reign, each specie, thus followed aprogressive evolution enabling to pre-serve the source of its existence, thewater.

Our skin needs a sufficient water uptake, firstto reconstitute the water pool in the dermis(to avoid dehydration and following decreasingskin elasticity and wrinkles appearance), andalso to renew the hydrolipidic film that isessential for the epidermis barrier function.
The barrier function is vital since it allows theorganism protection against potential dange-rous external agents (pollution, temperaturevariation, UV, etc.). The epidermis also takespart in general homeostasis regulation by limi-ting water loss across the skin.
Nevertheless water applied on the skin mois-turises or re-moisturises under two mainconditions:
• the water has to be fixed by lipids that preventevaporation;
• the water has to circulate in the whole skintissue
In a normal and young skin, the water contentof the epidermis is about 10 to 15% in thestratum corneum and about 60 to 70% in thedeeper levels of the skin. There is a water gra-dient maintained by the water flow from themore hydrated cells to the less ones.
When the water content of the stratum cor-neum decreases under the critical thresholdof 10%, the epidermis surface becomes dry,tends to crack and lose the elasticity that isessential for a normal skin softness and velvety.Indeed, the more dry is the keratine structure,the less supple it is.
As a matter of fact, a drying up of the skin isnoticed when it is getting old: the skin loses itssooth aspect, it is wizened and furrowed withwrinkles.
To bring an answer to this phenomenon,Alban Muller International (A.M.I.) hascreated Amiporine®, the first restructu-ring active that restores water circula-tion in the skin, and thus preventsageing.

The fundamental role played by cell membra-nes in the plants water capital preservationwas highlighted when aquaporins have beendiscovered. These proteins are water chan-nels vital for some plants functioning.
As a response to environmental changes,aquaporins expression, reveals an extremelyperforming adaptive strategy developed byspecies.
Pomegranate tree is bearing pomegranate andperfectly illustrates this adaptive phenomenon.
This tree preferentially grows in southerncountries and on the opposite of most of fruit

Aquaporins were discovered for the first time byelectronic microcopy in 1974. They were reallycharacterised in 1991 by Peter AGRE, who recei-ved the Nobel Prize in Chemistry in 2003 for thiswork.
Up to date, 200 aquaporins were listed in the ani-mal kingdom of which 10 in the human organism.These molecules are generally named AQP follo-wed by a number corresponding to their disco-very time. The aquaporin 3 (AQP3) is the only oneknown to be expressed in the epidermis.
Aquaporins are inter-membrane molecules for-ming, specialised tunnels for water flow in the cellwall.
In the human body, water flow in the skin is ensu-red by a specific aquaporin known as aquaporin 3(AQP3). This protein is expressed by epidermiscells and is essential in the general skin metabo-lism. It is actually a glycero-aquaporin, that is to saythis protein is permeable both to water and glyce-rol (or glycerine) which play an important role inthe hydro-lipid barrier formation.
Aquaporins allow to preserve a water balance inthe skin even in unfavourable conditions (oppositeosmotic pressure): they facilitate water and smallsolutions (glycerol for example) diffusion acrossthe membranes.
When aquaporins metabolism is altered, there is awater gradient modification leading to a bad watercirculation. The skin is then deprived of its watercapital and loses its suppleness and young aspect.

The A.M.I. research team has performed astudy on aquaporins 3 expression dependingon the age. The results demonstrate the dra-matic decreasing of their presence in the skinwhen subjects are 40 to 45 years old.
The aquaporins expression decreaseswith age. When these "water channels"do not work anymore, the whole skinmetabolism is disturbed.

Analytical method: indirect immunofluorescence
Skin explants were obtained by surgery on 3and 53 years old subjets. They were sectionedusing a cryostat and fixed on glass slide.
The aquaporins expression is highlighted usinga specific antibody for human aquaporin 3.This antibody is revealed using a second onecoupled with fluoresceine.
The experiment reveals the difference ofaquaporins expression between a young (1)and an old (2) skin.
Obviously, by maintaining its water capi-tal aquaporins play an essential role topreserve the young and tonic aspect ofthe skin.

(1) Young skin organisationThe stratum corneum is steady and thick, keratinocytesof the epidermis widely show aquaporin 3. This pre-sence is gradual and more intense in the deep levelsthan in the external level of the epidermis.
(2) Old skin organisationThe skin organisation appears widely modified with age:the stratum corneum is thin and the aquaporin 3 isweakly expressed by keratinocytes. The staining noticedin the dermis matrix is a non-specific staining.

Analytical method
Keratinocytes used for these experiments arefrom HaCat cell line. One night after beingcultured, the media is exchanged with a newone containing Amiporine®.
After a 72-hour incubation, cultures are rinsedand fixed for immunostaining.
Wiser for these results on aquaporins, A.M.I.has developed a purified pomegranate extractable to rehabilitate the natural water flow bystimulating the skin specific aquaporins(AQP3).
By restoring water balance, Amiporine®
brings a renewed youth to skin: the epidermisis perfectly moisturised and the stratum cor-neum regains its suppleness and brightness.

3 - Consequences of Amiporine® activity on skin metabolism:communication between epidermis cells is optimal
Re-establishing water flow in the epidermis allows the skin to restore its supplenessand beauty. This unequalled effect of Amiporine® is the result of a general improve-ment in cells metabolism. As an example, the communication between keratinocy-tes was studied.
Analytical method
The following test was performed using a spe-cific fluorescent tracer: Lucifer Yellow, thatpenetrates in cells if they have first been arti-ficially cut.
When entered in a cell, Lucifer Yellow can dif-fuse in neighbouring cells using gap junctions.These are involved in the skin tissue organisa-tion and coherence since they favour cellscommunication.
The following pictures were taken using afluorescent microscope. In this manner, theeffect of Amiporine® on cell communicationvia gap junction was visualised.

4 - Amiporine® effect on collagen I and III and matrix collgenase synthesis by human dermal fibroblasts
Analytical method
In order to demonstrate the Amiporine® acti-vity on human dermal fibroblasts via the epi-dermis, keratinocytes were previously incuba-ted with different doses of the active. Themedium obtained was then used for fibro-blasts culture.
This experiment demonstrates that Amiporine®
also acts on fibroblasts via the epidermis cells.The active stimulates collagen I and III synthe-sis and decreases collagenase synthesis. Onthe opposite of collagen that build the dermismatrix, collagenase damages it.
• Keratinocytes treatment
Human keratinocytes were cultured andincubated overnight with a standard culturemedium. Supernatants were then eliminatedand replaced by a new medium (control) ormedium containing Amiporine®.
After a 6-hour incubation, supernatantswere eliminated, cells were rinced andagain incubated with a new medium during24 hours. After this period, supernatantswere centrifuged and filtered for fibroblastsculture.
• Fibroblasts treatment
Fibroblasts were cultured with keratinocy-tes supernatants.
After a 48 hours incubation, supernatantswere filtered and freeze at -20°C. The mea-surement of collagenase using ELISA dosagewas then performed.
The measurement of collagen I and III wasperformed by using a semi-quantitativemethod of fluorescent immuno-labelling :after the labelling of collagens I and III, cellswere lysed and fluorescent supernatantswere then quantified with a spectrofluori-meter (Fluoroscan).

5 - Hormonal variation and aquaporins
All their life long, women are subjected toseveral hormonal variations, particularlyduring menstrual cycle and on pregnancy.
These changes widely alter cells behaviourand can lead to a dysfunction affecting thegeneral aspect of the skin.
Our research team performed a test whichdemonstrate that an increase in a sporadichormonal concentration such as the estradiol,can alter aquaporins expression. This experimentwas realised thanks to a flow spectrofluorimeter:this high-performance device allows to labelthe cells with a specific fluorescent antibodyand to analyse cells precisely, one by one.
The results obtained prove thatAmiporine® is able to reverse the estra-diol effects on aquaporins expression andeven to stimulate it despite the hormonepresence.

The Amiporine® solvent is glycerine which is apolysaccharide perfectly tolerated by the skinand which contributes to its preservation andfirmness.
Indeed, disposable scientific bibliographydemonstrates that glycerine favours the epi-dermis barrier healing and increases the stra-tum corneum hydration.
These effects are partly caused by the aqua-porin 3, one of the few proteins permeableboth to water and other solutions such as gly-cerine or urea.

Formulation Amiporine® can readily be incorporated in an oil-in-water emulsion or pre-diluted in water before being added in a cream or a gel.
Amiporine® should be cold-incorporated at the end of the formulation process (temperature between 30 and 40°C).
