Bacterial Transformation with Plasmid DNA

Uptake of new genes by bacterial cells resulting in an altered genotype and phenotype (the phenotype has been “transformed”)

How can we get our plasmid DNA into the bacteria?

Treat cells so they are “competent” to take up DNA from their surroundings

1. Treat cells with CaCl2

2. Add plasmid DNA

3. Heat Shock the cells
