lysosome exocytosis as well as mature IL-1� release in theP2X7R-null BMDM.

P2X7R-induced exocytosis of secretory lysosomes is LPSdependent and requires protein synthesis, proteasome activity,and NF-�B signaling

Given our previous finding (30) that LPS priming of BMDM wasrequired for efficient coupling of P2X7R to inflammasome activa-tion and IL-1� export, we tested whether a similar requirement forLPS priming characterized the activation of secretory lysosomeexocytosis by ATP. Notably, non-LPS-primed BMDM secretedonly a minimal amount of cathepsin B and no active caspase-1 inresponse to ATP stimulation (Fig. 3A). The coupling of P2X7R toinflammasome activation could be observed within 30 min of LPSpriming, as indicated by the ATP-stimulated accumulation of in-tracellular caspase-1 p10. Similarly, an increase in ATP-elicitedcathepsin B release was apparent within 30 min after initiationof LPS priming. In cells primed with LPS for longer durations(1 or 4 h), the ATP-stimulated release of both active caspase-1and cathepsin B was further increased in a coordinated manner.In the absence of ATP stimulation, even 4 h of LPS primingalone did not result in inflammasome activation, IL-1� secre-tion, or cathepsin B release. Furthermore, the ability of LPS topotentiate ATP-dependent caspase-1 activation and secretorylysosome exocytosis occurred before significant expression ofpro-IL-1� protein. These observations indicate that LPS prim-ing provides a necessary and common signal(s) for efficientcoupling of P2X7R to exocytosis of secretory lysosomes as wellas inflammasome activation.

Other experiments tested whether de novo protein synthesis ormaintained synthesis of rapid turnover proteins was necessary forP2X7R-dependent inflammasome activation and P2X7R-activatedlysosome exocytosis. Pretreatment of cells with the protein syn-thesis inhibitor cycloheximide, the proteasome inhibitor MG132,or the I�-B kinase inhibitor Bay 11-7085 before LPS priming com-pletely repressed not only intracellular pro-IL-1� accumulation,but the P2X7R-induced activation of the caspase-1 inflammasome.As indicated in Fig. 3, B and C, these three inhibitors similarlyrepressed the correlated lysosomal protein secretion, but did notchange the expression levels of procaspase-1 or cathepsin B. Theexpression of P2X7R and LAMP-1 protein, as well as P2X7Rchannel function, was not altered in cells treated with these phar-macological reagents (data not shown).

ASC, active caspase-1, and pannexin-1 (Px1) are required forIL-1� secretion, but not secretory lysosome exocytosisstimulated by P2X7R

The adaptor protein ASC, which is essential for assembly ofcaspase-1 inflammasome complexes, is also coreleased withcaspase-1 and IL-1� (10) (Fig. 8B). This suggests that the entireinflammasome complex is copackaged and released as a unit withits IL-1� product from ATP-stimulated cells. We used BMDMfrom ASC-null mice to test whether ASC was also required for theP2X7R-regulated exocytosis of secretory lysosomes, which coin-cided with IL-1� secretion. Fig. 4A shows that ASC�/� BMDMwere completely deficient in ATP-induced activation of inflam-masome complexes as well as the coupled release of matureIL-1� and active caspase-1. However, release of the soluble
