ESTIMATION OF TOTAL PHENOLIC CONTENT
(Folin-Ciocalteau Method)
[Swain, T. and Hillis, W.E. (1959) as modified by Thaipong et al.,(2006) ]
PRINCIPLE
The Folin-Ciocalteau assay relies on the transfer of electrons in alkaline medium from
phenolic compounds to phosphomolybdic/phosphotungstic acid complexes to form blue
complexes (possibly (PMoW11O40)4−) that are determined spectroscopically at approximately 725
nm. In this assay phenols present in vegetables and fruits extracts react with phosphomolybdic
acid in Folin-ciocalteau reagent and sodium carbonate, produces blue coloured complex
(molybdenum blue).
REAGENTS AND CHEMICALS REQUIRED
1. Folin – Ciocalteau reagent for phenols
2. 1 N Sodium carbonate
3. Standard – Gallic acid- 5mg in 100mL De-mineralised water (50µg/mL)
PROCEDURE
Preparation of Extract is given in Methodology (Section 3.8). Standardisation of assay
protocol is given in Methodology (Section -3.4.2) and result of standard plot is given in Section
4.3. The aliquots of extracts (200µL) were made up to 2450µL using de-mineralised water. Then
added 150 µL of Folin-ciocalteau reagent mixed well and allowed to react for 3 minutes at room
temperature. Then added 300 µL of 1 N sodium carbonate and mixed well. The solution was
incubated at room temperature in the dark for 30 minutes. Simultaneously a blank was prepared
devoid of sample extract. The absorbance was measured at 725 nm using UV-Vis double beam
spectrophotometer (Systronics- Model-2201). Standard graph was plot using different
concentrations of Gallic acid. The results were expressed in GAE equivalence for gram fresh
weight of the sample.

FRAP (Ferric Reducing Antioxidant Power) ASSAY
[Benzie and Strain, (1996) with some modifications as per Thaipong et al. (2006)]
PRINCIPLE
In the presence of antioxidants, ferric tripyridyltriazine (Fe 3+) complex is reduced to
ferrous ion at low pH (3.6) which has an intense blue colour which can be monitored by
measuring the change in absorption at 593nm in spectrophotometer. The change in absorbance is
directly related to the combined or total reducing power of the electron-donating antioxidants
present in the reaction mixture.
REAGENTS AND CHEMICALS
1. Reagent A -Acetate buffer (pH-3.6)- 300 mM/L
Sodium acetate (3.1g) was mixed with 16mL glacial acetic acid and made up to 100mL
with distilled water. This solution was prepared freshly and warmed at 37°C
2. Reagent B -TPTZ (2, 4, 6- Tripyridyl-S-triazine)
TPTZ 10 mM was dissolved in warm 40mM hydrochloric acid (40°C)-Prepared
freshly
3. Reagent C -Ferric chloride hexahydrate -20mM/L-
Ferric chloride reagent was prepared by dissolving 3.24 mg ferric chloride in 100mL
of water- Prepared freshly
4. Working Standard FRAP reagent
Mix Reagent A, B and C in the ratio of 10:1:1 – Prepared freshly
5. Standard Trolox was prepared in 50% methanol (20µM/mL)
PROCEDURE
Preparation of Extract is given in Methodology (Section 3.8). Standardisation of assay
protocol is given in Methodology (Section -3.4.2) and results of standard plot was given in
Section 4.3. Pipetted 200 µL of sample extract in a clean dry test tube added 2900 µL of working
standard FRAP reagent and vortexed. Then the tube was kept in the dark at room temperature
(37°C) for 30 min. The intensity of colour developed was read at 593 nm in spectrophotometer
(Systronics-2201) against reagent blank. The standard curve was plot between 0.2 to 4.0 µM and
the results were expressed in µM of Trolox per gram Fresh Weight.

DPPH (2,2-diphenyl 1-picryl hydrazyl)Assay
[Brand-Williams et al. (1995) with some modification as per Thaipong et al. (2006)]
PRINCIPLE
The spectrometric method for assessing the total antioxidant activity is based on the
absorbance decrease monitoring of the DPPH radical (2, 2- diphenyl-1-picrylhydrazyl assay) in
the presence of antioxidant (Refer Methodology- Section 3.4.5). DPPH is characterized as a
stable free radical, the delocalization of the spare electron over the molecule rise to a deep violet
colour characterized by an absorption band at about 517nm. When a DPPH solution is mixed with
a substance which can donate hydrogen atom, the reduced form is generated, accompanied by the
loss of the violet colour.
REAGENTS
1. Methanolic solution of DPPH- Weighed accurately 24 mg of DPPH and dissolved in
100 mL methanol and stored at -20° C. The working solution was obtained by mixing
10mL stock solution with 45mL methanol to obtain an absorbance of 1.1 to 1.2 units
at 517nm using spectrophotometer.

PROCEDURE

protocol is given in Methodology (Section -3.4.2) and result of standard plot is given in Section

DPPH. Mixed well and allowed to stand for 30 minutes at 37 ° C in dark. The discolouration of
DPPH solution in the presence of sample occurs and it was read at 517nm against methanol as
blank. Prior to sample treatment, the DPPH solution devoid of sample was read at 517nm. This
serves as control. The difference in sample and control absorbance was taken to calculate Total
antioxidant activity against DPPH radical in the food sample. The standard plot was prepared
between the concentrations of 0.2 to 4.0 µM by measuring the radical scavenging activity of
Trolox (Standard).

ABTS (2, 2 - Azinobis - 3 - ethyl - benzo thiozoline - 6 - Sulphonate) Assay
[Arnao et al. (2001) with some modification as per Thaipong et al. (2006)]
PRINCIPLE
In this assay the ABTS radical is produced continuously by reaction between the
ferrylmyoglobin radical generated H2O2 and peroxidase and the activity of antioxidants to
scavenge the ABTS.+
radical cation is measured by the decrease in its absorbance at 734nm.
Since ABTS.+
is a relatively long lived radical, the TEAC assay was modified by generating the
ABTS.+
radical , the TEAC assay was modified by generating the ABTS.+
radical by direct
oxidation of ABTS with potassium persulphate prior to reaction with test antioxidants.
REAGENTS
1. Solution A -ABTS solution (7.4mM) (2,2- Azino-bis (3-ethylbenzothiazoline-6-Sulfonic
acid) diammonium salt
2. Solution B- Potassium persulphate – 2.6 mM
3. Working solution
The equal quantities of solution A and B were mixed and allowed them to react at 37°C
for 12-16 hour at room temperature in the dark. The solution was then diluted by mixing
with methanol to obtain an absorbance of 1.1 to 1.2 units at 734nm using
spectrophotometer. Fresh ABTS assay was prepared for each assay.
4. Standard Trolox was prepared in 50% methanol (20µM/mL)
