    GENERAL

    LH (Luteinizing Hormone) is one kind ofglycoprotein hormone. Just like FSH (Follicle-Stimulating Hormone), its one of the main factorsmotivating the ovulation. In female mammals, LH iscooperating with FSH to aid the follicle growingmaturely and motivate the ovulation. In addition, itenhances the blood flow. Ovulation is the process inwhich the ovary releasing the ovum and its the keyevent in ovary period. The ovulation of mammals isa complex process involved with many genes and

    steps. A lot of research indicates that ovulation isregulated by hormone and partial active substanceof follicle. Several or more hours before theovulation of mammals, secretion of LH would rise inresponse to female hormone positive regulation andLH surge appears. After the LH surge, the wall ofthe enlarged follicle ruptures at ovulation and themature ovum is extruded. After ovulation, LHreturns to its base line level with the concomitantincrease of progesterone level to initiate lutealphase. Before ovulation, the LH in urine, bloodserum and milk rises gradually and the ovulationoccurs approximately 12-14 hours after the LHsurge.

    In view of the characteristic variation of LH, rapidand sensitive measurement of LH is an importanttool in the diagnosis and management of infertilityin female mammals.

    HOW TO USE

    As the cow come into heat, perform this test afterten hours for normal cows which shows heat inproper cycle. If found positive, breed that cow withinterval of 12 hours. Incase of negative result,perform this test again with the interval of 5 to 6

    hours and breed at positive result.For disturbed heat cycle cows, perform this testafter 12 to 14 hours later to start of heat. If foundpositive, breed that cow with interval of 12 hours.Incase of negative result, perform this test againwith the interval of 5 to 6 hours depending upon

    the heat cycle history of cow. It is important thcow should be breed twelve hours later to positresult.

    SPECI MEN COLLECTI ON AN D PREPARATI ONTake the urine sample as it is as or take bloocentrifuge it and separate blood serum as sampfor this test.

    ASSAY PROCEDURE

    1. Remove the test device and dropper from its fwrapper by tearing along the slice.

    2. Using the dropper, withdraw the urine or serusample from the specimen cup and slowdispense 3-4 drops into the circular sample weWhen the sample liquid moves slowly to tother end due to the capillarity, it will dissolthe solid gold labeled antibody and broughtahead in the mean time.

    3. Read results within 10 minutes.I NTERPRETATI ON OF RESULTSPositive: If the test band (T) is more intensthan or equivalent to that of control band (C), tspecimen is likely at surge level.

    Negative: If only the control band (C) appears, the color intensity of the Test line is less than thof Control line, this means the specimen is belowthe surge level.I nvalid: When neither test nor control baappears on the membrane, the test should voided since improper test procedure deterioration of reagents probably occurred. It recommended that the test be repeated.

    2. Do not uses test kit beyond expiration date.3. Do not reuse the test devices and discard it in

    the dustbin after single use.4. Once opened, the test device should be us

    immediately. Prolonged exposure to atmosphewill lead to deterioration.
